1.Construction of the fusion gene lipL32/1-lipL41/2 from Leptospira interrogans and the immuno-reactivity of its expressed products

In the present study, the fusion gene lipL32/1-lipL41/2 from Leptospira interrogans was constructed by using PCR with linking primers, expressed in prokaryotic expression system. And the immuno-reactivity of its expressed product was determined. Meanwhile, SDS-PAGE and BioRad agarose image analyzer system was used to examine the expression output of the target recombinant protein rLipL32/1-LipL41/2, and the immune-reactivity of this recombinant protein was identified by Western blot assay. ELISA was used to detect the level of antibodies against the recombinant proteins in sera of patients with leptospirosis. It was demonstrated that the percentages of similarity in nucleotide and the putative amino acid sequences of the fusion gene lipL32/1-lipL41/2 with the corresponding sequences previously reported were 99. 9% and 98. 9%respectively. The expression output of the target recombinant protein was approximately 10% of the total bacterial proteins.Both the rabbit antisera against rLipL32/1 and rLip41/2 could recognize and combine to the recombinant fusion protein rLip32/1-Lip41/2 as well. The positive rates of antibodies in 228 leptospirosis patients with the recombinant proteins rLip32/1,rLip41/2 and rLip32/1-Lip41.2 were 97.4%, 78. 5% and 99. 1% respectively. The results of the present study leads to the conclusion that the fusion gene lipL32/1-lipL41/3 with its prokaryotic expression system is successively constructed and the expressed recombinant protein shows excellent immuno-reactivity, thus suggesting that it may be used as the antigenic candidate for the development of the leptospiral genus-specific engineering vaccine and for the preparation of diagnostic kits.
2.Study on MDA and SOD of Cervical Cord at Early Stage after Decompression of Chronic Compressive Spinal Cord Injury

Objective We investigate lipid peroxidation of compressed myeloid tissue at early stage after decompression of chronic compressive spinal cord injury.Method SOD and MDA of compressed myeloid tissue are measured respectively before compression,before decompression and 3h after decompression.Result Increased MDA while decreased SOD of compressed myeloid tissue at 3h after decompression than before decompression.Conclusion The increased lipid peroxidation of compressed myeloid tissue at early stage after decompression of chronic compressive spinal cord injury.It is possible that it was resulted from ischemical reperfusion injury.
3.Bedside Chest Radiography in the Intensive Care Unit

Objective To evaluate the applied value of bedside chest radiography in the intensive care unit ( ICU ) . Methods One hundred and ninety-three cases with 507 chest radiographs (CXRs) in the ICU were analysed retrospectively. The appearances of cardiopulmonary abnormalities were analysed.Results 97.4% of the CXRs were satisfied for diagnosis. 83.9% of the CXRs showed cardiopulmonary abnormalities, including atelectasis, pneumonia, pulmonary edema, ARDS, pneumothorax, mediastinal and subcataneous emphysema,pleural effusion,pericardial effusion and pulmonary thromboembolism, and 6.2% out of these positive cases was discovered accidentally. The chest radiograph has moderate accuracy (77.9%) in visualizing cardiopulmonary abnormalities at the initial CXR including atelectasis,pneumonia,pulmonary edema and pleural effusion. Misdiagnosis occurred mainly on the patients with a small quantity of pneumothorax and pulmonary thromboembolism. About one-third cases of cardiogenic edema and ARDS were indistinguishable.Conclusion Bedside chest radiographs are of significant value for the patients in the ICU.
4.The Evaluation of Monitoring Devices in the Intensive Care Unit with Bedside Chest Computed Radiography

Objective To assess the contribution of the portable chest computed radiography (CR) in evaluation of monitoring devices of the patients in the intensive care unit (ICU). Methods One hundred and sixty-two cases with 387 chest radiographs in the ICU were analysed retrospectively. The location of the catheters of monitoring devices and complications were observed.Results The malposition of the catheters was detected in 47 cases(16.9%),including the endotracheal (ET) tubes too deep at the position, the central venous catheters placed into the internal jugular veins,and the position of the thoracic drain tubes to be deep not enough causing the drain to fall.The complications after operation of monitoring devices were not common,including pneumothorax caused by ventilatory assistance,atelectasis and pneumonia caused by malposition of the ET tubes,totally in 11 cases.10 cases with cardiopulmonary abnormalities were discovered accidentally in all 162 cases(6.2%) when evaluation of monitoring devices.Conclusion Bedside chest CR not only can show the catheter position and the complications of the monitoring devices ,but also the cardiopulmonary abnormalities of patients in the ICU.
5.Correlative study among CT signs, prognosis and the expression of CD44v3 and CD44v6 in peripheral non-small-cell lung cancer

Objective To observe the relationship between CT signs, prognosis and the expression of CD44v3 and CD44v6 in peripheral non-small-cell lung cancer (NSCLC). Methods The expression of CD44v3 and CD44v6 was detected with immunohistochemistry (SP method) in 46 patients with peripheral NSCLC confirmed by surgery and pathology. The expression, CT signs and prognosis of NSCLC were analyzed respectively. Results The expression of CD44v3 and CD44v6 correlated with deep lobulation, spinous protuberant sign, short spiculation and lymph node enlargement of hila or mediastinum (P<0.05), but not with the tumor size, vacuole sign, vessel convergence sign and pleural indentation in CT images of peripheral NSCLC (P>0.05). The expression of CD44v3 and CD44v6 correlated with the three-year survival rate in peripheral NSCLC patients (P<0.05). Conclusion Deep lobulation, spinous protuberant sign, short spiculation and lymph node enlargement of hila or mediastinum may indicate strong potential invasion and metastasis in peripheral NSCLC. The positive expression of CD44v3 and CD44v6 may suggest shorter survival time and poorer prognosis in peripheral NSCLC patients.
6.The Posterior Contour Sign of the Lumbar Intervertebral Disc----A Sign to Show the Posterior Border of Disc in Plain Film

Objective:To eveluate and study the anatomic basis of the posterior contour sign of lumbar intervertebral disc.Methods:The most protruding length of the soft tissue mass behind intervertebral spase on lumbar plain film were measured in 100 cases and compared with CT or MR findings.Results:41 posterior contour sign of the interlumbar disc were fined in 39 cases,of which there were 25 in L4/5(61.0%),15 in L3/4(15.6%)and 1 in L2/3(2.4%).The protruding was less than 0.2 cm in 26 and larger than 0.2 cm in 15,there were 10 lumbar disc protruding confirmed by CT or MR ,7 of which the pesterior contour ≥0.2 cm,and 3>0.3 cm,none was ≤0.2 cm.Conclusion:The adipose tissue in the posterior border of lumbar body and disc was the anatomic basis of the sign,and it was an importent sign for diagnosing of lumbar disc hernia on plain film.
7.Simultaneous Determination of 3 Effective Ingredients in Compound Thymol Solution by HPLC

OBJECTIVE:To establish amethod for simultaneous determination ofbenzoic acid,methylis salicylas and thymol in Compound thymol solution.METHODS:HPLC was performed on the column of Thermo Hypersil BDS C18 with the mobile phase ofacetonitrile-water-acetic acid(40∶60∶0.02,V/V/V)at a flow rate of 1 ml/min,the detection wavelength was 275 nm,column tem-perature was 30 ℃,and injection volume was 10 μl. RESULTS:The linear range was 0.045 2-0.9030 mg/ml for benzoic acid, 0.0492-0.9844 mg/ml for methylis salicylas and 0.046 4-0.927 0 mg/ml for thymol;RSDs of precision,stability and reproducibility tests were lower than 2%;recoveries were 98.40%-108.88%(RSD=3.26%,n=9),98.21%-104.07%(RSD=1.87%,n=9) and 96.83-107.09%(RSD=3.33%,n=9),respectively. CONCLUSIONS:The method is simple,accurate and specific,and accurately de-termine the contents of benzoic acid,methylis salicylas and thymol in Compound thymol solution.
8.Mechanism Analysis of the Antidepressant Effect of Acupuncture by Regulating the HPA Axis

Studies have found that depression is closely related to the hyperactivity of the hypothalamo–pituitary–adrenal (HPA) axis, which is manifested in increases in HPA axis-related hormones CRH, ACTH and CORT contents. Traditional Chinese medical acupuncture can down-regulate HPA axis levels to produce a marked antidepressant effect. However, at present there is no HPA axis mechanism analysis of the antidepressant effect of acupuncture. From two aspects: the relationship between depression and HPA axis, and clinical and animal mechanism studies of the antidepressant effect of acupuncture by regulating the HPA axis, an analysis has been made to provide a more powerful scientific basis and the idea of further in-depth study for clinical acupuncture treatment of depression.
9.Finite element analysis of anti-impact loading of wrist protectors

BACKGROUND: Skidproof wrist protection devices can well protect the waist.OBJECTIVE: To analyze the efficacy of wrist protectors against fracture using three-dimensional finite element analysis.METHODS: Based on original data of Mechanical Virtual Human of China, three-dimensional finite element models of wrist with normal soft tissues and wrist wearing protectors were established using Abaqus6.51 software. The model was subjected to loading of 2 m/s. Stress-strain distribution of finite element model of the wrist before and after protectors wearing was compared, and maximum stress variation with time in the distal ulna was analyzed.RESULTS AND CONCLUSION: Compared with wrist not wearing protectors, the equivalent stress at soft tissues of hypothenar eminence, dorsal wrist, inferior segment of the radioulna, and distal radioulna was significantly decreased, which the stress at the second and fourth metacarpal bone, proximal segment of the index finger, and hamate bone was significantly increased, indicating the wrist protector can disperse, or transfer some stress from the distal radius and ulna to the wrist, palm or digital bone, thereby playing a positive role in preventing dital radial and ulnar fractures.
10.Construction,Expression and Identification of Structural Gene for Porin I,the Major Outer Membrane Protein of Neisseria gonorrhoeae

Objective To construct,express,purify and identify the gene encodi ng major outer membrane protein of Neisseria gonorrhoeae (Porin I, or PI). Metho ds The gene encoding for PI of N.gonorrhoeae was amplified by PCR and cloned int o expression plasmid pGEX-4T-2 to form pGEX-4T-2/PI recombinants. A high lev el expression of GST-PI fusion protein was obtained in GST gene fusion system (GST:glutathione S transferase). The analysis indicated that the expressed pr otein was present predominantly in the insoluble form. Therefore, the induced pr otein was purified by SDS-PAGE, and bands corresponding to polypeptides of GST-PI fusion protein were excised and subjected to electroelution. A dot immunoch romatographic assay was employed to demonstrate whether the purified protein was gonococcal PI specific. Results The pGEX-4T-2/PI expression recombinants were constructed,expressed,purified and identified successfully. SDS-PAGE analysis and dot immunochromatographic assay suggested that the recombinant GST-PI fusio n protein was a 60 000 molecular weight protein andidentical in size to native PI and reacted with anti-PI monoclonal antibody. Conclusion Our results may lead to a potentiality for further study of diagnosti c kits and vaccine for Neisseria gonorrhoeae.
