1.In vitro antitumor efficacy of lymphokine-activated killer cells

We have investigated, in both cases of normal individuals and tumor- bearing patients, the cytotoxic activity of lymphokine- activated killer (LAK) cells to cultured cell lines or fresh solid tumor cells with a 4- hour chromium 51 (~(51)Cr) - release assay. The LAK cells were generated by incubating peripheral blood mononuclear (PBM) cells in vitro with recombinant interleukin 2 (rIL- 2)or partially purified interleukin2(PPIL- 2). Our results indicated that these two sources of L \K cells were cytotoxic to either K562, NK- sensitive or Daudi, NK resistant tumor cell lines. These two kinds of LAK cells could also kill a broad rang of fresh solid tumor cells, suggesting that these cells be indeed LAK cells. Furthermore, the results demonstreated that some fresh solid tumor cells were resistant to LAK cell killing to some extent as they showed different sensitivities to LAK cytotoxicity.
2.Progress of renal voltage-gated chloride channel

Voltage-gated chloride channel (ClC) is a class of very important chloride ion channels in vivo,participating in the chloride ion transporter,cell volume regulation,cell control acidification and many pathological physiological process.The studies have found that ClC-K was related to dent disease,and ClC-5 was associated with bartter syndrome.This article mainly reviews ClC-type chloride ion channel classification,structure,regulation and distribution in the kidney,and summarizes the related diseases.
3.Heart Rate Variability Analysis based on Multi-Resolution Analysis

Objective:Heart rate variability (HRV) contains a lot of information concerning the cardiovascular regulation,and can be an important indicator,quantitatively reflecting of autonomic nervous function and the normal heart activity,also can play an important role in cardiovascular regulation.Therefore,HRV signals are analyzed using common HRV analysis methods.Methods:Decompose HRV signals of congestive heart failure and normal persons into 6 scales using Multi-resolution analysis.The wavelet coefficient energy is calculated,especially the frequency energy of cd3 to cd6.Results:Energy is mainly concerned in the lower band of high frequency band and low frequency band,where the energy differences between the two groups are more evidence.It's more precise compared to the result with frequency-domain analysis.Conclusions:To gain results with universal law,more different analysis methods need to be improved and lots of researches are necessary.
4.In vitro antitumor mechanisms of human Lymphokine-Activated killer cells

We previously demonstrated the cytotoxicity of human LAK cells to solid tumors in vitro. The work we reported here investigated the mechanisms involved in killing of solid tumor cells by LAK cells. It was shown that the cytotoxicity of LAK cells was mediated by some factors secreted by LAK cells and effects of direct contact with targets. Following lysis, the nucli of targets were destroyed and fragments of DNA were released into the medium. The cytotoxicity of LAK cells, under some circumtances, was of a positive correlation to the proliferation of themselves.
5.Preliminary Study of Immunodiagnosis of Trichinosis by ELISA Using Urea Soluble Antigen of Trichinella spiralis

Urea soluble antigens of Trichinella spiralis infective larva were prepared and the efficiency of the antigens in diagnosis of trichinosis by ELISA are studied. Specific IgG antibodies are detected by ELISA using urea soluble antigens in 91.67%(22/24) of 24 sera and 91.67% (ll/12)of 12 blood spots of filter paper of patients of trichinosis. Specific IgM antibodies are seen in 95.24% (20/21) of 21 sera of patients using the antigens. Sera of patients infected with Schistosoma japonicum have some cross reactions on the antigens. Sera from mice infected with - Schistosoma japonicum, Ancyclostoma caninum larva, and Plasmodium yoelii are negative. The urea soluble antigen has a relatively high sensitivity, specificity and good reproducibility in diagnosis of trichinoisis by ELISA, and has a practical and economic value.
6.Effects of pioglitazone combined with radiotherapy on colon carcinoma xenograft in mice

Objective To investigate the effects of peroxisome proliferator activated receptor γ(PPARγ) agonists pioglitazone combined with radiotherapy on colon carcinoma xenograft in mice.Methods The colon cancer cells CT26 of mouse were inoculated in the right side of the rib loins of BALB/c mouse.Established BALB/c mice tumor models.The mice were randomly divided into six groups:untreated group,tumor contral group,DMSO group,pioglitazone treated group,radiotherapy group,radiotherapy combined with pioglitazone treated group.To observe the tumor growth of each group,to compute and compare to the tumor inhibition rates.the pathological varieties of the tumor were observed.Results The tumor volumes were compared with each group after two weeks of radiotherapy.Compared with tumor contral group,pioglitazone treated group,radiotherapy group and radiotherapy combined with pioglitazone treated group could decrease the volume of tumor (P =0.008,P =0.001,P =0.001).Compared with pioglitazone treated group,the effect of radiotherapy group or radiotherapy combined with pioglitazone treated group was more evident (P =0.026,P =0.018).Contrasted with radiotherapy group,radiotherapy combined with pioglitazone treated group had no significant difference (P =0.335).The tumor inhibition rates of pioglitazone treated group,radiotherapy group and radiotherapy combined with pioglitazone treated group were 46.30 %,68.60 % and 70.01%.The pathological varieties of tumor were obvious.Conclusion Pioglitazone,radiotherapy and radiotherapy combined with pioglitazone could inhibit the growth of tumor.PPARγ is a new target for tumor treatment.
7.Effect of Aerobic Exercise and Dietary Fat on the Formation of Atherosclerotic Plaque in ApoE-Deficient Mice

To approach the effects of aerobic exercise anddietary fat on the formation of atherosclerotic plaque, we have adopted the ApoE-gene knockout mice as the model to study the effects on an animal treadmill. The results show that the plaque area at the aortic sinuses of the exercised and low fat diet fed mice group were significantly reduced as comparing with the control group respectively. Mice of aerobic exercise and low fat diet combined treated group have the least lesion area of plaque as compared with the other groups. These results have indicated that either aerobic exercise or low fat diet is beneficial and effective to alleviate the formation of atherosclerotic lesions. However, aerobic combined with exercise can provide the greatest benefit in the alleviation of the formation of atherosclerotic plaque as compared with the single factor of either exercise or low fat diet.

Flow fluorescence microbeads array is a novel high throughput technique based on the combina tion of use of flow cytometry and use of fluorescence encoded microbeads as the solid phase for conventional im munoassay,affinity assay and DNA hybridization assay,.which offers a new approach to multiplexed assays for ap plications of large-scale screening.By utilizing appropriate fluorescence encoded micron-sized polymer particles, flow fluorescence microbeads array can be created to enable highly muhiplexed analysis of complex samples.Indi vidual sets of microbeads can be modified with reactive components such as antigens,antibodies or oligonu eleotides,then mixed with each other to form a multiplexed assay set.Flow fluorescence microbeads array com bines the specificity of fluorescence-encoded microbeads and the high sensitivity of flow cytometry,bears signifi cant potential as a general analysis platform for both research and clinical applications.Recent applications and advances are reviewed in this article.
9.The correlational study between the depression after acute stroke and nerve function defect

Objective To explore the correlation between the depression status of patients with post-stroke depression(PSD) and nerve function defect.Methods 75 patients in the first 24h of acute stroke onset were selected.According to the Zung depression self rating scale(SDS),75 cases were divided into simple stroke patients group and PSD group,and the Hamilton depression scale (HAMD) and Scandinavian nerve function defect rating scale (SSS) were used for further evaluation.The score results were statistically analyzed.Results Of 75 cases with acute stroke,32 cases(42.7%) were diagnosed PSD.Admission day 2,at the end of 1,2 and 3 weeks,the HAMD scores between PSD patients and simple acute stroke group had statistically significant differences (all P < 0.01).The HAMD scores of three groups of PSD patients (light,medium,heavy) appeared obvious changes with the prolonged time,which were significantly higher than the simple stroke group.The SSS score of simple stroke group and mild PSD patients increased with prolonged observation time,the difference between the two groups was significant (q =9.75,P < 0.05).The SSS score of moderate and severe PSD patients decreased with prolonged observation time,compared with the simple stroke group,the differences were statistically significant(q =50.23,46.82,all P < 0.05).In simple stroke patients and mild PSD patients,HAMD scores and SSS score had no correlation (r =0.062,P > 0.05).In moderate and severe PSD patients,HAMD score and SSS score had correlation (r =4.682,P < 0.05).Conclusion The incidence rate of PSD in acute brain stroke patients was 42.7%,and in patients with moderate and severe PSD,HAMD scores correlated with the degree of nerve function defect.Early intervention on depression treatment and promote the recovery of neural function is also important.
10.Change of thrombin antithrombin Ⅲ complex after acute brain infarction

ObjectiveTo determine the change of coagulation in acute brain infarction.MethodsBlood samples were taken intravenously from 57 patients with acute brain infarctions (within 24hr) and the thrombin time(TT), prothrombin time(PT), activated partial thromboplastin time(APTT), fibrinogen(FIB) levels were detected serially at the time of instant admission, 1st day ,3rd day and 7th day after routine therapy, including 21 patients whose thrombin antithrombin III complex(TAT) levels were detected at the same time.ResultsTAT levels were significant increased in acute brain infarctions, especially in acute progress stroke, but TT, PT, APTT were not significantly different before and after routine treatment. FIB levels were higher at 7th day than pre-treatment. ConclusionTAT levels can be served as a marker of progress stroke.
