1.Clinical effect of laparoscopic common bile duct exploration combined with endobiliary drainage in treatment of acute biliary pancreatitis

Objective To evaluate the clinical effect of laparoscopic common bile duct exploration combined with endobiliary drainage in the treatment of acute biliary pancreatitis.Methods A retrospective analysis was performed on the clinical data of 90 patients with acute biliary pancreatitis admitted to our hospital from January 2012 to May 2014.Among them,42 patients received laparoscopic common bile duct ex-ploration combined with endobiliary drainage (laparoscopic group),and 48 patients underwent endoscopic retrograde cholangiopancreatogra-phy (ERCP)combined with endoscopic sphincterotomy (ERCP group).The operation situation,changes in white blood cell (WBC) count,total bilirubin (TBil),alanine aminotransferase (ALT),and C -reactive protein (CRP)after operation,and follow -up results were analyzed.Comparison of continuous data between the two groups was made by t test,while comparison of categorical data was made by chi -square test.Results There were no significant differences in operation time,intraoperative blood loss,hospitalization expenses,and the incidence of complications between the two groups (all P >0.05).On the 1st,3rd,and 7th days after operation,the two groups had re-duced WBC count and serum levels of TBil,ALT,and CRP,but the reduction was significant only on the 3rd and 7th days in both groups (all P <0.05).On the 3rd day,the laparoscopic group had significantly lower WBC count and CRP level than the ERCP group (both P <0.05),but none of all the indices showed significant differences between the two groups at other time points (all P >0.05).In the 6 -month follow -up,the incidence of regurgitation cholangitis in the ERCP group was 6.25% (3 /48),but no complications were found in the laparoscopic group (χ2 =2.716,P =0.099).Conclusion Laparoscopic common bile duct exploration combined with endobiliary drainage is safe and feasible for acute biliary pancreatitis,and it can retain the function of the Oddi′s sphincter.
2.Effect of histone lysine methtyltransferase G9a in sevoflurane-induced long-term cognitive impairments in the developing brain of neonatal rats

Objective To investigate the role of histone lysine methyltransferase G9a in sevoflurane-induced cognitive impairment in the developing brain of neonatal rats.Methods Thirty-six 5-day-old male SD rats were randomly divided into 3 groups (n =12):control group,sevoflurane group and Bix01294 (the inhibitor of histone lysine methyltransferase G9a)group.The rats in the sevoflurane group and the Bix01294 group received 3% sevoflurane anesthesia for 2 hours once a day at postnatal 5-7 days (P5-P7 ).The rats in the Bix01294 group received Bix01294 (10 mg/kg)subcu-taneously at 1 5 min before anesthesia,and the rats in the control group and sevoflurane group received normal saline for injection (0.1 ml)at the same time.The open-field test and fear condition-ing test were performed at P3 5 and P3 9-P41 ,respectively.The tissues of hippocampus were collected at P42 to measure the levels of G9a,histone H3 lysine 9 dimethylation (H3K9me 2 )and synapsin 1. Results Compared with the control group,the percentage of freezing time of sevoflurane group was significantly decreased in the contextual fear condition test,while the levels of G9a and H3K9me 2 were significantly increased and the level of synapsin 1 was significantly decreased (P <0.01).How-ever,the percentage of freezing time of Bix01294 group was significantly increased,while the levels of G9a and H3K9me 2 were significantly decreased and the level of synapsin 1 was significantly in-creased compared with the sevoflurane group (P <0.05).There was no difference in the total distance and residence time in the central grid in the open-field test,and the percentage of freezing time in the cued fear condition test among the three groups.Conclusion Histone lysine methyltransferase G9a is involved in the sevoflurane-induced long-term cognitive impairment in developing brain of neonatal rats,which may be associated with the increase of H3K9me 2 and the down-regulation of synapsin 1 in the hippocampus.
3.Changes of serum GP73 after hepatectomy and its relationship with recurrence in hepatocellular carcinoma patients

Objective To investigate changes of GP73 after hepatectomy and its correlations with hepatocellular carcinoma (HCC) recurrence. Methods Perioperative serum GP73 was monitored in hepatic hemangioma and HCC patients undergoing hepatectomy. Clinicopathologic features and follow-up results were collected to evaluate the relationship between serum GP73 level and patients' prognosis.Results There was no statistical difference between preoperative GP73 and postoperative GP73 in hepatic hemangioma group.While preoperative GP73 in HCC group was 9.9(3.7 - 15.8) relative unit (RU),and that on POD3 (postoperative day 3 ) was 9.1 ( 3.4 - 13.3 ) RU,on POD7 was 74.3 ( 1.7 - 9.0) RU,on POD14 was 3.3(2.1 -5.4) RU ( F =72.606,P < 0.001 ).HCC recurred in 21 cases during follow-up,GP73 in recurrent cases [ 11.0 (8.4 - 13.8 ) RU ] was significantly higher than postoperative trough values while it was not different from their preoperative GP73 level [ 9.9 ( 2.9 - 15.0) RU ] ( Z =1.185,P >0.05). The preoperative GP73 level between recurrent subgroup and nonrecurrent subgroup was not significantly different (Z =- 1.546,P > 0.05 ).Preoperative GP73 did not correlate to patients' survival.Conclusions Hepatectomy for HCC leads to a significant decrease of GP73 and postoperative HCC recurrence accompanies reelevation of GP73. GP73 could be used as a postoperative monitor for HCC recurrence.
4. The mechanism of Annexin A1 regulating the proliferation, apoptosis and migration of bladder cancer cells

Objective:
Explore the function and regulatory mechanism of Annexin A1 (ANXA1) in bladder cancer cell proliferation, apoptosis and migration.
Methods:
From February 2018 to June 2019, we use T24 cells as the model and divide it into over-expression control group (ctrl), ANXA1 over-expression group (ANXA1), knockdown control group (shctrl), ANXA1 knockdown group 1 (shANXA1-1), ANXA1 knockdown group 2 (shANXA1-2) and ANXA1 knockdown group 3 (shANXA1-3). 24 hours after the culture, the cells were collected and the mRNA expression level of ANXA1 was detected by Real-Time quantitative PCR. The cell activity was detected by CCK-8; the cell apoptosis and cycle were detected by flow cytometry. The cell migration was detected by Transwell assay.
Results:
The Real-Time quantitative PCR showed that the expression of ANXA1 in the over expression group was significantly higher than that in the over expression control group (15 369.00±874.20 and 1.00±0.07,
5.Effect of enhanced UV-B radiation on photosynthetic structure and photosynthetic characteristics of Mentha piperita.

OBJECTIVETo reveal the effects of UV-B radiation on the growth of medical plant Mentha piperita, simulate an enhanced UV-B radiation and evaluate intensity of radiation on the photosynthesis of M. piperita.
METHODThree different levels of UV-B radiation were set in the experiment which included: natural light control (0 W x m(-2)), light UV-B radiation stress (0.15 W x m(-2)) and heavy UV-B radiation stress (0.35 W x m(-2)). The chloroplast ultrastructure, photosynthesis indexes and chlorophyll fluorescence parameters of the M. piperita were observed under the three treatments.
RESULTAlthough the chloroplast ultrastructure was destroyed to some degree under the light UV-B radiation stress, F(v)/(F)m, F(v)/F(o), qP, phiPS II and ETR could resume to the comparative level of natural light control. At the same time, qN increased firstly and decreased thereafter. But under the high strength UV-B radiation stress, the photosynthetic structures were badly destroyed, which could not recover through protecting mechanism by itself.
CONCLUSIONIt was showed that M. piperita was able to protect photosynthetic structures by increasing respiration and dissipation when photosynthetic capacity reduced under light UV-B radiation stress. It is demonstrated that M. piperita has high adaptation to light UV-B radiation stress, which is kind of promising medical plant for area with higher UV-B radiation.

6.Diagnosis and treatment of genetically related urinary calculi

In recent years, the incidence of single-gene nephrolithiasis has been increasing year by year. With the application of whole-genome analysis and whole-exome sequencing technology, the etiology of single-gene mutations leading to the development of urinary calculi has been extensively verified. Therefore, this article reviews the research on urinary calculi-related genetic diseases at home and abroad, and introduces transport proteins and channels; ions, protons and amino acids. The role of urinary calculi in the majority of clinicians realizes the significance of genetic testing in such diseases, thereby increasing the understanding of genetically related urinary calculi and improving the level of clinical diagnosis and treatment.

BACKGROUND:
LY01005 (Goserelin acetate sustained-release microsphere injection) is a modified gonadotropin-releasing hormone (GnRH) agonist injected monthly. This phase III trial study aimed to evaluated the efficacy and safety of LY01005 in Chinese patients with prostate cancer.
METHODS:
We conducted a randomized controlled, open-label, non-inferiority trial across 49 sites in China. This study included 290 patients with prostate cancer who received either LY01005 or goserelin implants every 28 days for three injections. The primary efficacy endpoints were the percentage of patients with testosterone suppression ≤50 ng/dL at day 29 and the cumulative probability of testosterone ≤50 ng/dL from day 29 to 85. Non-inferiority was prespecified at a margin of -10%. Secondary endpoints included significant castration (≤20 ng/dL), testosterone surge within 72 h following repeated dosing, and changes in luteinizing hormone, follicle-stimulating hormone, and prostate specific antigen levels.
RESULTS:
On day 29, in the LY01005 and goserelin implant groups, testosterone concentrations fell below medical-castration levels in 99.3% (142/143) and 100% (140/140) of patients, respectively, with a difference of -0.7% (95% confidence interval [CI], -3.9% to 2.0%) between the two groups. The cumulative probabilities of maintaining castration from days 29 to 85 were 99.3% and 97.8%, respectively, with a between-group difference of 1.5% (95% CI, -1.3% to 4.4%). Both results met the criterion for non-inferiority. Secondary endpoints were similar between groups. Both treatments were well-tolerated. LY01005 was associated with fewer injection-site reactions than the goserelin implant (0% vs . 1.4% [2/145]).
CONCLUSION:
LY01005 is as effective as goserelin implants in reducing testosterone to castration levels, with a similar safety profile.
