Objective To study the effect of human Slitrk1 gene on proliferation and differentiation of PC12 cells. Methods The CDS sequence of Slitrk1 was amplified by PCR, and then cloned into pcDNA4 vector. The recombinant plasmid pcDNA4/Slitrk1 were transfected into rat PC12 cells by lipofectamine. The stable expression cell clones were screened by RT-PCR. MTT method was used to detect the proliferation rate of PC12 cells. The morphologic changes in PC12 cells were observed microscopically. Results The stable cell lines expressing pcDNA4 (pcDNA4/PC12) and pcDNA4/Slitrk1 (ST1/PC12) were established. Compared to pcDNA4/PC12 cells, the growth rate of ST1/PC12 cells was decreased. In addition, pcDNA4/PC12 cells tend to grow as well as the normal PC12 cells. However, most of the ST1/PC12 cells adhered to the plate with one or two neurites. Conclusion Over-expression of human Slitrk1 gene inhibited the proliferation of PC12 cells and promoted the outgrowth of neurites. It is suggested that human Slitrk1 gene may be involved in differentiation of PC12 cells.

About 50% to 60% of the stage Ⅱ-Ⅲ colorectal carcinoma patient suffer from liver metastases.Recently, there is a research hotspot on how to improve the prognosis of the colorectal carcinoma liver metastases (CRLM) patient. It can elucidate the way how the carcinoma cells transfer from the original tumor to the liver, through establishing a CRLM animal model. An ideal CRLM animal model should perfectly mimic the total procedure of the metastases pathway, including the change of the cytology behavior and the development of the tumor biology.Besides, the model should be practical,tumor-predictable and accord with the ethical consideration.Until now, there is not an ideal CRLM animal model can perfectly match those conditions. This review goes through the advantages and disadvantages of different CRLM animal model methods used for research.

Chinese Pharmacological Bulletin 1986;0(05):-
Aim To explore the mechanism of polydatin (PD) treating shock through investigating influence of PD in protein kinase C (PKC) activity of myocardial cells when myocardial cells are treated by ischemia and hypoxia. Methods The hypoxia model was set up by drawing off the oxygen of an enclosed acryl glass box and ischemia model by low sugar culture medium. The activity of PKC was measured by ?-scintillation counter. Results The activity of PKC of myocardial cells cytoplasm in normal situation after the cells were treated by PD decreased(vs normal group P0.05). Conclusion PD antagonizes the damage by reversing the PKC activities, which is associated with the anti-shock effect of PD.

Chinese Journal of Microbiology and Immunology 2017;37(3):188-193
Objective To investigate the role and mechanism of retinoblastoma protein-associated proteins 48 (RBBP4) in HIV-1 latency. Methods CEM-Bru cells latently infected with HIV-1 were stimulated with 25 ng/ml of tumor necrosis factor alpha (TNF-α) in combination with 10 ng/ml of interleukin-2 (IL-2).Chromatin immunoprecipitation (ChIP) was performed to detect the changes in RBBP4 and in histone deacetylases 1 and 2 (HDAC1/2) binding to long terminal repeat (LTR).Binding activities of HDAC1/2 and RNA polymerase Ⅱ (RNA Pol Ⅱ) to LTR and acetylated histone H3 at LTR region were detected by ChIP after partially interfering the expression of RBBP4 in CEM-Bru cells with electroporation.Initiating and elongated transcripts were measured by RT-PCR.Results The binding activities of RBBP4 and HDAC1/2 to LTR in HIV-1 latently infected cells were enhanced significantly as compared with those in TNF-α and IL-2 co-stimulated cells.Fewer RBBP4 and HDAC1/2 bound to LTR following the interference of RBBP4 expression, which was accompanied with enhanced histone acetylation and strengthened binding activity of RNA Pol Ⅱ to LTR.Moreover, more initiating transcripts were detected in HIV-1 latently infected cells after the RBBP4 expression was interfered by electroporation. Conclusion RBBP4 contributes to the maintenance of HIV-1 latency, in which HDAC1 and HDAC2 might be involved.
