1.Study on Optimum Conditions of Isolation and Purification of Specific Toxin Fractions Produced by Exserohilum turcicum

Five fractions,Ⅰ(R_f0.06)、Ⅱ(R_f0.21)、Ⅲ(R_f0.45)、Ⅳ(R_f0.60)、Ⅴ(R_f0.75)have been obtained after HT-toxin from race 1 of Exserohilum turcicum 99-2 isolated by TLC. In all of these fractions,only fractions Ⅱ had specific toxicity to the corn leaves with Ht1 gene. Then fractions Ⅱ-1、Ⅱ-2、Ⅱ-3 were isolated from fractions Ⅱ by HPLC purification,and the bioassay result showed only fraction Ⅱ-3 was toxigentic to corn leaves with Ht1 gene but non-toxigentic to corn leaves without Ht1 gene. Fractions Ⅱ-1、Ⅱ-2 and Ⅱ-3 were scanned by UV-Vis spectrophotometer. It was shown that the fractions Ⅱ-1 and Ⅱ-2 had analogous spectrum,and especially the fraction Ⅱ-3 had a special peak at 300nm.
2. Determination of five saponins in Ginseng Radix et Rhizoma-Bupleuri Radix herb pair and its preparations by QAMS method

Objective To study the effects of ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1, saikosaponin a, and saikosaponin d in Ginseng Radix et Rhizoma-Bupleuri Radix (GRRBR) herb pair and its preparations by using quantitative analysis of multi-components by single-marker (QAMS). Methods On the basis of ginsenoside Rg1, the relative correction factors between the ginsenoside Rg1 and the other four saponins were established, and then the contents of the other four saponins were calculated. At the same time, the contents of the five components were determined by external standard method and compared with those evaluated by QAMS. The relative retention time was determined by different chromatographic columns. It could be considered that QAMS was feasible and accurate in the determination of saponins in GRRBR herb pair. Results A quantitative control method of five kinds of saponins was established, and the methodological results were good. The results showed that there was no significant difference in the contents of five kinds of saponins in GRRBR herb pair, Xiaochaihu Decoction, and Kaixin Jieyu Prescription between QAMS group and external standard method group. Conclusion QAMS is suitable for the determination of saponin in GRRBR herb pair, which can be used as a reference for the determination of its effective components and the establishment of compound quality control method.

4.Determination of Anticancer Activity Components of Flavonoids from Hedyotis diffusa by HPLC

OBJECTIVE:To establish the method for the simultaneous determination of anticancer activity components of flavonoids from Hedyotis diffusa,i.e. quercetin,kaempferol. METHODS:HPLC was applied to determine the contents and performed on Alltima C18(250 mm?4.6 mm,5 ?m) column. Mobile phase consisted of methanol(A)-0.5% glacial acetic acid,(gradient elution). The detection wavelength was aet at 350 nm. RESULTS:The linear range of quercetin was 0.006 2~0.244 0 ?g(r=0.999 8)and that of kaempferol 0.007 8~0.310 6 ?g(r=0.999 9). The average recovery of quercetin was 101.84%(RSD=1.79%,n=6) and that of kaempferol 99.04%(RSD=2.90%,n=6). CONCLUSIONS:The method is simple,accurate and reproducible for the quality control of H. diffusa.
5.Studies on Stability of Microbial Flocculant and Its Application to Municipal Thickened Sludge Dewatering

The stability of microbial flocculant (MBF) produced by Aspergillus sojae and its application to municipal thickened sludge dewatering were studied. The results showed that the MBF had high heat and acid-base endurance with high flocculating activity in a wide range of pH from 1.5 to 12. The MBF retained 96% of flocculating activity after 35 days preservation at 4℃, but in different pH the flocculating activity difference was very apparent after 35 days store at room temperature. The experimental results also demon- strated that the MBF was better than PAM and PAC in reducing specific resistance filtration. The optimal dose of MBF used for intensifying thickened sludge dewatering is 7%(volume fraction). And the more the volume of sludge is treated, the less the cost of MBF for unit volume sludge treatment.
6.Distribution and clinical significance of HBV genotypes in patients with HBV infection in 30 regions of China

Objective To explore the distribution and clinical significance of HBV genotypes in patients with HBV infection in China. Method Serum samples were collected from 2922 patients with HBV infection. HBV genotyping was performed with type-specific primers polymerase chain reaction, and the virological and biochemical markers were detected, which differences in the genotypes distribution between various regions and liver function and virological markers between various HBV genotyping were analyzed. Result The genotype B, C, B + C, D of 2922 patients with HBV infection accounted for 15.9% , 83.5% ,0. 41%, 0. 21% respectively. In Northern China, genotype C was most prevalent, accounting for 90% of all cases, while it was less common in Southern China; genotype C was present in Zhejiang and Jiangsu provinces, but genotype B was comparatively more common in Guangdong, Hunan, Hubei, and Jiangxi provinces. B, C genotype HBV infection patients in the sex difference was not statistically significant; B genntypes compared with C genotype HBV infection patients, the average age of is less ( P <0.001 ) ;HBeAg positive rate of C genotype HBV infection patients are higher than that of B genotype ( P = 0. 023 ) ;Viral load of genotype C HBV infection patients is higher than that of genotype B ( P =0. 038 ) ;.Cholinesterase and Albumin levels of genotype C HBV infection patients are lower than that of genotype B ( Pvalues were 0. 016, < 0. 001 ). Conclusion There were HBV genotype B, C, B + C and D in Chinese patients with HBV infection, with genotype B and C being the major ones. Mainly in northern regions of genotype C, C genotype significantly reduced the southern region, some of the southern region dominated by B genotype. Genotype C HBV infection patients are older, and their HBeAg-positive rate is higher, and their liver damage is more severe, but their viral load is less.
7.Expression of HLA class Ⅱ antigens, interleukin-2 receptors and transferrin receptors of monocytes from patients with insulindependent diabetes mellitus

Expression of HLA class Ⅱ antigens(HLA-DR, DQ and DP), interleukin2 receptors(IL-2R) and transferrin receptors(TfR) of blood monocytes from 10 patients with insulin-dependent diabetes meIlitus (IDDM) were assayed with the indirect immune fluorescence technique using corresponding monoclonal antibodies and the FITC-labelled second antibody. The results showed that the number of HLA-DQ~+ monocytes was much more in diabetics than in normal controls. The percentages of HLA-DR~+ and HLA-DP~+ monocytes in diabetics were not different significantly from those in normal controls. Besides, IL-2R~+ and TfR~+ monocytes were also found to be very much increased in diabetics as compared with controls. It was possible that increased expression of HLA-DQ antigen, IL-2R and TfR of monocytes in patients with IDDM might play a role in the pathogenesis of the autoimmune reaction.
8.Comparison and analysis of therapeutic effects of different therapies on simple obesity.

OBJECTIVETo search for the best therapy for simple obesity.
METHODSEighty cases were randomly divided into 3 groups. The body acupuncture group were treated based on the syndrome of heat of stomach and intestine, syndrome of spleen deficiency and stagnation of dampness, and syndrome of spleen and kidney yang-deficiency; the auricular and body acupuncture group were treated by the syndrome treatment of body acupuncture combined with auricular point sticking; the observation group were treated by the combined auricular and body acupuncture treatment plus moving cupping on back-shu points. Body weight, BMI, body fatd, blood lipids and clinically main symptoms before and after treatment were investigated.
RESULTSThe total effective rate was 69.6% in the body acupuncture group, 76.0%. in the auricular and body acupuncture group, and 90.6% treated in the observation group, with significant differences in the therapeutic effect, clinically main symptoms, external indexes of obesity and kidney ang-blood lipid metabolism between the observation group and the body acupuncture group. The various indexes in the auricular and body acupuncture group were superior to those iwere tthe body acupuncture group, with no significant differences in most indexes.
CONCLUSIONAuricular and body acupuncture combined with moving cupping at back-shu points has obvious therapeutic effect on simple obesity and this is a better therapy for simple obesity.

9.Comparison of different threshold ~(18)FDG PET with computer tomography for defining gross tumor volume in non-small cell lung carcinoma

Objective Under different standard uptake value(SUV),to assess gross tumor volume (GTV) definition for non-small cell lung cancer(NSCLC) with 18-fluoro-deoxy-glucose positron emission tomography (~(18)FDG PET) both under definite threshold (42 percent threshold) and various relative threshold (threshold SUV/maximum SUV) derived from the linear regressive function,threshold SUV=0.307?(mean target SUV)+0.588,with computer tomography (CT).Methods Of 20 patients with non-small cell lung cancer,the CT GTV (GTV_(?)).PET GTV with 42 percents threshold (GTV_(42%)) and PET GTV with relative threshold (GTV_(?)) were obtained and compared.Results The mean GTV_(42%),,mean GTV_(?) and mean GTV_(CT) was (13 812.5?13 841.4),(24 325.3?22 454.7) and (28 350.9?26 079.8)mm~3,respectively,with the difference in mean GTV among these three methods significant (F =10,P<0.01).The GTV_(42%) was smaller than the GTV_(?) and the GTV_(CT)(P<0.01),with i(?)significant difference between GTV_(?) and GTV_(CT)(P=0.125).Conclusion The relative threshold is more suitable to define the gross tumor volume than the definite threshold.
10.Regulative mechanism of budesonide on endogenous hydrogen sulfide, cystathionine-gamma-lyase and cystathionine-beta-synthase system in asthmatic rats.

OBJECTIVETo investigate plasma hydrogen sulfide (H₂S) levels and cystathionine-gamma- lyase (CSE) and cystathionine-beta-synthase (CBS) mRNA expression in the lung tissues in asthmatic rats and to explore the roles of endogenous H₂S, CSE and CBS system in the pathogenesis of asthma.
METHODSThirty male Sprague-Dawley rats (age 5 to 7 weeks) were randomly divided into three groups: control, asthma and budesonide treatment (n = 10 each). The asthma model was established by ovalbumin (OVA) sensitization and challenge. The budesonide treatment group received inhaled budesonide before challenge. The contents of plasma H₂S were measured by spectrophotometry. The levels of CSE and CBS mRNA in the lung tissues were examined by reverse transcriptase polymerase chain reaction (RT-PCR).
RESULTSThe contents of plasma H₂S in the asthma group (61 ± 16 μmol/L) were significantly lower than those in the control group (84 ± 15 μmol/L) (P<0.01). The contents of plasma H₂S in the budesonide treatment group (71 ± 14 μmol/L) were not statistically different from those in the control and asthma groups. CSE mRNA and CBE mRNA expression in the asthma group were significantly lower than those in the control group (P < 0.01). The budesonide treatment group had a decreased CSE mRNA expression and CBE mRNA expression compared with the control group, but had significantly increased CSE and CBE mRNA expression compared with the asthma group (P < 0.01). There was a significantly negative correlation between H₂S contents in plasma and total inflammatory cells in bronchoalveolar lavage fluid (n = 30, r = -0.549, P < 0.01).
CONCLUSIONSPlasma H₂S levels and CSE and CBS expression in the lung decrease in asthmatic rats, which possibly promotes inflammatory cell aggregation to the airway. Budesonide may alleviate airway inflammation in asthmatic rats possibly through the system of endogenous H₂S, CSE and CBS.
