1.Ultrastructural Changes of Cochlea Nerve Following Gunshot Wound in the Maxillofacial Region

This experiment was done by using the Sweden trauma model. Ten dogs, divided into two experiment groups, were hit in the lelf maxillofacial region with a small steel sphere (1.03g, 1400m/s). The specimens from the cochlea nerves were examined under electron microscope immediately (the first group) and at 6 hour (the second group) after trauma, respectively. Another four uninjured dogs were used as a crontrol (the third group). Both experiment groups were found to have mitochondrion swell, crista irregularity and break in axons of the cochlea nerves after trauma. The mechanism of the hearing loss following maxillofacial wound by high velocity missile is discussed.
2.Domelike decompression of the lumbar vertebral canal in the treatment of lumbar spinal stenosis

3.Reliability and validity of the Chinese-version Hypertension Self-care Profile

Objective To test the reliability,validity and Chinese adaptation of Chinese-version Hypertension Self-care Profile (HBP SCP).Methods HBP SCP was translated into a Chinese version and had some Chinese adaptation,the reliability and validity of HBP SCP was tested in 377 patients with hypertension.Results Item-total correlations was 0.396~0.881 and the determination coefficient for each scale was 5.890~20.874.The Cronbachs'α was 0.950 for the total scale,the test-retest correlation was 0.918.The content validity index for the scale was 0.83~1.00,2 and 3 factors were extracted by principal components analysis,which contributed 58.934%,67.224% and 66.601% of the variance.Conclusions The Chinese-version of HBP SCP has good psychometric quality and can be used as a measurement tool for Chinese patients with hypertension.
4.Application of allogeneic amniotic membrane for myringoplasty in the treatment of traumatic perforation of tympanic membrane

OBJECTIVE To investigate the clinical efficacy of using allogeneic amniotic membrane for myringoplasty in the treatment of traumatic perforation of tympanic membrane.METHODS 27 cases of traumatic perforation of tympanic membrane underwent myringoplasty with allogeneic amniotic membrane between July 2003 and February 2008 were analyzed retrospectively.The perforation healing rate and hearing improvement were studied.RESULTS All cases were followed-up for more than 6 months. The perforation healing rate was 96.3%and PTA gain was(12?3.7) dB.CONCLUSION The virtue of myringoplasty with allogeneic amniotic membrane is the plenty of service,convenient operation,little damage and good histocompatibility and effect.

50.0%).ESBLs isolates reached up 63.5%.CONCLUSIONS E.coli has a high detection rate and serious drug resistance in urinary infections.Conventional ESBLs tests can help to appropriate use antibiotics and lower the occurrence of drug resistance.Clinical doctors are advised to restrict the usage of antibiotics in order to lower the occurrence of drug resistance strains,as well as to prevent the spread of ESBLs.
7.Comparison of SPECT and CT in detecting skull base invasion in nasopharyngeal carcinoma

Objective To investigate the detecting ability of single photon emission computed tomography (SPECT) and CT in skull base invasion in nasopharyngeal carcinoma. Methods Sixty three patients with nasopharyngeal carcinoma were examined by whole body and skull base SPECT and CT of nasopharynx and skull base before radiotherapy. The results were double blind compared and evaluated. Results The overall positive rates of skull base invasion detected by SPECT and CT were 63.5% and 25.4%. In patients with headache, cranial nerve palsy and both, they were 87.9%,93.3%,92.3% and 42.4%,46.7%,46.2%. In patients with T 1+T 2 and T 3+T 4 lesions, they were 37.5%,90.3% and 0.0%,51.6%. Objective To evaluated the effect of the regional cooperative rescue model implemented on the length of time from first medical contact (FMC) to balloon dilation (B),economic expense and prognosis in patients with acute coronary syndrome (ACS).Methods Patients with ACS (including ST-segment elevation and non-ST-segment elevation) selected from other hospitals within 24 hours after onset were treated with emergency percutaneous coronary intervention.Patients were divided into two groups, regional cooperative rescue group and control group without the regional cooperative rescue model approved.The lengths of FMC-to-B time and Door-to-B time (from arrival at emergency department or OPD to balloon dilation),time required for patients referred to our hospital,cardiac function,averaged hospital costs,average hospital stay,percentage of medication used and a major adverse cardiac event (MACE) were analyzed.Results Mean FMC-to-B time,Door-to-B time,referral time and time consumed to obtain informed consent were significantly shorter [(106±33) min,(31 ±8) min,(62 ±18,8 ±3) min] vs.[(231 ±35) min,(109 ±26) min,(98 ±31) min,(28 ±11) min,respectively] by implementing the regional cooperative rescue compared with control group,and LVEF was increased,and LVED was deceased inregional cooperative rescue group.The mean costs [(44 123.0 ±3 427.0) yuan vs.(51 587.0 ±5 621.0)] yuan,days of hospital stay [(8.7 ±4.1) vs.(13.2 ±6.4)] and percentage of medication used were significantly decreased in the regional cooperative rescue group.The incidence of MACE inregional cooperative rescue group was 6.2%,whereas the incidence in control group was 16.8%.Conclusions The regional cooperative rescue model can improve the prognosis and decrease the FMC-to-B time,the rate of MACE and financial burden in patients with ACS.
9.Molecular modeling of peptide antibiotics hPAB-?and its artificial mutants construction

Objectives: To design the mutants of peptide antibiotics hPAB ? based on its molecular structure. Methods: The three dimension structure of hPAB ? was constructed by protein homology modeling method. The mutant molecules were designed and generated by PCR and inserted into pQE CP4 expression plasmid. The recombinant plasmids were identified by PCR and DNA sequencing and then transformed into Escherichia coli JM109 to express target fusion proteins. Results:Peptide hPAB ? shows one ? helical and three ? sheet in its structure. Its ? helical regions seem play a key role in the formation of active oligomer. Aside from positioning Thr 7 and Lys 10 into contact positions, the orientation of the ? helix is conserved about the oligome core, forming a ridge around it. Additionally, the dipoles of the helices would overlap to create a positively charged region near the core. These dipoles may be offset, however, by the presence of Asp 4 at the base of the helix. Two mutant molecules, hPAB ? 38 and hPAB ? 34, were designed by deleting N or/and C terminal 2~5 amino acid residues based on hPAB ? structure. The recombinant plasmids containing the mutants gene can express interest fusion proteins in E. coli JM109 successfully. Conclusions: Design, cloning and expression of the mutants of peptide antibiotics hPAB ? lay down the foundation for screening of the mutant of shorter peptide chain and having high or same antimicrobial activity.
10.Application of carrier molecule PaP3.30 in fusion expression of small bioactive peptides

Objective:To screen and clone a carrier molecule for the expression of small bioactive peptides at high levels. Methods: A carrier molecule, PaP3.30, was screened out from the genome of Pseudomonas aeruginosa phage PaP3 and its gene was cloned by PCR method and inserted into pQE 32 expression plasmid, this recombinant plasmid was named pQE PaP30. The peptide antibiotics hPAB ? gene was then inserted into pQE PaP30 and induced to express the fusion protein in Escherichia coli . The ability of PaP3.30 to express other bioactive peptides was evaluated by fusing 6 different origins, varies in sizes and isoelectric points selected peptides to it. Results: After fused to PaP3.30, the peptide antibiotics hPAB ? could express as fusion protein above 30% of total bacterial proteins. Six selected peptides were also expressed by the level of 35%~44% total bacterial proteins when fused to carrier molecule, PaP3.30. Conclusion: The new carrier molecular, PaP3.30, is versatile in the expression of small bioactive peptides.
