1.Combined Probe for Determining Canal Filing Cutting Path.

In order to help a surgeon to determine a proper canal filing cutting path in a hip replacement operation conveniently, this paper presents a kind of probe with combined structure. Firstly, the doctor can use this kind of combined probe to choose canal filing cutting path. Then, the doctor can use computer to guide the surgeon to file femoral cavity along the selected canal filing cutting path. Through hip replacement corpse experiments, filing effects and used time of using combined probe group and separate control group were analyzed. The experiment results showed that the methods introduced in this paper could lower the difficulty of hip replacement operations, improve the implantation of hip stem prostheses further, and reduce the incidence of surgical complications.

2. Clinical research on acupoint application on pulmonary function and quality of life in patients with pneumoconiosis

OBJECTIVE: To explore the effects of acupoint application on pulmonary function and quality of life in patients with pneumoconiosis. METHODS: By random number table method,74 pneumoconiosis patients were randomly divided into two groups: acupoint application group and control group,37 cases in each one. The control group was given conventional treatment. Based onconventional treatment,the acupoint application group was given acupoint application with Chinese herbal medicine on the first day of Chu Fu,Zhong Fu,Mo Fu every year for three years. The changes of pulmonary function was observed in each group. The quality of life of patients was investigated by the World Health Organization Quality of Life-BREF. RESULTS: Before treatment,the pulmonary function indicators such as vital capacity,forced vital capacity(FVC),forced expiratory volume in first second(FEV1),FEV1/ FVC% 、peak expiratory flow( PEF),maximal voluntary ventilation and quality of life score between the two groups had no statistical significance( P > 0. 05). Compared with the same group before treatment,all indexes of pulmonary function,except FEV1/ FVC% and PEF,and life quality score were improved in patients in the control group after treatment( P < 0. 05). After treatment,all indexes of pulmonary function and life quality score were improved in patients in acupoint application group( P < 0. 01). The remaining 5 lung function indexes,except PEF,and the quality of life of the patients in acupoint application group were higher than those in the control group( P < 0. 05). CONCLUSION: Acupoint application therapy can improve lung ventilation function and improve quality of life in patients with pneumoconiosis.
3.Expression and Significance of Serum Mir-132 in Patients with Obstructive Sleep Apnea with Cognitive Impairment

【Objective】To explore the relationship between the miR- 132 expression in serum and cognitive deficits of OSA. 【Methods】 66 Chinese adults age 30 to 60 years old were enrolled and categorized into two groups based on Montreal Cognitive Assessment(MoCA)scores: OSA patients with cognitive impairment(OSAI,n=36),OSA patients without cognitive impairment(OSAN,n=30),and thirty adults without OSA as healthy control group(HC,n=30). Out- of- center cardiopulmonary sleep testing (OCST) and MoCA assessment were performed and the relative expression of miR-132 in serum was detected by PCR.【Results】No significant difference was observed in age,education,gender and hypertension(P>0.05). The relative expression level of miR-132 was significantly up-regulated in OSAI patient′s serum compared to the OSAN and HC patients (P<0.001),and had a positive correlation with MoCA score (r=- 0.726,P<0.001). ROC analyses showed that the areas under the curve(AUC)were statistically significant from the line of identity in OSA with cognitive impairment(AUC=0.935,95% CI:0.890- 0.981,P<0.001)and in OSA(AUC=0.787,95% CI:0.695-0.879,P<0.001).【Conclusions】Elevated serum miR-132 expression levels are closely related to the diagnosis of OSA and its cognitive dysfunction. Detection of serum miR- 132 may be a potential indicator of cognitive dysfunction and diagnosis in OSA patients.
4.Analysis of the monitoring of radioactive hazard factors in non-medical institutions in Jinan, China

Objective To investigate the exposure level of radioactive hazard factors and the health management of radiation workers in non-medical radiation institutions (excluding military institutions) in Jinan, China through radioactive hazard factor monitoring, to identify the weak links, and to provide a scientific basis for future work priorities. Methods According to the monitoring plan formulated by Jinan Municipal Health Commission, the task undertaking institutions at all levels in Jinan investigated the types of radioactive hazard factors, detection, training, and health monitoring of 101 non-medical radiation institutions in Jinan. In addition, the workplace radiation levels were detected in 25 institutions of 6 types of monitoring objects, including industrial flaw detection, non-medical accelerator, non-sealed radioactive material workplace, nuclear instrument, baggage detector, and others. Results The investigation objects included institutions engaged in industrial flaw detection, nuclear instrument, luggage detector, non-medical accelerator, non-sealed source workplace, and others. Of these institutions, 91.84% were equipped with radiation protection detectors, 92.86% were equipped with personal dose alarm, 97.73% were equipped with personal protective equipment, 94.36% performed radiation protection training, 92.69% employed radiation workers with certificates, 95.77% performed personal dose detection, 94.83% performed occupational health examination, and 100.00% were qualified for radiation protection detection in workplace. Conclusion There is still a gap between the radiation protection status of non-medical institutions in Jinan and the national regulations and standards, so it is necessary to further strengthen supervision and law enforcement and make greater efforts in training and publicity.
5.Influence of Combination with 1q21 Amplification or-No in Patients with Newly Diagnosed MM on the Clinical Effecacy of Bortezomib-based induction chemotherapy and Long-Term Prognosis of Patients.

OBJECTIVE:
To investigate the influence of influence of combination with 1q21 amplification or-no in patients with newly diagnosed MM on the clinical effecacy of bortezomib-based induction chemotherapy and long-term prognosis of patients.
METHODS:
148 patients with newly diagnosed MM treated from January 2010 to May 2018 were selected and divided into 2 groups: group A (70 patients) without 1q21 amplification and group B (78 patients) with 1q21 amplification; and the survival benefit and influence on clinical efficacy of bortezomib were compared between 2 groups, and the factors influencing clinical prognosis in the patients with newly diagnosed MM were analyzed.
RESULTS:
The median PFS and OS of patients in B group were significantly shorter than those in group A (P<0.05). There was no significant difference in the median OS and PFS between patients with 1q21 amplification copies number =3 and >3 (P>0.05). Multivariate Cox model analysis indicated that the adverse factors for OS were ISS staging, Hb levels, β2 microglobulin levels and 1q21 amplification respectively, and the adverse factors for PFS were Hb levels and 1q21 amplification respectively in patients with newly diagnosed MM (P<0.05). The very good partial remission rate of newly diagnosed MM patients with 1q21 amplification and bortezomib-based induction chemotherapy were significantly higher than that in the patients without bortezomib-based induction chemotherapy (P<0.05). The median PFS time of newly diagnosed MM patients with 1q21 amplification and auto-HSCT after bortezomib-based induction chemotherapy was significantly longer than that of patients without auto-HSCT (P<0.05).
CONCLUSION
1q21 amplification should be the independent risk factor for poor prognosis of patients with newly treated MM. The application of bortezomib-containing induction chemotherapy in patients with 1q21 amplification can efficiently improve the remission rate, while auto-HSCT consolidation therapy may prolong patients' PFS.

6.A novel method to evaluate myocardial injury: cloning of anti-cTnI murine antibody Fab fragment and DNA sequence analysis

BACKGROUND: To apply mouse anti-human cTnI monoclonal antibody as the drug vector in the treatment and diagnosis of myocardial injury, it is important to degrade the immunity of murine antibody and overcome human anti-mouse reaction. Humanization has been applied as an attempt to resolve this problem.OBJECTIVE: To clone murine anti-cTnI Fab fragment and analyse the nucleotide and deduced amino acid sequences.DESIGN: Single sample study.SETTING: An institute of cardiovascular disease under a medical university-affiliated hospitalMATERIALS: The study was conducted in the Institute of Cardiovascular Diseases, First Affiliated Hospital of Nanjing Medical University from January 2003 to May 2004. The hybridoma cell line JS200202 which secrets the anti-cTnI monoclonal antibody was provided by Institute of Cardiovascular Disease, First Affiliated Hospital of Nanjing Medical University.METHODS: IgG heavy chain primers and κ light chain primers of amplified mouse were designed. Total RNA was extracted from hybridoma cells which secrete cTnI. Reverse transcription polymerase chain reaction(RT-PCR) was amplified. Cloning and subsequent sequence analysis of the Fab fragment was performed. The deduced amino acid sequence was compared and analysed with previously published sequences.MAIN OUTCOME MEASURES: Heavy chain Fd segment and κ light chain gene sequence and its subgroups.RESULTS: A band of approximate 700 and 800 base pairs were amplified using IgG heavy chain primers and κ light chain primers respectively. Sequence analysis indicated that the deduced amino acid sequences were in consistent with the characterization of the amino acid in the murine IgGl Fab fragment(GenBank accession NO AY484430, AY484431; Protein Bank accession NO AAR83243, AAR83244).CONCLUSION: A complete murine anti-cTnI Fab fragment was obtained in this study, which may provide basis for the production of the chimeric anti-cTnI antibody.
7.Prophylactic effect of matrix metalloproteinase inhibitor on acute graft-versus-host disease in a murine model.

OBJECTIVETo explore the prophylactic effect of tissue inhibitor of matrix metalloproteinase (TIMP) on acute graft-versus-host disease (aGVHD) after allogeneic hematopoietic stem cell transplantation (allo-HSCT) in a murine model.
METHODSThe murine model of aGVHD after allo-HSCT was established by using female C57BL/b (H-2b) mouse as the donor, and male BALB/c (H-2b) as the recipient. After allo-HSCT, recipient mice were divided into 3 groups. For aGVHD prophylaxis group A was given TIMP (2 mg/d) , group B CsA (5 mg x kg(-1) x d (-1)) was given, and group C nothing. Physical signs, mean survival time (MST), peripheral blood counts and aGVHD histopathology were observed.
RESULTSMice in group C developed typical aGVHD and 100% of mortality, with a MST of 8 days, and those in group A and B had longer survival, the MST being (4.8 +/- 1.4) d and (4.3 +/- 0.9) d respectively, with no statistical difference in peripheral blood count between these two groups. Mice in group A showed less severe signs.
CONCLUSIONSTIMP markedly prolongs MST of allo-HSCT recipients, delays the onset of aGVHD signs, and has no adverse effect on hematopoiesis reconstitution.

8.Research on short-term biological stability and cytotoxicity of allogenic arteries cross-linked by dye-mediated photooxidation in vivo.

This study evaluated the biological stability and cytotoxicity of rabbit thoracic aorta cross-linked by dye-mediated photooxidation in vivo environment of blood flow. Rabbits, whose allogenic thoracic aorta was cross-linked by dye-mediated photooxidation (DMP, n=6) and glutaraldehyde (GA, n=6), were in the DMP group and GA group, respectively; rabbits, whose homogenic abdominal aorta was orthotopic transplanted (OT, n=6), were in the OT group. Then the donor arteries were transplanted into the position of the abdominal arteries of rabbits. After operation, the animals were fed for 1 month, then each of graft arteries was removed for observation. Biological stability was evaluated through histological analysis under optical microscope. Cytotoxicity was evaluated through ratio of coverage of endothelial cell (ES) under scanning-electron microscope. The results showed that no statistically significant difference was noted between DMP group and OT group (P>0.05); however, there was a significant difference between DMP group and GA group (P<0.01). The stability index in DMP group was much higher than that in GA group. There was a statistically significant difference between DMP group and GA group (P<0.01). Ratio was much higher in DMP group than in GA group. In conclusion, the arteries cross-linked by dye-mediated photooxidation treatment appeared to have higher biological stability and lower cytotoxicity in rabbit allogenic transplation model.

9.Relationship between physical activity and trajectories of anxiety and depression scores in college students

Objective:
To investigate the relationship between different physical activity pattern and trajectories of anxiety and depression scores in college students, and to provide evidence for exploring the influence of physical activity on the development of anxiety and depression.
Methods:
A total of 1 265 college students from grade 2017 in a comprehensive university were investigated longitudinally for five times through stratified cluster sampling. Physical Activity Rank Scale-3(PARS-3), Self-rating Anxiety Scale(SAS), and Self-rating Depression Scale(SDS) were used, and latent class linear mixed models(LCLMM) were used to identify the trajectories of anxiety and depression scores and multinomial Logistic regression models were used for exploring the relationship between small persistent status of physical activity among the five surveys and subgroups of trajectory and anxiety and depression scores.
Results:
A total of 940(74.3%) college students were indentified as low persistent physical activity group and 325(25.7%) of them were identified as moderatre-to-high persistent physical activity group. The trajectories of anxiety score were classified into three groups:low level group, high level-decreasing group, and high level group. The number and proportion of the three groups were 686(54.2%), 144(11.4%), 435(34.4%), respectively. The trajectories of depression score were classified into four groups:low level group, moderate level-decreasing group, moderate level-increasing group, and high level group. The number and proportion of the five groups were 604(47.8%), 170(13.4%), 177(14.0%), 314(24.8%), respectively. Compared to the students with moderate-to-high persistent physical activity, higher persistent physical activity was associated with higher risk of keeping anxiety score at a high level(OR=1.78), and higher odds of decreasing depression score at moderate level(OR=0.67), as well as with higher risk of increasing depression score from a moderate level(OR=1.79), and increase the risk of keeping depression score at a high level(OR=1.97)(P<0.05).
Conclusion
Low persistent physical activity of college students could increase the risk of anxiety and depression at a high level. Relevant departments in universities should encourage students to take part in physical activity,with special focus given to individuals with less amount of physical activity and take effective interventional measures to modify the development of anxiety and depression.
10.Study on the sensitivity of leukemic cells to arsenic trioxide enhanced by targeted suppression of mIRNA-21.

OBJECTIVETo study the effect of antisense oligonucleotide targeted on miRNA-21 (AMO-miR-21) for enhancing the arsenic trioxide (As2O3) sensitivity of leukemic K562 cells and its possible acting mechanism.
METHODSChemosynthetic AMO-miR-21 was transfected to K562 cells using Lipofectamine TM 2000. The inhibitory effects of As2O3 and AMO-miR-21, used singly or in combining, on cell proliferation were detected by MTT, their inhibition rate and IC50 were calculated. Cell cycle and apoptosis were assessed with PI stain; expression of miRNA-21 in cells was detected quantitatively by real-time PCR, and the potential target gene PDCD, protein expression was detected by immuno-fluorimetry.
RESULTSUsed in combining with AMO-miR-21, the IC50 of As2O3, could be lowered from 2.1 micromol/L to 1.23 micromol/L, and the sensitivity of cells to As2O3 increased to 1.78-fold; with the amount of apoptotic cells increased significantly. Transfection with AMO-miR-21 alone could downregulate the expression of miRNA-21 in cells (P < 0.01), and up-regulate PDCD, protein expression level significantly.
CONCLUSIONSCombined use of AMO-miR-21 and As2O3 could increase the sensitivity of K562 cells to As2O3, which provides a novel potential approach for treatment of leukemia. AMO-miR-21 realizes it anti-tumor action by way of targeted inhibition on miRNA-21, and further up-regulates the expression of anti-tumor gene PDCD4.
