3.Discrimination of varieties of peony in Huangqin decoction.

By studying the varieties of peony and analyzing the prescription of Huangqin decoction, the authors explored the differences between Paeoniae Radix Alba and Radix Paeoniae Rubra in varieties, origin, processing method and clinical efficacy and compare their efficacies to define Paeoniae Radix Alba or Paeoniae Radix Rubra in Huangqin decoction recorded in Treatise on Febrile Diseases. In the study, the authors clarified the development and change of the variety in various historical periods according to the earliest ancient herbal book recording the variety and the development sequence of main herbs, and made clear the relations between Paeoniae Radix Alba and Paeoniae Radix Rubra. The modern application of Paeoniae Radix Alba started in the Song Dynasty. Although it was processed in different ways from Paeoniae radix Rubra, they shared the same original plant varieties. On the basis of the historical origin, botanical origin, producing place and processing method, the authors made clear the evolvement of peony varieties, discussed and analyzed the developments and changes for the combined to the separate administration of Paeoniae Radix Alba and Paeoniae Radix Rubra and defined the peony variety in Huangqin decoction. Through the textual research on ancient herbal books, the authors confirmed that more than 2 000 years ago, Paeoniae Radix Alba didn't appear when Treatise on Febrile Diseases was written. According to the records in Miscellaneous Records of Famous Physicians that "its roots were collected and dried in February and August", it was inferred that the use of Paeoniae Radix Rubra conformed to the historical facts.

Physiologically based pharmacokinetic model (PBPK), a mechanistic mathematic model, which can simulate the absorption, distribution, metabolism and excretion of drugs, is being more widely used in pharmaceutical research and development areas. This article reviews primarily the recent advances in the procedure of establishing a PBPK model, including specifying of the PBPK model structure, specification of the tissue model, writing of equations, set of model parameters, simulation and evaluation. Application significance, major challenges and future developments of PBPK model in pharmaceutical areas are also discussed.

9.Research Progress in Cofactor Engineering of Xylose Metabolism in Recombinant Saccharomyces cerevisiae

Cofactor engineering, a vital part of metabolism engineering, changes the redox cofactor regeneration approach. Its main goal is to rebuild the components of metabolic products. The bioconversion of xylose for the production of ethanol is being studied intensively because ethanol is an alternative energy source and a potential liquid fuel. Saccharomyces cerevisiae has been traditionally used in producing ethanol from fermentable sugars but it cannot utilize xylose, only its isomer xylulose. Introduction of the xylose fermentation pathway from Pichia stipitis into S. cerevisiae enables xylose utilization in recombinant S. cerevisiae, but the ethanol yields of xylose fermentation with recombinant S. cerevisiae has been low and large amounts of the byproduct xylitol are produced. The major reason is that the catabolism of xylose with the fungal pathway leads an imbalance of redox cofactor. The process of the catabolism of xylose requires NADPH and NAD~+, both of which have to be regenerated in separated processes. More and more attention has therefore focused on the redox cofactor balance in S. cerevisia. The research progress of cofactor engineering to solve the imbalance of redox cofactor in xylose metabolism recombinant S. cerevisiae was introduced. This included expression of transhydrogenase, increasing the utilization of NADPH, and achieving the anaerobic reoxidation of NADH. Reversing the cofactor specificity of enzymes is another effective way.
10.Analysis of characters and biochemical profiles of 27 cases with EDTA-Dependent Pseudothrombocytopenia

Objective To analyze the characters about anticoagulation EDTA on the platelet aggregation ,and discuss the risk factors about EDTA‐PTCP by analyzing the biochemical parameters .Methods 30 700 EDTA‐K3 blood samples were collected from January 2014 to August 2015 .WBC was performed using Sysmex XN‐3000 and 27cases were identified of EDTA‐PTCP .The blood samples of 27 subjects were recollected and WBC was repeated using EDTA‐K3 and sodium citrate blood ,respectively .Manual platelet counting and smear microscope examination on periphery blood were also performed .Biochemical parameters of 27 subjects with EDTA‐PTCP were compared with 50 randomly selected non EDTA‐PTCP controls .Results Platelet counts in citrated blood were significantly higher than EDTA blood ,the difference was statistically significant(P< 0 .05) ,but similar to manual platelet counting(P>0 .05) .ALT ,Glu and TG in patients with EDTA‐PTCP were significantly higher than normal people ,but HDL was significantly lower than control group ,the difference was statistically significant(P<0 .05) .Conclusion EDTA‐dependent pseudo‐thrombocytopenia varies from people with different disease or using different medicine .It may be relative to hyperglycemia and hy‐perlipidemia .Another approach for rectifying ,changing anticoagulant or manual platelet counting is a better way lead to correctly di‐agnose when EDTA‐PTCP occurred .
