3. Fucoidan induces apoptosis of multiple myeloma cells by inhibiting Ras-p38MAPK and Shh-GLI1 signaling pathways

Objective: To investigate the effect of fucoidan on multiple myeloma U266 cells and its possible molecular mechanism. Methods: The multiple myeloma U266 cells were treated with 10, 25 and 50 μg/mL fucoidan for 24, 48 and 72 h, respectively. The growth inhibitory rate of U266 cells was determined by cell counting kit-8 (CCK-8) assay. The apoptosis rate was measured by flow cytometry. The expression levels of RAS, p38, phospho-p38, sonic Hedgehog (Shh) and glioma-associated oncogene homolog 1 (GLI1) were detected by Western blotting. Results: The growth of multiple myeloma U266 cells treated with different concentrations of fucoidan (10, 25 and 50 μg/mL) was inhibited only in a dose-dependent manner. The apoptosis rates of U266 cells treated with 10, 25 and 50 μg/mL fucoidan for 24 h were (6.73±2.30)%, (9.12±1.90)% and (20.13±2.10)%, respectively, which were higher than that of the U266 cells without fucoidan intervention [control: (4.08±1.60)%; P < 0.05). The expression levels of RAS, phospho-p38, Shh and GLI1 proteins in U266 cells treated with fucoidan were significantly lower than that of the control (P < 0.05). Conclusion: Fucoidan inhibits proliferation and induces apoptosis of U266 cells by inhibiting Ras-p38MAPK and Shh-GLI1 signaling pathways. Copyright © 2013 by TUMOR.
7.Relationship between psychological state and the mode of delivery in pregnant women

Objective To investigate the dynamic changes of psychological states and the results of delivery in pregnant women,simultaneously,study the effective way to reduce the rate of cesarean section.Methods Through applying the self-compiled test paper,the self-evaluating state table(SCL-90)and the table of overall feeling of happiness,we assessed the psychological states of 89 pregnant women.The results were recorded at 35 and 38 weeks of gestation and 72 hours postpartum.Results 48 delivered vaginally(53.93%).4 were managed by cesarean section(46.07%),including 12 non-indicated cesarean section(29.27%).The mark of anxiety,terror and depression factors increased in the group of non-indicated cesarean section before and after dilivery compared with the group of vaginal delivery and indicated cesarean section.There was remarkable difference(P<0.01).The value of overall feeling of happiness in the group of non-indicated cesarean section was less than that in the group of vaginal delivery.The difference was extremely significant(P<0.001).The rdevant analysis showed that the feeling of happiness,the desire of labor and educational level were related negatively to the level of symptom during pregnancy(P<0.01).The mode of delivery was related positively to the psychological state of pregnant women(P<0.01).Conclusion Non-indicated cesarean section is mainly interrelated to anxiety,terror and depression that proceed gradually during pregnancy and delivery.So it is suggested that the aimed psychological education be beneficial to decrease the rate of cesarean section.
8.Effect of psychological counseling skills on nurse-patient communication

Objective To explore the effect of psychological counseling skills on medical treatment and satisfaction degree of patients in hospital.Methods Toally 60 discharged patients during December 2015 were assigned as the control group and another discharged one during January 2016 as the intervention group.Training on psychological counseling skills was done for the nurses by the psychological counselor in the intervention group while the control group was not.The two groups were compared on the basis of investigation with the patient's consultation experience scale as well as the medical satisfaction questionnaire.Result The satisfaction levels of consultation experience and medical consultation satisfaction in the intervention group were both higher than those of the control group (all P<0.01).Conclusion Psychological counseling skills used in the nurse patient communication can enhance the patient's medical experience and their satisfaction with medical treatment.
10.Pathological changes of nasal mucosa after instillation of 37℃ normal saline in rats

[ABSTRACT]OBJECTIVE To obser ve t he pathological changes of nasal mucosa in rats after instillation of 37℃ normal saline. METHODS Forty-eight qualified rats were given instillation of 37℃ nomal saline 30 times, once per minute. Another 2 qualified rats served as control. After instillation, in the 15, 30 minutes and 1st, 3rd, 7th, 14th, 42th, 28th day respectively, the nasal septum mucosa of 6 rats was observed by light and electron microscopy. The corresponding area of the mucosa of control group was also observed by light and electron microscope. RESULTS Under light microscope, the arrangement of cell was disordered in the 1st, 3rd, 7th days. But there is no damage of vascular gland structure. After 14 days, the disorder recovered. With electron microscope, edema of cells, expansion of perinuclear gap, disorder of cilia and microwilli was found in the 30 minutes, 1st, 3rd day. All these began recover in the 7th days, and completely restored in the 28th day. CONCLUSION 37℃ nomal saline drip can damage nasal mucosa, but the damage is light, which is characterized by disorder and shortage of cilia and microvilli, edema of the epithelial cells. And the damage can recover quickly. It began in the 30 minutes after instillation, reached its peak in the 3rd day. And the recovery began in the 7th day, and completed in the 28 day.
