1.Effects of macrophage migration inhibitory factor on glucocorticoid release and glucocorticoid receptor in rats

Objective To investigate the effects of macmphage migration inhibitory factor (MIF) on glucocorticoid (GC) re]ease and glucocorticoid recer (GR) in mts.Methods Test Ⅰ Thirty-two male SD rats weighing 250-300 g were randomly divided into 4 groups(n=8 each):control group(C),low dose recombinant MIF (rMIF) group (rMIF-L),middle dose rMIF group (rMIF-M) and high dose rMIF group (rMIF-H).The animals received l ml normal saline via the right femoral vein in group C.The animals received rMIF50.100 and 200 ng in l ml normal saline though right femoral vein in group rMIF-L,rMIF-M or rMIF-H respectively.Blood samples were taken from left femoral artery immediately before inection(T0,baseline),and at 5 min,3 h,6 h.12 h and 24 h after injection of rMIF(T1-5) for determination of serum concentration of corticosterone.Test Ⅱ Primary cultured neonate rat(2-3 days)myocardial ceils were randomly divided into 3 groups(n=24 each):group C,group rMIF-L and group rMIF-M.The ceUs in group C,rMIF-L and rMIF-M wefe incubated with DMEM.rMIF 50 ng+DMEM and rMIF 100 ng+DMEM for 3 h respectively.The expression of GR and HsPg0 wag determined by Western blot.ResuBs Test Ⅰ The serum concentration of corticosterone was signifieemily higher in the other 3 groups than in group C at T1-5(P<0.05).The sertlm concentration of corticostemne was significantly increased at T1-5 in group rMIF-L,rMIF-M and rMIF-H compared with the baseline values(P<0.05).Test Ⅱ HSP90 expresion was significantly lower in the other two groups than in group C(P<0.05).Them was rio signifieanf difference in HSP90 expression between group rMIF-L and group rMIF-M(P>0.05).There was no significant difference in GR expression among the 3 groups ( P > 0.05). Conclusion MIF druing sepsis can weaken GR function through down-regulating HSP9O expression, resulting in CC resistance.
2.Glibenclamide treatment for gestational diabetes mellitus: A systematic review

Objective To evaluate the effect and safety of glibenclamide for gestational diabetes mellitus (GDM).Methods Cochrane Central Register of Controlled Trials (CENTRAL),Medline,EMBASE,Science Citation Index,CNKI,VIP,Wanfang Data,and CBM were searched for the randomized controlled trials (RCTs) of glibenclamide for gestational diabetes from the date of establishment of the databases to March 2013.The bibliographies of the included studies were searched,too.The included studies were evaluated by GRADE.The extracted data were analyzed by Rev-Man 5.1 and GRADEprofiler 3.2.2.Results Five RCTs were included.Effectiveness (such as maternal postprandial glucose,HbA1C) and adverse perinatal outcomes (such as the change of the pregnant women body weight,cesarean section rate,maternal hypoglycemia incidence,birth body length,incidence of hypoglycemia,jaundice,newborn being in the care unit,congenital malformations,stillbirth,and mortality neonatal) showed no differences between glibenclamide (alone or complemented with insulin) group and insulin group (P>0.05).However,compared with insulin group,higher maternal fasting blood glucose,higher birth weight of newborn infants,and incidence of macrosomia were shown in glibenclamide group.Conclusion Glibenclamide can effectively control maternal blood sugar,and would be a promising alternative therapy for GDM,without adverse effect on fetal growth and development,but with the higher incidence of neonatal macrosomia.Due to the limitations of the included studies,more large-sample,high-quality RCTs are required.
3.Analysis of the Development Strategies for Hospital Preparations in Our Hospital by SWOT Method

OBJECTIVE:To provide reference for the development of hospital preparation in our hospital. METHODS:The method of SWOT analysis was applied to analyze competitive strengths,weaknesses,opportunities and challenge about the develop-ment of pharmaceutical preparations in our hospital,and to explore the development strategies of hospital preparation. RESULTS &CONCLUSIONS:The strengths include the prominent specialty characteristics of clinical key subject,a long R&D history,diverse dosage forms and varieties,definite therapeutic efficacy,etc. The weaknesses contain production capacity shrinkage,deficient con-struction of professional team,external exchange and cooperation deficiency,etc. The opportunities contain national health con-sciousness enhancement,hospital preparation included into local medical insurance reimbursement,etc. The threats contain great difficulty to register new preparation and low preparation pricing,etc. It is suggested to rely on advantage resources to enhance clin-ical formulation development,strengthen human resource management and team building,strengthen cooperation and exchanges to enhance research and development,strengthen the meticulous management to improve operational capacity,in order to formulate scientific development strategy.
4.Pharmacogenetics of individualized treatment of asthma

Asthma, a complex polygenetic disease, has individual differences in patient′s response to asthma drugs,which is mainly related to gene polymorphism. These genes include beta 2-adrenaline receptor genes(ADRB2),the glucocorticoid receptor genes(NR3C1) and leukotriene synthase genes(LTC4S and ALOX5),etc. Pharmacogenomics can realize individualized treatment of asthma,reduce the admission rate and the mortality rate,and reduce the cost of treatment. Pharmacogenomics of children asthma is reviewed in this arti-cle,hoping to provide help for the individualized treatment of asthma.
5.Effect of high glucose on the expression of activating transcription factor 4 in cultured retinal Müller glia cells

Objective To observe the effect of high glucose on the expression of activating transcription factor 4 (ATF4) in cultured retinal Müller glia cells.Methods The retinal tissue of Sprague Dawley (SD) rats was collected,and Müller cells were isolated and cultured.The glial fibrillary acidic protein (GFAP) and glutamine synthetase (GS) of Müller cells were identified by streptavidin-biotinperoxidase complex.Cultured rat Müller cells were divided into control group (5.5 mmol/L glucose),group A (20 mmol/L glucose),group B (30 mmol/L glucose) and group C (40 mmol/L glucose).ATF4 protein expressions in Müller cells of four groups were measured by Western blot four days after cultured.Results GFAP and GS expressed in more than 95% of Müller cells.Over 95% of Müller cells of group A,B and C were positive for GFAP and GS.Western blots indicated that ATF4 protein in group A,B and C increased obviously compared with the control group (q=0.293,0.754,0.484; P<0.05).Conclusion High glucose can increase the expression of ATF4 protein and cause endoplasmic reticulum stress in retinal Müller glia cells in vitro.
8.Protective effects of geranylgeranylacetone on rat retinal ischemia/reperfusion injury

Background Retinal ischemia/reperfusion injury is a common pathological process in ophthalmology.It can cause permanent visual impairment.The prevention and treatment of retinal ischemia/reperfusion injury is one of the focus topics at home and abroad up to now.Objective Present study was to evaluate the protective effects of geranylgeranylacetone(GGA)on retinal ischemia/reperfusion injury.Methods The retinal ischemia/reperfusion models were established in 40 SD rats by the infusion of normal saline solution into the anterior ehamber to elevate the intraoeular pressure for 1 hour and then remove the process.GGA of 2 ml (800 ms/kg)was intragastrieally administered in 20 model rats as GGA group,and the equal volume of normal saline solution was used at the same way in other 20 models as model group.Four normal matched SD rats were served as normal control group.The histological changes of retina in 6,24,48,72 hours after modeling was examined by hematoxylin-eosin staining under the light microscope.and the expression of heat shock protein 70(HSP70)and caspase-3 in retina at the time points mentioned above was detected by immunohistochemistry.Results The edema of cornea and retina of rats appeared in 1 hour and peaked in 24 hours after modeling.In 72 hours after modeling,the edema was alleviated but the disorder of retina structure was found in model group.However,although the pathological change process was similar to that of model group,the damage of retina tissue was milder in the rats of GGA group under the light microscope.HSP70 and caspase-3 were absent in the retina of normal rats.But。The expression of HSP70 in retinal ganglion cells(RGCs)was found in 6 hours after modeling,reached peak in 24 hours and declined after that in model group.The numbers of positive cells for HSP70 in retina was significantly increased in various time points in GGA group,showing statistically significant differences between model group and GGA group(P<0.05).In model group,a few of caspase-3 positive cells were found 6 hours after reperfusion and increased gradually until reaching the peak 24 hours and then decreased gradually 48 hours later.Few caspase-3 positive cells were found 72 hour8 after reperfusion.The numbers of positive cells for caspase-3 decreased in GGA group compared with model group during the experimental duration with the considerable difference between these two groups(P<0.05).Conclusion GGA could protect RGCs against isehemia/reperfusion injury by upregulating the expression of HSP70 and downregulating the expression of caspase-3 in retina.
9.Effects of the position of posterior chamber intraocular lens on visual quality

lntraocular lens ( lOL) often has different degrees of tilt, decentration and rotation in the eye. The deflection of lOL is an important factor to impact on the visual quality of the patients with cataract surgeries. Vision was commonly used to evaluate visual quality before. ln recent years, some new methods are also introduced to evaluate visual quality, such as optical aberration, modulation transfer function ( MTF ) and subjective visual perception. They show that the lOL deflection will cause decreases in eyesight and MTF, cause increase in aberration, lead to ametropia and so on. Therefore, a better evaluation of the effects of lOL deflection on visual quality, has significance for lOL selection, positioning and cataract individualized treatment.
10.Comparison of Electrogastrogram and Gastroscopy in 136 Children with Gastric Disease

Objective To study and evaluate the consistency and diagnostic value of electrogastrogram(EGG) and gastroscopy in children′s gastrosis diagnosis.Methods A total of 136 children were checked with gastroscopy and EGG during the same period,then compared the diagnostic accordance rate with 2 ways,and analyzed the characteristics of EGG.Results EGG showed certain characteristics in different gastrosis.Diagnosis of EGG in the group of functional illness was consistence with gastroscopy at 95.8%;others were less than 40%.Conclusion EGG can be used as a method of special value to diagnose children′s functional gastrosis and a screening means in general gastrosis.
