1.Inhibitory effect of ciprofloxacin on proliferation of cultured fibroblast in human conjunctiva

Objective:To estimate the anti-proliferative effect of ciprofloxacin on the cultured human conjunctiva fibroblast.Methods:The cultured human conjunctiva fibroblasts were treated with different concentrations of ciprofloxacin.The inhibitory effect of ciprofloxacin was measured with MTT.Results:Ciprofloxacin induced a dose-dependent inhibition for growth of cultured human conjunctiva fibroblast.Conclusion:Ciprofloxacin can prevent the development of PVR effectively,which provides an evidence for the use of ciprofloxacin in clinic.

OBJECTIVE:To study the inhibiting effects of ligustrazine and ciprofloxacin on proliferation of cultured human fibroblasts of conjunctiva.METHODS:The human fibroblast cells of conjunctiva were treated with different concentrations of ligustrazine and ciprofloxacin.The inhibitive effects was measured with MTT.RESULTS:Both ligustrazine and ciprofloxacin induced inhibition of growth of cultured human fibroblasts of conjunctiva in a concentration-dependent manner.CONCLUSI_ ON:Ligustrazine and ciprofloxacin can prevent the development of PVR effectively and may provide a clue to use them in the clinical practice.

Post-transcriptional regulation of gene expression is an important contributor to disease pathogenesis.MicroRNAs (miRNAs) are endogenous, conserved, single-stranded non-coding small RNA with 18-25 nucleotides in length, which regulate gene expression post-transcriptionally.Several studies found that miRNAs could affect the development of nonalcoholic fatty liver disease through multiple ways.This article reviews the roles of miRNAs in the development of nonalcoholic fatty liver disease by regulating lipid metabolism, insulin resistance, oxidative stress, inflammatory, intestinal mucosal barrier, cell apoptosis and so on.
6.Causes of job burnout in medical professionals and relevant intervention strategies

Job burnout has been a common problem in modem society, and medical professionals are vulnerable to it. The causes of burnout in medical professionals involve factors of social environment and system, institutional factors, occupational factors and personal factors. The intervention strategies at multiple levels are put forward, including to advocate concerning about the well-being of others and attaching importance to cultural value orientation of qualitative life-style, to construct harmonious doctor- patient relationship and good working environment, to enhance social public relations and establish good media image, to improve management system and operational mechanism at organizational level, and to enhance the coping ability at individual level by training.
7. Abnormalities in glucose metabolism and pancreatic endocrine function induced by 5-fluorouracil in vitro and in vivo

Objective: To explore the effects of 5-fluorouracil (5-FU) on glucose metabolism and the endocrine function of pancreatic beta-cells. Methods: The NIT-1 cells were treated with different concentrations (5.0, 10.0, 20.0 and 40.0 mg/L) of 5-FU. The insulin concentration in the culture medium was determined by radioimmunoassay. Twenty Wistar rats were divided into 5-FU group (intraperitoneally administered with 5-FU at a daily dose of 20 mg/kg for 5 days) and the control group (intraperitoneally administered with normal saline). Glucose tolerance test was performed on d 2 and d 7 following 5-FU administration, and the plasma insulin concentration was determined by radioimmunoassay. Results: The high glucose (16.7 mmol/L)-induced release of insulin from NIT-1 cells was inhibited by 5.0 to 20.0 mg/L of 5-FU in a dose-dependent manner (P<0.01). Compared with the control group, on d 2 and d 7 following 5-FU administration, the plasma glucose levels were markedly increased at 30, 60, 120 and 180 min following a glucose load (P<0.01); however, the fasting plasma insulin levels were not significantly elevated or decreased (P>0.05). Postprandial plasma insulin level at 60 min following a glucose load on d 7 was significantly lower than that of the control group on d 2 (25.32 ± 6.07 vs 33.98 ± 4.94, P<0.05). The peak secretion time of plasma insulin was delayed to 120 min. Conclusion: Hyperglycemia following 5-FU treatment appears to be related to a relative deficiency in insulin secretion response to glucose stimulation. 8.Research on Herbal Medicine Application Regulation of Professor Wu Liangcun in Gastric Cancer Treatment

Objective]To summarize herbal medicine application regulation of Professor Wu Liangcun in gastric cancer treatment. [Methods]Collect 116 gastric cancer patients from Chinese medicine clinic system during January 1,2014 to December 31, 2014 and establish clinical prescription underlying database. Apply descriptive analysis to drug cumulative frequency, drug-induced taste and nature and apply cluster analysis to 42 drugs using more than 10 times. [Results] In the 116 gastric cancer patients,159 kinds of herbs were used ,the cumulative frequency of herbal medicine was 1816.GanCao was the herb with highest usage frequency, and then was FuLing, ShanYao, TaiZishen;According to accumulated frequency, herbs with tonifying function were at the top position(herbs with tonifying qi function were most), and then was the herbs with function of herbs with the function of clearing excessive heat, herbs with function of herbs with the function of relieving dyspepsia, herbs with function of herbs with the function of regulating qi. In terms of herbs' nature and taste, herbs with sweet taste, plain nature were used most. Cluster analysis divided all used herbs into 5 sections, involving prescriptions Sijunzi decoction, Erchen decoction, and couplet medicines DaoYa with GuYa, JiNeijin with MiHoutao, SheShecao with XianHecao and so on. [Conclusions] Professor Wu Liangcun treats gastric cancer with the law of tonifying qi, the main prescription is the same as Sijunzi decoction, emphasizing nourishing Yin and clearing away the heat-evil, protecting gastric, accompanied with promoting digestion and regulating qi.
9.Stability of Reduced Glutathione Sodium in Four Different Injections

OBJECTIVE:To investigate the stability of reduced glutathione sodium after mixing with glucose injection (5% or 10%),glucose and sodium chloride injection or 0.9% sodium chloride injection. METHODS:After mixing of reduced glutathione sodium with glucose injection (5% and 10%),glucose and sodium chloride injection,and 0.9% sodium chloride injection,respectively,the 4 mixtures at 0,0.5,1.0,1.5,2.5,3 h after mixing which were stored respectively in 25℃ or 37℃ thermostatic bath were observed in respect of the changes in appearance,pH values,absorbability,contents and the insoluble particles. RESULTS:After mixing of reduced glutathione sodium with glucose injection (5% and 10%),glucose and sodium chloride injection,and 0.9% sodium chloride injection,respectively,the appearance and pH values of the 4 mixtures showed no obvious change,but the contents and the insoluble particles of the 4 mixtures experienced changes of different degree. CONCLUSION:Reduced glutathione sodium is stable in glucose injection (5% or 10%),glucose and sodium chloride injection or 0.9% sodium chloride injection within 2h after mixing.
10.Differentiation of vitreous proteome in human eye with proliferative Vitreoretinopathy

Background Proliferative vitreoretinopathy (PVR) is a a common cause of anatomic failure in retinal detachment surgery.Proteomics is the critical method to investigate the different proteins. Objective This study was to search for related vitreous proteins in rhegmatogenous retinal detachment patients with proliferative vitreoretinopathy (PVR)and screen for the related protein expression in the vitreous with PVR. Methods Vitreous samples were obtained from 8 moderate PVR (grade B)eyes and 8 severe PVR(grade C or D)eyes during pars plana vitrectomy (PPV)for the proteomics analysis by two-dimensional gel electrophoresis coupled with mass spectrometry.Normal vitreous from 8 healthy donor eyes served as control.First dimension isoelectric focusing (IEF) was performed with the Pharmacia IPGphor in solvent B using 18 cm non-linear pH 3-10 immobilized pH gradient (IPG)strips.IPG strips were rehydrated with sample,and then IEF was performed.The second dimension 12%sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis (PAGE) was performed after the IPG strips were equilibrated.After silver staining,the gels were analyzed by the 2-DE gel analysis software.The matched spots were excised and tryptica digested in-gel.The peptides were analyzed by MALDI-TOF-MS and the MS/MS spectral were searched against the human protein databases using MASCOT.This study process complied with the Declaration of Helsinki.Wfitten informed consent was obtained from each patient prior to this trial. Results In the current study,a total of 47,184 and 336 protein spots were detected from the normal donor eyes,moderate PVR eyes and severe PVR eyes,respectively,by 2-DE gels.Among 13 protein spots with significant differences in the 3 groups,7 types of proteins were successfully identified.The results indicated that Enolase 2 was a specific protein for normal donor vitreous.whereas transthyretin monomer and retinol-binding protein(RBP) chain B were distinct proteins found in severe PVR vitreous.and prostaglandin D synthase and RBP3 precursor were common proteins with up-regulated expression in vitreous samples with moderate PVR,but down-regulated levels in vitreous samples with severe PVR.The levels of albumin and transferrin in vitreous showed dynamic elevation with the enhancement of severity of PVR.Conclusion The changes in the vitreous proteome imply that some types of vitreous proteins degrade and some common serum proteins are expressed in the vitreous body with PVR,which indicated the disruption of the blood retinal barrier in the pathogenesis of PVR.
