Objective: To establish a Beagle dog model for the study of the mechanism of implant-bone interface remodeling following the restoration of submerged and nonsubmerged implant denture. Methods: 8 adult purebred beagle dogs were used to build the animal model. Fixed metal full crown was used to carry out submerged and nonsubmerged implant denture in the dogs to mimic the normal chewing status in animal models. Results: The submerged and nonsubmerged implant dentures were made successfully. After the implant denture restoration, all experimental dogs were fed with granular horniness forage to guarantee enough biting stimulation. The chewing fashion of experimental dogs was not changed obviously during the experiment process. Although the horniness forage may result in fairly bigger bite force, no visble biting hurt was found 12 weeks after loading and the fixed implant dentures were all preserved. Conclusion: The beagle dog model can preferably mimic the normal chewing fashion and the models are both availability and credibility.

The b eagle dogs were sacrificed by steps after loading of the dentures. HE staining t echnique was used to observe the dynamic remodeling process of implant-bone int erface.Results:2 weeks after loading of implant denture, a major ity of the implant surface attached to bone tissue directly, however, at the int erface, especially at the top of the screw thread, bone tissues were absorbed an d substituted by fibrous tissues. 4 weeks after loading, fine attachment was fou nd at the implant-bone interface and the previouly observed fibrous tissue at t he interface was gradually remodeled to form new bone. 8 weeks after loading, i mplant directly attached to bone tissues by osteo-interface, and the cellular c omponents and capillaries were decreased at the interface.12 weeks after loading , all implants attached to bone tissues by osteo-interface with high combinatio n level, typical Havers system was observed at the interface.No obvious differen ce in the interface remodeling was observed between the submerged and nonsubmerg ed implant denture. Conclusion:There is no distinct difference i n the implant-bone interface remodeling after the loading of submerged and nons ubmerged implant denture.

Glycerol,mannitol has been used as additives for the separation of deoxyribonucleic acid (DNA)fragments in entangled solution by capillary electrophoresis. Differing from glycerol and mannitol,the structure of glucose is a ring with 5 hydroxyl groups.This structure helps it easily to form complex with boric acid and hydroxypropyl methylcellulose (HPMC).On the other hand,glucose has large space obstacle compared with glycerol and mannitol.So glucose can markedly improve separation of DNA in HPMC solution by capillary electrophoresis compared with other additives.The factors such as boric acid concentration and pH,influencing formation of complex of HPMC and glucose were studied. High concentration of boric acid could improve the DNA separation in glucose-HPMC matrix system. The optimization pH to separate DNA in HPMC-glucose solution by capillary electrophoresis was 8.3.

砄bjectives:To observe the difference of programmed cell death (PCD) and the expression of related gene p 53 in the development of normal palate and in the formation of cleft palate.Methods:The model of the development of cleft palat was estabished with retinoid acid (80 mg?kg 1 for each pregnant mouse). The palate samples were obtained at GD13 14 (at 13rd day 14 hours of prenancy),13 22 ,14 8,14 14 ,14 33 ,15 8,15 22 and 16 8 respectively.PCD was detected with TUNEL staining,while the mRNA transcription of p 53 was observed by in situ hybridization.Rssults:In early development of palate process,the positive index of PCD in the cleft palate samples was significantly higher than that in the normal ( P 0.05) between the two groups.Conclusion:The proliferation of mesenchymal cells of the early developmental palate process may be inhibited due to the abnomal PCD in the formation of cleft palate.The mechanism of PCD in this study may be a p 53 independed pathway.

Objective To investigate the effect of hyperoxia on the expressions of cysteinyl aspartate specific proteinase -3(Caspase -3)and proliferating cell nuclear antigen (PCNA)in bone marrow mesenchymal stem cells (BMSCs).Methods Primary BMSCs from SD rats were cultured in vitro,BMSCs grew to 70% -80% degrees of fu-sion and then were randomly divided into room air group and hyperoxia exposure group.Each group was divided into 5 sub -groups,and cultured for 0,3,6,1 2,and 24 h respectively.Morphology investigation with inverted phase contrast microscope was adopted.The expressions of Caspase -3 and PCNA protein levels were detected by Western blot. Results (1 )As time wenty by,compared with room air group,the gap between cells increased,some of the cells be-came circular,and cell detachment and floating appeared in the hyperoxia -exposure group (>1 2 h)compared with room air group.(2)As time went on,compared with the room air group,the expression levels of Caspase -3 protein in the hyperoxia -exposure group were increased,and the difference was significant after 6 hours of culture (0.27 ± 0.04 vs 0.1 4 ±0.02,t =5.03,P =0.007).(3)Compared with room air group,the PCNA levels of the hyperoxiaexpo-sure group(6 h)decreased,and the difference in PCNA protein expression levels was significant after 6 hours of culture (0.27 ±0.04 vs 0.38 ±0.04,t =3.37,P =0.028).Conclusions Hyperoxia exposure increases Caspase -3 expres-sion levels and decreases PCNA expression,which may affect the proliferation and apoptosis of BMSCs.

In recent years,along with the deepening study of bone marrow mesenchymal stem cells (BMSCs),the repair effect of BMSCs in various tissue injury have been gradually revealed.And in recent years,except the ability of differentiate to the target cells,its paracrine effect,for example,a variety of cyto-kines secreted by BMSCs,also plays an important role in the process of repairing.

Background and purpose:Promoter methylation ofPCDH10, a gene encoding protocadherin 10, has been found to be correlated to poor prognosis in gastric cancer (GC) patients. However, the relationship between the expression of PCDH10 and prognosis in GC remained unknown. This study aimed to explore the relationship be-tween the expression of PCDH10 and clinicopathological features and prognosis of GC, and to identify biomarker for predictions of recurrence and survival of GC.Methods:mRNA expressions of PCDH10 in 115 pairs of GC tissues and adjacent normal tissues were detected by real-time lfuorescence quantitative polymerase chain reaction (RTFQ-PCR). The correlation between PCDH10 expression level and clinicopathological features and prognosis of GC was analyzed. Prediction models for 5-year recurrence and 5-year survival were established using logistic regression method.Results:Progression-free survival (PFS) and overall survival (OS) were signiifcantly prolonged in patients with PCDH10 low expression compared to patients without PCDH10 low expression (P=0.046 andP=0.033 respectively). PCDH10 low expression signiifcantly correlated with less lymph node metastasis (P=0.001) and earlier TNM staging (P=0.001), and was more common in female than in male (P=0.040). The mRNA expression of PCDH10 did not correlate with age, Lauren classiifcation, T stage, neural invasion or vascular invasion. Univariate Cox analysis showed Lauren classiifca-tion, T stage, N stage, M stage and PCDH10 expression signiifcantly correlated with PFS and OS. Logistic regression models for the prediction of 5-year recurrence or 5-year survival based on clinicopathological features included Lauren classiifcation, T stage, N stage and M stage as variables. Logistic regression models for the prediction of 5-year recur-rence or 5-year survival based on PCDH10 expression included Lauren classiifcation, T stage, M stage and PCDH10 expression level but not N stage as variables. The models based on PCDH10 expression had the same effciencies as models based on clinical parameters in predicting 5-year recurrence or 5-year survival for GC patients.Conclusion:PCDH10 low expression correlated with better prognosis, less lymph node metastasis and earlier TNM stage in GC patients. Low expression of PCDH10 may be a biomarker of better survival for GC patients. Logistic regression model based on PCDH10 mRNA expression may serve as a prediction model when patients have unknown lymph node metas-tasis status.

Objective Lactate dehydrogenase (LDH) is shown to be an important virulence factor resulting in acid production of Streptococcus mutans(S.mutans), on which the cariogenic potential of S.mutans depends.Differences in cariogenic abilities of S.mutans isolates may be determined by genetic heterogeneity from virulent factors.The relationship between enclosed Uyghur children LDH activity or genetic diversity and cariogenicity of S.mutans(serotype c)isolates was studied in this research.MethodsChoose 6 original inhabitants of the countryside from the age of 3 to 5 years old closed Uighur children in kashika.These isolates came from 35 caries-active individuals and 37 caries-free enclosed Uyghur ones, in which 27 strains showed the high LDH activity and 24 strains showed the low LDH activity.Then genetic diversity of PCR products were analyzed or assessed by restriction fragment-length polymorphism(RFLP).Some of amplified LDH genes from different group were sequenced and assayed.ResultsIt is testified that two genotypes A and B of ldh-RFLP were revealed when LDH genes were digested with MseI,but MnlI,DdeI,NlaⅡI and AluI digesting fragments of ldh gene did not show different pattern.Furthermore,Fisher Exact one-Tail Test showed that the proportion of Genotype B strains from caries-active individuals was higher than that from caries-free ones(P<0.05),while the distribution genotypes A,B with different LDH activity was different between two groups(P<0.05),high proportion of the LDH active group genotype B strains than the low activity group.ConclusionThis study indicated that LDH gene of S.mutans strains from Enclosed Uyghur children different individuals is conservative, while there still is the gene mutation in.The ldh genetic diversity may be related to the high caries sensitivity, and closely correlated with the differences in LDH enzyme activity of S.mutansstrains.

OBJECTIVE:To establish a method for determining the content of Ornidazole gel by first-order derivative spectrophotometry.METHODS:The first-order derivative spectrophotometry was used to determine the content of Ornidazole gel at a wavelength of 259nm and 299nm.RESULTS:The linear range of Ornidazole was 4.01～44.1?g?mL-1.The average recovery was 101.6%,with RSD of 1.3%.CONCLUSIONS:This method can meet the requirements for quality control of Ornidazole gel both in terms of precision and accuracy.
