1.Proliferation of dermal fibroblasts from human,Yorkshire pigsor SD rats cultured in DMEM with different concentrations of serum

Objective: To establish an optimized culture condition f or the growth of dermal fibroblasts. Methods: Human, Yorks hire pig or SD rat fibroblasts were cultured in DMEM with fetal boven serum(P BS) at the concentrations(ml/L) of 100 (A), 50 (B), 20 (C) or 20 supplemente d with 0.025 g/L boven pituitary extract (BPE)(D). The proliferation of the cell s were examined by microscopic observation,MTT assay and BrdU analysis. Results: Human dermal fibroblasts grew better in the medium with FBS at 100 ml/L, 50 ml/L and 20 ml/L supplemented with BPE, Yorkshire pig dermal fibroblasts did in medium with FBS at 50 ml/L and 20 ml/L supplemented with BPE . SD rat dermal fibroblasts did in the medium with 20 ml/L FBS supplemented wi th BPE. Conclusion:The favarable serum concentration in me dium for human, Yorkshire pigs and SD rats dermal fibroblasts was different,but the medium with 20 ml/L FBS supplement with BPE is suitable for all 3 kinds of f ibroblasts.
2.Anterior approach or combined anterior and posterior approaches for severe ossification of cervical posterior longitudinal ligament

BACKGROUND:It is controversial whether anterior approach alone, or combined anterior and posterior approaches were used for high level and multiple segments of severe ossification of cervical posterior longitudinal ligament.
OBJECTIVE:To explore the difference of anterior approach versus combined anterior and posterior approaches for the treatment of high level and multiple segments of severe ossification of cervical posterior longitudinal ligament.
METHODS:A total of 21 cases of high level and multiple segments of severe ossification of cervical posterior longitudinal ligament were included in this study. There were 9 males, aged 56-72 years, and 12 females, aged 58-70 years. We used anterior decompression and titanium mesh bone graft fusion in 11 cases which lesion located between C2-5 vertebra, and ossification excision, combined anterior (titanium mesh plate and screw) and posterior (lateral mass screw) approaches in 10 cases which between C3-7 vertebra. Japanese Orthopaedic Association score system was used to evaluate the results. The excellent and good rate and improvement rate were calculated.
RESULTS AND CONCLUSION:The excellent and good rate was 90%and improvement rate was 82%in 10 cases using combined anterior and posterior approaches. The excellent and good rate was 73%and improvement rate was 73%in 11 cases using anterior treatment alone. Significant differences in the excellent and good rate and improvement rate were detected between the two groups (P<0.05). These suggested that combined anterior and posterior approaches for high level and multiple segments of severe ossification of cervical posterior longitudinal ligament is a better operative procedure.
4.Physical and chemical factors affecting the allergenic potency of the house dust mite dermatophagoides farinae extract

The physical and chemical factors affecting the allergenic potency of the house bdust mite Dermatophagoides farinae extract were determined by skin test and ELISA technique in the asthmatic patients sensitive to the mite. The results showed that the allergenic potency of the extract could be reduced by heat sterilization, trypsin trea-tment and lyophization,but not lost completely,and that the potency could be increasedby supersonic treatment and repeated freeze-thaw(but less than 10 times).It was alsoshowed that the allergenic potency of the extract preserved at -20℃ was more stablethan those preserved at 4℃ or at room temperature,and that the mite extract preservedat 4℃ more than 9 years still had allergenic potency.
5.XBJ Oxgen Inhalation Treat Chronic Pulmonary-cardiac Disease in Acute Period

[Objective] To observe the clinical therapeutic effect of XBJ oxygen inhalation on chronic pulmonary-cardiac disease in acute period.[Method] Randomly divide 150 cases into blank control group(1),vein administration group(2),oxygen inhalation group(3),observe their clinical effect,lung function,blood-gas analysis,cardiac function grade and other indexes.[Result] Group 3 could improve patients lung function,blood-gas analysis,cardiac function grade and other indexes,which had marked advantages compared with other 2 groups.
6.Correlation between revascularization effect and calcium content of a kind of man-made compound bone on the femoral head defects and necrosis of canines

[Objective]To observe the effect of composite(AACB/BMP/bFGF)of allogeneic antigen-extracted cancellous bone(AACB),bone morphogenetic protein(BMP)and basic fibroblast growth factor(bFGF)on the repair of defective and necrotic areas of the femoral head,and evaluate the revaseularization the femoral head defect and necrosis model using liquid nitrogen,and randomly divided into 3 groups with 12 femoral heads in each group,which were implanted with nothing(group A),AACB/BMP(group B)and AACB/BMP/bFGF(group C)respectively.Every femoral head was implanted with about AACB 0.33 g,BMP 12.5 mg and bFGF 2000 U.Two canines were sacrificed respectively at 3th,6th,and 12th weeks after operation in every group.Histologically,immunohitochemical vessel staining,vessel count and vessel surface area image analysis,calcium content and the correlation between revascularization effect and calcium content were investigated in the femoral heads after the operation.[Result]Histology,the detects were still infilled by fibrillar tissue in group,A;there were a great deal of new bone formed in group B;and the detects were repaired completely in group C 12 weeks after operation.Measurement of vessel count and vessel surface area showed a large quantity of new vessel proliferated around the infilling material in group C on the 3rd week.The vessel count and vessel surface area continuously increased on the 6th week and 12th week.They were more than those of group B and group A at every definite time point(P
7.The management and prognosis of postmastectomy local recurrence in 477 breast cancer patients

Objective To explore an optimal treatment and to study the prognosis related factors of breast cancer patients with local recurrence after mastectomy. Methods From 2002. 7 to 2005. 2, 477female patients with loco-regional recurrence of breast cancer treated in Tianjin Cancer Hospital were analyzed retrospectively. Results In 477 cases, recurrence within 1 year after mastectomy accounted for 26. 2% , recurrence within 2 years accounted for 61. 2%. There were 310 cases with metastasis after local recurrence was diagnosed, accounting for 65.0%. 5-year total survival rate after recurrence is 48.4%.Local control rates varied in subgroups with different recurrence site, clinical subtypes, radiotherapy fields,with or without radiotherapy, surgical resection or excisional biopsy ( P<0. 05 ). There was a statistical difference in distant metastasis rate and 5-year survival rate among subgroups which had different clinical stage of primary tumor, disease-free interval, clinical subtypes or treatment methods ( P<0. 05 ). Simplistic treatment option, late clinical stage of primary tumor and triple-negative breast cancer were the independent factors predicting poor prognosis for recurrent breast cancer ( P<0.05 ). Conclusions Multi-site recurrence and triple-negative breast cancer lead to a poor local control. Local expansion of radiotherapy combined with surgery improves the local control rate. Patients with late clinical stage of primary tumor,recurrence within 2 years, triple-negative breast cancer are likely to have distant metastasis when recurrence is diagnosed. Combined treatment program improves survival rate.

9.Detection and grading of endolymphatic hydrops in Ménière's disease using 3D FLAIR MRI.

OBJECTIVE:
To evaluate the extent of endolymphatic hydrops as shown by three-dimensional fluid-attenuated inversion recovery magnetic resonance imaging (3D FLAIR MRI) performed 24 hours after bilateral intratympanic gadolinium administration and discuss the positive rate of endolymphatic hydrops in vestibule and cochlea.
METHOD:
Twenty-four hours after bilateral intra-tympanic 8 times diluted gadolinium administration, three-dimensional fluid-attenuated inversion recovery MRI, using a three-Tesla unit, was performed in 48 patients, and then assessed the extent of endolymphatic hydrops in the MRI images.
RESULT:
Forty-eight patients showed different levels of enhancement of perilymth in the inner ear. In these patients, obvious signs of endolymphatic hydrops were visualized in vestibule,including 8 slight hydrops, 17 mild hydrops and 23 severe hydrops with a diagnostic rate of 83.3%. There were almost no complications in all 48 patients after bilateral intra-tympanic injection except short vertigo in some of them.
CONCLUSION
3D FLAIR MRI resonance imaging has a high positive diagnostic rate in assessing endolymphatic hydrops of Ménière's disease and provides direct imaging evidence for diagnosing Ménière's disease.

10.A thin-slice radioanatomic study of jugular foramen

Objective To observe and analyze the CT and MR imaging of the structures in the region of the jugular foramen (JF) on the base of thin-slice anatomic study. Methods Having been scanned by multislice CT and 1.5T MR scanner, two formalin-preserved adult cadavers were dissected into 1.0 mm thickness contiguous sections in transverse plane. Twenty cases without skull base and nasopharyngeal history received routine and post-contrast CT examinations with spiral and HQ mode. Twenty healthy volunteers received MR scanning, including SE T 1WI, FSE T 2WI, and 3D RF-FAST (3D Radio-Frequency Fourier Acquired Steady-State) sequences. Results JF region was divided into three levels, which included inner aperture, the jugular cavity, and the outer aperture. At the entrance of JF, there were glossopharyngeal canal and vagal canal, which wrapped the Ⅸ nerve and Ⅹ and Ⅺ nerves, respectively. CT images could display these canals in 20 cases (100%). Furthermore, the Ⅸ, Ⅹ, and Ⅺ nerves could be identified on different MR sequences. 17 cases (85%) were displayed on 3D RF-FAST, 14 cases (70%) on SE T 1WI, and 10 cases (50%) on FSE T 2WI, respectively. From the anterior to the posterior compartment within the JF cavity, the structures ranged as follows: the Ⅸ nerve, the inferior petrosal sinus, the Ⅹ and Ⅺ nerves, and the jugular bulb. These neuro-vessel structures could not be distinguished on CT, SE T 1WI, and FSE T 2WI images, except for 3D RF-FAST (16 cases, 80%). The outer aperture of JF was adjacent to the hypoglossal canal. The shape of JF outer aperture could be evaluated on CT images. On the oblique plane, which was parallel to the hypoglossal canal, the posterior cranial nerves could be detected to enter/exit the skull through the JF and hypoglossal canal separately. Conclusion The complement of CT and MR imaging would be helpful to distinguish the structures in the region of JF.
