Objective:To study the expressions of hepatocyte growth factor (HGF) and basic fibroblast growth factor (bFGF) in fresh and two preserved amniotic membranes (AM),and to explore their biological vitality.Methods:The amniotic membranes from the healthy puerperia with abdominal delivery were randomly divided into three groups:fresh group,DMEM/glycerol preserved group at -80℃ and glycerol preserved group at 4℃ (for 1 and 3 months).Immunohistochemical method was used to investigate the changes in the expressions of HGF and bFGF among the three groups.Results:HGF and bFGF were distributed in the epithelium of AMs mostly.The expressions of HGF in both preserved AMs for 1 and 3 months decreased significantly compared with fresh AM (P

OBJECTIVE:To determine the content of hydroxyproline in liver tissue in rats with hepatic fibrosis.METHO_ DS:RP-HPLC was used with precolumn derivation by2,4-dinitrochlorobenzene(DNCB)in the Borax buffer solution for lh in boiling water.Acetonitrile-acetate buffer linear gradient elution was used.Detecting wavelength was360nm.RESULTS:Derivation of hydroxyproline could be separated from other amino acids and accurately determined.CONCLUSION:The method is rapid,accurate,and suitable for detection of biopsy samples obtained by liver puncture.
4.Effects of Anzi Mixture on TLR4/MyD88/NF-κB Signaling Pathway of Antiphospholipid Antibodies Posi-tive Abortive Mice

OBJECTIVE:To study the effects of Anzi mixture on Toll like receptor 4(TLR4)/myeloid differentiation factor 88 (MyD88)/nuclear facter-κB(NF-κB)signaling pathway of antiphospholipid antibodies(APA)positive abortive mice,and to inves-tigate the mechanism of anti-APA positive abortion. METHODS:BALB/c mice(female)were randomly divided into blank control group,model group,aspirin group (positive control,0.0195 g/kg) and Anzi mixture low-dose,medium-dose and high-dose groups (37.7,75.4,150.8 g/kg,calculated by crude drug),with 10 mice in each group. Except for blank control group,other groups were given human β2-glycoprotein Ⅰ as derivant to establish APA positive abortion model. From the first day of pregnancy, treatment groups were given relevant medicine intragastrically,and blank control group and model group were given constant vol-ume of normal saline intragastrically,once a day,for consecutive 9 d. mRNA and protein levels of TLR4,myeloid differentiation 2 (MD2),MyD88 and NF-κB in placental tissue of mice were determined by RT-PCR and immunohistochemical method. RE-SULTS:Compared with blank control group,mRNA and protein expression of TLR4,MD2,MyD88 and NF-κB in placental tis-sue were increased markedly in the model group(P<0.01). Compared with model group,mRNA and protein expression of TLR4, MD2 and MyD88 in aspirin group and Anzi mixture low-dose and medium-dose groups were decreased significantly as well as the protein expression of TLR4 in Anzi mixture high-dose group and the protein expression of NF-κB in all medicine groups(P<0.05 or P<0.01). mRNA expression of TLR4 and MD2 and the protein expression of MD2 and MyD88 in Anzi mixture low-dose groups were lower than those in aspirin group (P<0.05 or P<0.01). CONCLUSIONS:Anzi mixture can inhibit TLR4/MyD88/NF-κB signaling pathway of APA positive abortive mice,which may be one of anti-APA positive abortion mechanisms.

Multiple myeloma is a malignancy of plasma cells,which is still incurable now.Despite dramatic improvement of the response rate and overall survival have been made due to the application of target therapy and stem cell transplantation,the treatment effect is difficult to maintain,nearly all patients will eventually relapse and become refractory.At relapse,the challenge is to select the appropriate treatment for each patient.The decision will be both disease-dependent and patient-dependent.
6.Optimization of Ultrasonic Extraction Technology for Polysaccharide from Aconiti Lateralis Radix Praepa-rata by Uniform Design Method

OBJECTIVE:To optimize ultrasonic extraction technology for polysaccharide from Aconiti lateralis radix praepara-ta(ALRP). METHODS:Uniform design method was applied to investigate the effects of liquid material ratio,ultrasonic time and ultrasonic temperature on extraction rate of polysaccharide from ALRP. Verification test was conducted and compared with the re-sults of conventional decoction and boiling method. RESULTS:Optimized ultrasonic extraction technology was as follow as liquid material ratio of 10 mL/g,ultrasonic time of 34 min and ultrasonic temperature of 73 ℃. The polysaccharide extraction rate in veri-fication test was 19.05%(RSD=0.60%,n=3),relative error of the predicted value (19.44%) was 2.0%,while the extraction rate was higher than decoction and boiling method(16.42%). CONCLUSIONS:Optimized ultrasonic extraction technology is sim-ple,rapid and stable with high extraction rate,which is suitable for extracting polysaccharide from ALRP.
7.Clinical Observation of Acupuncture plus Moxibustion for Chemoradiotherapy-induced Leukopenia

Objective To observe the therapeutic efficacy of acupuncture plus moxibustion in treating chemoradiotherapy-induced leukopenia in patients with malignant tumors.Method A total of 120 patients with malignant tumors who presentedleukopenia during receiving chemoradiotherapy were randomized into two groups. Sixty cases in the treatment group were intervened by acupuncture plus moxibustion; sixty cases in the control group were intervened by oral administration of Batilol tablets and Leucogen tablets.The peripheral white blood cell (WBC) count in the two groups was detected on treatment day 3, 7, 14, 21, and 28.The clinical efficacies of the two groups were compared.ResultThe WBC started toincreaseafter 1-week treatment in thetreatment group (P<0.05) and the increase was statistically significant after 2-week treatment (P<0.01); in the control group, the WBC started to increase after 2-week treatment (P<0.05)and the increase was statistically significant after 3-week treatment (P<0.01); the effect of increasing WBC was more significant in the treatment group compared to the control group (P<0.05,P<0.01).The total effective rate was 90.0% in the treatment group versus 53.3% in the control group, and the between-group difference was statistically significant (P<0.01).Conclusion Acupuncture plus moxibustion can produce a significant efficacy in treating leukopenia induced by chemoradiotherapy.

9.Clinical Observation of Acupuncture-moxibustion for Endometriosis

Objective To observe the clinical efficacy of acupuncture-moxibustion in treating endometriosis (EM). Method Fifty EM patients were randomized into two groups, 25 cases in each group. The treatment group was intervened by acupuncture-moxibustion, while the control group was intervened by oral administration of Mifepristone tablets. Abdominal pain, pelvic lump, and relevant indicators were observed before and after the treatment, and the therapeutic efficacies were compared.Result After 6-month treatment, symptoms including abdominal pain obviously subsided in the treatment group and the total effective rate was 92.0%, versus 52.0% in the control group, and the between-group difference was statistically significant (P<0.01). The pelvic lump size was significantly changed after the treatment in both groups (P<0.05), suggesting that the lump size became smaller in both groups after the treatment;there was a significant difference in comparing the pelvic lump size between the two groups after the treatment (P<0.05), indicating that the lump size was reduced more significantly in the acupuncture-moxibustion group. The level of serum CA125 changed significantly after the treatment in both groups (P<0.05), suggesting that serum CA125 dropped in both groups after the treatment; there was a significant difference in comparing the level of serum CA125 between the two groups after the treatment (P<0.05), indicating that serum CA125 dropped more significantly in the acupuncture-moxibustion group. A year later, there was a significant difference in comparing the relapse rate between the two groups (P<0.05), revealing that the relapse rate was significantly lower in the acupuncture-moxibustion group than in the medication group.Conclusion Acupuncture-moxibustion can produce a marked therapeutic efficacy in treating EM.
10.Therapeutic Observation of Aconite-cake-partitioned Moxibustion plus Acupuncture in Treating Peptic Ulcer

Objective To observe the efficacy and prognosis of aconite-cake-partitioned moxibustion plus acupuncture in treating peptic ulcer.Method Thirty-four patients in the treatment group were intervened by aconite-cake-partitioned moxibustion plus acupuncture, while thirty-three patients in the control group were by Omeprazole Enteric-coated Capsules. The clinical efficacy, the effect on ulcer under gastroscope and Hp, and the therapeutic efficacy 12 months after the intervention were evaluated.Result The total effective rate was 91.2% in the treatment group versus 72.7% in the control group, and the difference was statistically significant (P<0.05). For ulcer by gastroscopy, the total effective rate was 85.3% in the treatment group versus 69.7% in the control group, and the difference was statistically significant (P<0.05). After intervention, the Hp level didn’t show significant change (P>0.05). The relapse rate of the control group was higher than that of the treatment group 12 months after the intervention.Conclusion Aconite-cake-partitioned moxibustion plus acupuncture can produce a higher efficacy than Western medication in treating peptic ulcer.
