2.Expression of Notch2,Notch4 in Hyperoxia Induced Lung Injury in Premature Rats

Objective To explore the roles of Notch2,Notch4 in hyperoxia induced lung injury in premature rats.Methods At the postnatal 1 day Sprague-Dawley premature rats were randomly assigned to about 85% hyperoxia group and air group.At the 1,7,14,21 days after exposed,8 rats of each group were used to evaluate expressions of Notch2,Notch4 in lungs by immunohistochemistry and the level of Nothch2,Notch4 mRNA by reverse transcription polymerase chain reaction(RT-PCR).Results Expressions of Notch2,Notch4 had their rules in rats′ different stages of development;85% oxygen exposed would change their tracks.Conclusion Prolonged 85% oxygen exposure result in abnormal expressions of Notch2 and Notch4 ,which is likely to lead to the pathogenesis of hyperoxic lung injure in premature rats.
4.Expression of TRAP1 in esophageal carcinoma and its influence on the prognosis

Purpose To investigate TRAP1 expression in esophageal cancer and its relationship with clinicopathological features and prognosis.Methods Expression levels of TRAP1 in 60 pairs of cancer and adjacent normal tissues were detected by immunohistochemistry,and the relevance between TRAP1 and clinicopathological features was evaluated.Kaplan-Meier and Cox proportional regression analyses were performed to determine the association of TRAP 1 expression and survival of the patients.Results The positive expression rate of TRAP1 was 55.0%,and the amount of relative protein transcript level was 2.7 ± 1.1 in the cancer tissue.The positive expression rate of TRAP1 was 11.7%,and the relative expression protein was 0.5 ± 0.4 in the adjacent normal tissue.The expression level of TRAP1 in cancerous tissue was significantly higher than that of tissue adjacent to carcinoma.There was no statistically significant difference between the expression levels of TRAP1 with sex,age,location,and degree of differentiation (P > 0.05).There was statistically significant relationship between recurrence,metastasis and TNM stages with the expression of TRAP1 levels (P < 0.05).The average survival time of TRAP1-negative patients was 69.0 months (95% CI:60.2-77.9) while the TRAP1-positive patients was 34.2 months (95% CI:24.4-44.1).High levels of TRAP1 were correlated with decreased survival of postoperative esophageal cancer patients (P < 0.05).Conclusion TRAP1 is overexpressed in esophageal cancer and associated with the progression and prognosis of esophageal cancer.

9.Comparison of Endoscopic Clips Ligation and Snare Resection in Treating Giant Colonic Polyps

2 cm) who were treated with endoscopic polypectomy in 2 different means,respectively.Methods The clinic data of 68 children with giant colonic polyps were review analyzed.Thirty-five cases were received endoscopic clips ligation and the other 33 cases were received endoscopic snare resection.Results All the 35 cases out of endoscopic clip ligtion group were sucessfully cured.There were only 3 cases showedl a little bleeding in this group.In the endoscopic snare resection group,there were 10 cases showed bleeding,8 cases showed polypectomy syndrome,1 case transferred into(operation).Conclusions The complication incidence in endoscopic clips ligation group is lower than that in endoscopic snare resection group.The endoscopic clipping brings about an effective and safe way to treat giant colonic polyps in children.
10.Effects of high blood glucose fluctuation on DNA damage of diabetic rat retinal tissues

AIM:To observe the situation of rat retinal tissue DNA damage at early diabetic period, discuss the role of the blood glucose fluctuations, and provide a new method for studying the pathogenesis of diabetic retinopathy ( DR) .
METHODS: SD rats were randomly divided into four groups:normal control group (NC), normal fluctuation group ( NF ) , diabetes group ( DM ) and diabetes fluctuation group ( DF ) . Diabetic models were established through intraperitoneal injection of STZ. A certain amount of glucose was injected in the rats of group NF and DF in an intraperitoneal mode three times a day after the model was established, thereby causing blood glucose fluctuations. Rats were killed and the retinal tissues were taken in the 8th week. Single cell gel electrophoresis ( SCGE ) technique was adopted for detecting DNA injury extent in the retina tissue.
RESULTS:Groups NF and DF showed significant and regular fluctuations. The curve of blood glucose fluctuations was relatively stable. All values of MBG, SDBG, LAGE and M were significantly increased compared with group NC. Group DF was increased more significantly. It was statistically significant (P<0. 01). SCGE showed that there were DNA damages in different levels in the cells of group NF, DM and DF. Indicators of cells such as TL, TDNA %, TM, OTM were higher than that in group NC. It was statistically significant ( P<0-01). The comparison difference between two groups was also significant (P<0. 01).
CONCLUSION: Rat retinal tissues have DNA injury during early diabetic period. DNA injury is gradually aggravated with blood glucose fluctuation. It indicates that high blood glucose and blood glucose fluctuation are involved in the mechanism of cell DNA injury, and they may be one of DR early event, have played a certain role in the incidence of DR.
