Lyophilized storage of red blood cells(RBC) may provide a useful storage alternative Both shelf life and transportation logistics can be improved over those for existing refrigerated or frozen storage. The present study was designed to evaluate the effects of lyophilization process on mean RBC recovery. Blood was drawn from healthy adult having no history of either hemoglobinophathy or abnormal RBC metabolism. Blood stored in plastic transfer bags were lyophilized for 16 hours, then it was rehydrated . In group 1, samples were stored at 4℃ for 6 hours, while in group 2 were stored at 4℃ for 5 hours, then 25℃ for 1 hour. The mean RBCs recovery in group 2 was higher than group 1(81 4?7 6% vs 62?10 2%,respectively). Samples were lyophilized using buffer containing different concentrations of glucose, and studies showed that there was no significant difference in mean RBCs recovery between samples in lyophilization buffer containing high and medium concentration of glucose, but low concentration of glucose in buffer resulted in lower mean RBCs recovery. The mean RBCs recovery was 30% in low concentration of glucose buffer and 45 1%, 47 2% in medium and high concentrations of glucose buffer, respectively. We also observed effects of different rehydration buffer with different osmotic pressure, and it showed that high osmotic pressure buffer resulted in higher mean RBCs recovery than iso-osmotic buffer. There was little change in the cell size and cell volume. The results presented in this report clearly demonstrate that human RBCs can be successfully lyophilized. Lyophilization buffer and rehydration buffer may influence the mean RBCs recovery.

Objective To explore the effects of rosiglitazone on the expression of TNF-? in visceral adipose tissue of insulin-resistant rats induced by high-glucose diet. Methods Sprague-Dawley (SD) rats were fed with high-glucose diet for 6 months to reproduce the non-obesity insulin-resistant rat model (abbreviated as IR rats). The model rats were then randomized into insulin resistan (IR) group and rosiglitazone (ROS) group (10 each). Ten rats fed with normal diet were enrolled as controls (CN group). Rats in ROS group were administered with 10?mol/(kg?d) of rosiglitazone for 6 weeks from the 7th week of experiment,and those in CN and IR groups were administered with same amount of distilled water. At the 6th and 12th week,the systolic pressure,body weight,and biochemical parameters (fasting and 2-hour post-glucose load plasma glucose,insulin,free fatty acid,triglyceride,high density lipoprotein cholesterol) were measured,and insulin resistance index (HOMA-IR) was calculated. At the end of the 12th week,the visceral adipose tissue was collected to calculate the ratio of visceral fat to body weight (VF/BW),and to determine the protein and mRNA expressions of tumor necrosis factor-? (TNF-?) with inmmunohistochemical assay and RT-PCR. Results As compared with CN group,the systolic pressure,HOMA-IR,plasma insulin,triglyceride and free fatty acid increased,whereas high density lipoprotein cholesterol decreased (P0.05). Inmmunohistochemical examination showed positive expression of TNF-? in intercellular substance,but not in visceral adipocytes. The expressions of TNF-? protein and mRNA in visceral adipose tissue were dramatically higher in IR group than in CN and ROS groups (P0.05). Conclusion Rosiglitazone may significantly abate insulin resistance,reduce the abnormality of plasma lipid and blood pressure,and inhibit the over expression of TNF-? in visceral adipose tissue in the non-obesity insulin-resistant rats induced by high-glucose diet.

Objective To study the risk factors for ventilator associated pneumonia (VAP) in ICU and pathogens antimicrobial resistance.Methods 220 patients were selected and divided into observation group (88 patients) and control group(132 patients) depending on whetherhe has VAP or not.The clinical data were reviewed to explore the risk factors.And the secretions of respiratory tract were investigated by the routine bacterial culture and drag-resistance methods to analysis the distribution of pathogens.Results The risk factors associated with VAP were COPD,mechanical ventilation,the extensive use of antibiotics,age ＞ 60 years,APACHEII score ＞ 30(P ＜ 0.01).157 bacterial were cultured,68.9％ of pathogens was Gram-negative bacteria in which Pseudomonas aeruginosa was rated as the top one,and 26.1％ of pathogens was Gram-positive bacteria in which MRSA was the majority.The results of the antibiotic resistance monitoring indicated that all Gram-positive and Gram-negative bacteria showed high drugresistance to common antibiotics.Conclusion We should control these risk factors which are closely related to VAP,and select suitable antibiotics on the basis of etiological analysis and drug-resistance.

Objective To establish hypoxic-ischemic brain damaged (HIBD) rat model,investigate whether H2S and cystathionine-β-synthase (CBS), the key enzyme for its generation, may be a mediator of electro-acupuncture(EA) stimulation treatment for HIBD. Methods Thirty-two healthy Sprague-Dawley neonatal rats were divided into four groups randomly: sham control group ( n = 8 ); sham + EA group ( n =8); HIBD control group ( n = 8); and HIBD + EA group ( n = 8 ). HIBD rat models were established on their 7-day-old. From the next day ,rats of sham + EA group and HIBD + EA group were electric stimulated 30 min daily for 14 d,BAIHUI and DAZHUI as the acupoints. Control ones were just fixed at the same time,without acupuncture. The rats were sacrificed on the 22 nd day, one day after the treatment course. Cortical H2S concentration was measured by sensitive sulphur electrode assay. The CBS protein expression was measured by western blot analysis and immunohistochemistry for localization. Results The concentration of cortical H2S in HIBD control group was (26. 83 ± 4. 31 ) nmol/mg protein, which was significantly higher than that of sham control group[(22. 78 ± 1.54) nmol/mg protein]( P ＜ 0. 01 ). The H2 S levels in HIBD + EA group and sham + EA group were ( 18.08 ± 2.71 ) nmol/mg protein and ( 18.91 ± 2. 78 ) nmol/mg protein, respectively. Compared with corresponding control group, they were much lower( P ＜ 0. 01 ). The expression of CBS protein in rats with EA stimulation decreased in cortex compared to corresponding control group( P ＜0. 05 ).Conclusion EA can down-resulate H2S/CBS pathway. This may be one of the mechanisms of how EA contributes to the recovery of brain damage.

Objective To establish the quality standard of Alatan Wuwei Pill. Methods Chebulae Fructus and Granatii Fructus were identified by TLC. Gallic acid and ellagic acid in Gardeniae Fructus and Granatii Fructus were determined by HPLC. Results The TLC spots developed were clear. Gallic acid showed good linear relationship in the range of 0.058 72-1.056 96 mg (r=1.000 0), the average recovery was 99.15%(RSD=1.3%). Ellagic acid showed good linear relationship in the range of 0.079 64-1.194 6 mg (r=1.000 0), the average recovery was 100.02% (RSD=2.3%). Conclusion The method is simple and reproducible. It can be used to control the quality of Alatan Wuwei Pill.

Objective Summarize the interventional nursing experiences for patients with coronary chronic total occlucion by transraidal approach. Methods Preoperative evaluation, careful psychological nursing, perioperative observation and prevetion complications were used for 105 patients with coronary chronic total occlusion when curred by transraidal approach intervention. Results There were 97 cases had successful operation, while 8 cases failure. There were 14 cases of perioperative hypotension, 6 cases of coronary acute occlusion, 4 cases of coronary perforation and 3 cases of no- reflow. There were no radial- as-sociated complication in any patients. Conclusions Effective and careful nursing measures can prevent complications.

Objective To study the positions,features and sectional anatomy of the palatovaginal canal and vomerovaginal canal,comparing with CT and MRI findings. Methods The palatovaginal canals in 104 sides and vomerovaginal canals in 60 sides of dry skulls were observed.Three adult cephalic specimens were sliced into celloidin contiguous sections on coronal plane(0.5 mm in thickness) and the results were correlated with the CT studies of 11 patients and MR imaging of a cephalic specimen. Results In dry skulls,the palatovaginal canal displayed as complete canal in 79.8% of the cases,while appeared as groof in 15.4%.There was neither significant canal nor groof in 4.8%.The vomerovaginal canal demonstrated as complete canal in 36.7% of the speciments,while appeared as groof in 33.3%.This canal was communicated with the lateral palatovaginal canal in 15% and it was not detected in 15%.Four typical slices of the celloidin and 3 plane of CT and 3 plane of MRI were selected and the key structures were described.Conclusion The palatovaginal canal and vomerovaginal canal can be well depicted on coronal plane.

Objective To investigate the effect of pantoprazole (proton pump inhibitor, PPI) and gefarnate (gastric mucosa protectant) on the prevention of upper gastrointestinal bleeding in patients undergoing post-percutaneous coronary intervention (PCI) dual anti-platelet therapy.Methods This research included 1263 patients taking enteric aspirin and clopidogrel after PCI.The cases were divided into 4 groups: routine treatment group (n=332), PPI group (n=318), gastric mucosa protectant group (n=299), and PPI+gastric mucosa protectant group (n=314).A follow-up for 6 months was observed including gastrointestinal symptoms, upper gastrointestinal bleeding, major adverse cardiac events (MACE), and adverse reactions.Results There were 52 cases of upper gastrointestinal bleeding within 6 months, including 21 cases from routine treatment group, 9 from PPI group, 15 from gastric mucosa protectant group, and 7 from PPI+gastric mucosa protectant group.The incidence of upper gastrointestinal bleeding among the 4 groups within 6 months was statistically different (X2=8.883, P=0.031).The routine treatment group had significant higher rate than the PPI group and the gastric mucosa protectant group (P<0.05), while among other groups there was no significant difference (P>0.05).The upper gastrointestinal bleeding occurred within 3 postoperative months in 34 out of 52 cases (65.4%).There was no statistical significance among the four groups in regard to bleeding occurrence time (X2=4.212,P=0.648).Conclusions Patients undergoing post-PCI dual anti-platelet treatment can reduce the incidence of gastrointestinal bleeding by taking pantoprazole or combined with gefarnate.Intervention against upper gastrointestinal bleeding should start on the first day after PCI and last for a minimum of 3-6 months.

Objective: To explore pathogeny and clinical effect of physiotherapy in young patients with lumbar disc hemiation (LDH). Method: Eighteen young LDH patients at 20-30 years old were administered comprehensive physiotherapy including lumbar traction, pulsed short wave therapy, pulsed magnetotherapy or low frequency modulated medium frequency electrotherapy. Before and after treatment, lumbar function was assessed with JOA scale. Result: Nine patients had specific trauma history; nine patients had sedentary lifestyle; five patients worked lor more than 8h/d in standing position; four patients had body mass index (BMI) 24-28. There was no significant difference in age, BMI and JOA scores between male and female group (P＞0.05). Patients scored significantly higher after treatment (P＜0.05). The remarkable effective rate of physiotherapy was 72.22%. Conclusion: Trauma, sedentary lifestyle, standing for long time, overweight or obesity are primary risk factors of LDH for youth and should be prevented by health education. Physiotherapy is significant efficient for young LDH patients.
