Objective To optimize the RP-HPLC procedure for the determination of aristolochic acid. Method Different extraction solvents and methods have been screened to get the most efficient way for sample preparation. The influence of different mobile systems on quantitation were compared to choose an approprate mobile system for determination. Besides, the minimum limits of different detecting waves were measured to give the method to detect trace amount of aristolochic acid. Result Refluxing with 70% methanol was better than other ways in sample preparation. Both methanol-1% acetic acid (70∶30) solution and 0.3% ammonium carbonate solution (pH=7.5)-acetonitrile (75∶25) with wavelength of 250 nm and 319 nm can be used for quantitation while 0.3% ammonium carbonate solution (pH=7.5)-acetonitrile (75∶25) with wavelength of 224 nm for trace detection. The minimum detectable amount was 0.02 ng. Conclusion The method established can be applied to determinate aristolochic acid and detect trace amout exactly.

BACKGROUND:Studies have reported that the superfamily of transforming growth factors exert a role in the mineralization of various stem cells, but the combination effects of transforming growth factorβ3 and heparin on proliferation and mineralization ability of stem cells from human exfoliated deciduous teeth remains to be studied. OBJECTIVE:To explore the effect of transforming growth factorβ3 on odontoblast-like differentiation of stem cells from human exfoliated deciduous teeth. METHODS:Human deciduous teeth were col ected using enzyme digestion. The 3rd cells were incubated with 25μg/L recombinant human transforming growth factorβ3, 10 U/mL heparin or their combination. The dentin sialophosphoprotein mRNA and dentinsialoprotein expressions were detected by Q-PCR and western blot assay. Alkaline phosphatase activity was determined using alkaline phosphatase kit. RESULTS AND CONCLUSION:Stem cells from human exfoliated deciduous teeth grew wel after induction. The activity of alkaline phosphatase in the combination group was significantly higher than that in the transforming growth factorβ3, heparin and control groups (P<0.01). After combination induction, the cells were strongly positive for alizarin red staining. Results fromα-PCR and western blot assay showed that the expressions of dentin sialophosphoprotein were both remarkably increased at mRNA and protein levels. In summary, stem cells from human exfoliated deciduous teeth can differentiate into odontoblast-like cells under the induction of transforming growth factorβ3 plus heparin.

Objective To investigate the role of transmembrane protein C0-029 in epithelial mesenchymal transition (EMT) induced by tumor necrosis factor alpha (TNFα) of intrahepatic cholangiocarcinoma (ICC) cells. Methods RT-PCR and Western blot were conducted to detect the CO-029 expression in ICC cells and tissues. The effect of CO-029 silencing by lenti-virus on EMT induced by TNFα was investigated.Western blot and mass spectrometry after immunoprecipitation were used to confirm whether TNFαR1 can directly or indirectly bind with CO-029 to form complexes in ICC.Results Differential expression of CO-029 was observed in ICC cells and tissues. The expression of CO-029 was significantly reduced by lenti-viral interference in ICC cells, resulting in the failure of TNFα to induce EMT.TNFαR1 in ICC could directly or indirectly form complexes with CO-029.Conclusion CO-029 mediates TNFα/TNFαR1 induced EMT in ICC,which might play an important role in the invasion and metastasis.
