1.Advances in Study on Effects of Helicobacter pylori Eradication Therapy on Intestinal Microecology

It is documented that nearly half of the global population have been infected with Helicobacter pylori (Hp).In Chinese consensus report on management of Hp infection,bismuth quadruple therapy composed of proton pump inhibitor,bismuth and two antibiotics is recommended as the first-line Hp eradication regimen.However,large doses of oral antibiotics and PPI often disrupt the well-balanced host-microbial symbiotic state,and impair the protective effect of biological barrier.Furthermore,the alteration of microbial composition secondary to eradication therapy might be associated with the increased adverse effects of Hp eradication therapy and re-infection.This paper summarized the advances in study on effects of Hp eradication therapy on intestinal microecology and the advantages of probiotics supplementation in Hp eradication therapy.
2.Evaluate the effect of analgesia after total knee arthroPlasty of different analgesia models

Objective To compare the effect of postoperative analGesia of the continuous femoral nerve block analGesia( CFNB),continuous epidural analGesia( CEA)and intravenous analGesia( CIA)for patients with total knee arthroplasty. Methods The clinical informations of l44 four patients who underwent line unilateral total knee replacement surGery from April 20ll to May 20l2 were retrospective analyzed. AccordinG to the way of postoperative analGesia,the patients were divided into different CFNB Group( n=70 ),CEA Group( n=36 )and CIA Group(n=38). Record the pain score of 24 h,48 h and 72 h after surGery and adverse reaction. Results VAS score of CFNB Group and CEA Group at 24 h and and 48 h after surGery were siGnificantly lower than in CIA Group( P<0. 0l). While there was no siGnificant difference amonG three Groups in term of VAS score at the 72 h after surGery. Adverse reactions of postoperative Gastrointestinal and spiritual nerve symptoms of CFNB Group were siGnificantly lower than in CEA Group and CIA Group( P<0. 0l ). Postoperative catheter related problems of CFNB Group were siGnificantly hiGher than CEA Group and CIA Group( P<0. 05 ). Conclusion AnalGesic effect of CFNB and CEA for after total knee arthroplasty is Good,and better than that of CIA. But CEA and CIA have Greater adverse reactions.
3. Preparation of compound vitamin microcapsules using cross-linked gelatin as wall material

OBJECTIVE: To study the preparation and in vitro release profile of compound vitamin microencapsules using cross-linked gelatin as wall material. METHODS: At the presence of hydrogen peroxide, the cross-linked gelatin was prepared with ferulic acid as the cross-linking agent. During this reaction, horseradish peroxidase was used as the catalyst. Influences of reaction conditions on the cross-linking degree were investigated. Compound vitamin microcapsules were prepared by spray-drying technique using the obtained cross-linked gelatin as wall material. The core material was the mixture of thiamine hydrochloride (vitamin B1), riboflavin (vitamin B2), pyridoxine hydrochloride (vitamin B6), folic acid and nicotinamide. The effect of the ratio of wall material to core material on the encapsulation efficiency and loading of the vitamins were investigated. The size and surface morphology of the compound vitamin microcapsules were characterized. The encapsulation efficiency, loading and in vitro release property of the core material were determined by fluorospectrophotometry. RESULTS: A comparatively high cross-linking degree (ca. 10%) of cross-linked gelatin was obtained under the following reaction conditions:temperature of 40°C, pH value of 8.0, gelatin concentration of 9% (W/V), ferulic acid concentration of 40 mmol·L-1 and reaction time of 24 h. The vitamins were embedded by the cross-linked gelatin and the encapsulation efficiency was more than 85%. Scanning electron microscopy (SEM) study showed that the compound vitamin microcapsules had a regular spherical shape but the majority presented rough surfaces or dents. Particle size analysis indicated that the microcapsules had a mean diameter of 15.27 μm. At the ratio of coating material to core material was 10/1 (W/W), the vitamins encapsulated with the cross-linked gelatin released completely in 30 min in simulated gastric fluid, and they released completely in 16 min in simulated intestinal fluid. They released slower than the vitamins encapsulated with the gelatin accordingly. CONCLUSION: Compound vitamin microencapsules with high encapsulation efficiency and sustained release effect can be obtained by spray-drying using cross-linked gelatin as wall material.
6.Granulocyte Macrophage Colony Stimulating Factor as an Adjunctive Therapy for Acute Myeloid Leukemia:A Systematic Evaluation

OBJECTIVE:To evaluate the curative efficacy of granulocyte macrophage colony stimulating factor (GM-CSF) as an adjunctive therapy for the treatment of acute myeloid leukemia (AML). METHODS: Randomized controlled trials (RCTs) of GM-CSF in the treatment of AML were retrieved from PubMed,EMBase,Cochrane Library,CBMdisc and CNKI database by computer as well as from the leukemia-related journals manually for a quality evaluation of the methodology and Meta analysis using RevMan5.0 software. RESULTS: A total of 10 RCTs were included,of which,3 RCTs were graded as A,5 as B and 2 as C. The results demonstrated that there were no significant differences of indexes among groups as complete remission (CR) rate,relapse rate,infection rate and length of hospital stay,etc. CONCLUSION: The available clinical research results showed that GM-CSF adjunctive therapy could neither increase CR rate,nor decrease the relapse rate,infection rate or length of hospital stay.

8.Effect of sodium tanshinone II (A) sulfonate on Ang II -induced atrial fibroblast collagen synthesis and TGF-beta1 activation.

OBJECTIVETo observe the effect of sodium tanshinone II (A) sulfonate (STS) on Ang II -induced atrial fibroblast collagen synthesis and TGF-beta1 activation.
METHODAtrial fibroblasts of neonatal rats were cultured to determine the content of collagen protein. The original synthesis rate determined by the [3H]-proline incorporation method was taken as the index for myocardial fibrosis. The content of active TGF-beta1 and total TGF-beta1 in cell culture supernatants were tested and cultured by ELISA. The expression of thrombospondin-1 (TSP-1) was assessed by using Western blot.
RESULTAng II could significantly increase the content of atrial fibroblast collagen and the collagen synthesis rate, the TSP-1 expression and the concentration of active TGF-beta1, without any obvious change in total TGF-beta1. After the STS treatment, all of the indexes, apart from total TGF-beta1, were obviously down-regulated.
CONCLUSIONSTS could decrease the secretion of Ang II -induced atrial fibroblast collagen and the synthesis rate. Its mechanism is related to the inhibition of TSP-1/TGF-beta1 pathway.

10.Relationship between JAK2-V617F Gene Mutation in Peripheral Blood Mononuclear Cells and Thrombotic Events in Patients with Myeloproliferative Neoplasms

Objective To analyze the correlation between mutation in JAK2-V617F gene in peripheral blood mononuclear cells and thrombotic events in patients with myeloproliferative.Methods Investigation of thrombosis in 391 patients with MPN,using QRT-PCR to detect JAK2-V617F gene mutation in peripheral blood of patients.Result 105 cases of thrombotic events in 391 MPN patients (26.9%),84 cases of thrombotic events in 273 JAK2-V617F gene mutation positive patients (30.8 %),however,21 cases of thrombotic events in 118 J AK2-V617F gene mutation negative panents (17.8 %).The difference between the two groups was statistically significant (x2 =32.30,P<0.01).In the PV group,45 cases of thrombosis events in JAK2-V617F positive patients (29.2 %),6 cases of thrombosis events in JAK2-V617F negativepatients (42.9%),there was significant difference between the two groups (x2 =2.13,P>0.05).In the ET group,18 cases of thrombosis events in JAK2-V617F positive patients (32.2%),9 cases of thrombosis events in JAK2-V617F negative patients (15.8%),there was significant difference between the two groups (x2 =12.32,P<0.01).In the PMF group,21 cases of thrombosis events in JAK2-V617F posinve patients (33.3 %),6 cases of thrombosis events in JAK2-V617F negativepatients (12.8 %),there was significant difference between the two groups (x2 =14.32,P<0.01).Conclusion JAK2-V617F positive MPN patients were more susceptible to thrombotic events especially in patients with ET and PMF.
