Objective To investigate the effects of arsenic trioxide(As2O3)combined with ascorbic acid(AA) on the apoptosis and differentiation of myelocytic leukaemia cells.Methods The acute promyelecytic leukaemia cell lines (NB4 and MR2)and erythroleukemia cell line(KS62)were cultured in vitro.Grouping wasbased on different concentration of As2O3(0,0.1,0.2,0.5,1.0 μmol/L),which WaS used a8 control groups.Then,AA(113.0μmol/)was added into each group.Cell morphologic changes of cell lines NB4,MR2 and K562 were observed under light microscope;The apoptosis symbols [Annexin V(+)/PI(-),Annexin V(+)/PI(+)]and differentiation symbols(CD11b and CD33)were detected by flow cytometry 96 hours later.Results
2.The influence of PBL combined with CBS education on reporting cognition and reporting intention of adverse events

Objective To investigate the influence of PBL combined with CBS education on reporting cognition and reporting intention of adverse events.Methods Using convenience sampling method,66 registered nurses working for over 6 months were extracted as the study object.The training group was set up and the training files were established,PBL combined with CBS mode were implemented for 6 months from March to August 2013,once a month,2 hours per time.Training cases were designed and written,teaching staff was organized to carry out the training,problems were raised in class and discussed on site.Training staff looked up relevant data,collected information,organized another focused discussion and summarized the results.The Adverse Event Reporting Cognitive Questionnaire and Adverse Nursing Event Reporting Intention Questionnaire were adopted to evaluate reporting cognition and reporting intention before and after the training.Results After the implementation of PBL combined with CBS education,cognition of nurses for adverse events scored (7.23±1.20),higher than that before the intervention,(6.48±1.49),t=3.17,P < 0.01.The numbers of nurses to report adverse events such as potential vulnerabilities and not doing any harm were 42 and 45 before the intervention,while 53 and 56 after the intervention.Conclusions Using PBL combined with CBS model to implement educational interventions can improve reporting cognition and report intention on nursing adverse events of nurses.

OBJECTIVE:To establish an analytical method for the determination of prednisolone and prednisolone-hemisuccinate in biological media METHODS:Using RP-HPLC,biological media were extracted with a mixture of n-pentane and methyl tert-butyl ether(2∶3) The mobile phase consisted of acetonitrile and trisodium citrate buffer (33∶67) The sample was eluted on C18 column and detected at 254nm RESULTS:The extraction recoveries of two drugs were over 80% and the RSDs of within-day and day-to-day were below 3% CONCLUSION:The method is simple,rapid,sensitive,accurate and good enough to be applied to the pharmacokinetic study of new preparation of prednisolone and can also be applied to other similar glucocorticoids
4.Preparation of Compound Anisodamine and Neostigmine Sustained-release Tablet and Study on the in vitro Drug Release Behavior

OBJECTIVE:To prepare the Compound anisodamine and neostigmine sustained-release tablet and study the in vitro drug release behavior. METHODS:Using raceanisodamine and neostigmine methyl sulfate as main medicines,hydroxypropyl meth-yl cellulose as sustained release skeleton material,magnesium stearate as the lubricant,polyvinyl pyrrolidone as the adhesive,pre-gelatinized starch as the thinner,microcrystalline cellulose as the disintegrant and filler,Compound anisodamine and neostigmine sustained-release tablet was prepared by wet granulation method and direct compression method. The in vitro cumulative release rate within 12 h of the 2 main medicines was detected by HPLC method. RESULTS:Compound anisodamine and neostigmine sus-tained-release tablet was successfully prepared,and the in vitro release was basically completed within 12 h,with accumulative re-lease rate of 91.3% for anisodamine and 96.5% for neostigmine. CONCLUSIONS:Compound anisodamine and neostigmine sus-tained-release tablet that can cumulatively release for 12 h is successfully prepared.
5.Development of a nonlinear 3D solid finite element model for the calculation of bending moments of flexural members

A numerical model is developed in this paper to calculate the bending moments of flexural members through integration in 3D solid finite element analyses according to the nonlinear constitutive model of concrete and the elastoplastic constitutive model of steel, utilizing the stress condition of the cross-section, considering the destruction characteristic of reinforced concrete members, and based on the plane cross-section assumption. The results of this model give good agreement with those of the classical method. Consequently, we can also deduce the corresponding numerical expression for eccentrically loaded members according to the analysis method.

Methods:Using gene recombination techniques,four luciferase reporter gene plasmids containing different length of human MUC5AC gene promoter were constructed.Site-directed mutagenesis technique was used to establish mutants of Sp-l and NF-?B site in MUC5AC gene promoter; the relative luciferase activities were detected in the transfected human pulmonary A549 cells. Objective To study the correlation of the adiponectin receptor 1 gene rs1139646C/G and rs10920531C/A single nucleotide polymorphisms with Chinese Han women with polycystic ovary syndrome (PCOS).Methods For PCOS group (n =80) and control group (n =80),polymerase chain reaction (PCR) was used to amplify the upstream and downstream 250 bases of rsl139646C/G and rs10920531C/A,respectively.PCR products were directly sequenced.Results For rs1139646C/G loci sites between PCOS and control groups,all were the homozygous genotype CC,and no statistical significance was found for genotype frequency and allele frequency between two groups.For rs10920531C/A loci sites between PCOS and control groups,no statistical significance was found for genotype frequency and allele frequency between two groups (P =0.683,0.580),and no statistical significance was found for two loci with genetic linkage disequilibrium analysis (x2 =0.307,Fisher 's P =0.579610,Pearson' s P =0.579608).Conclusions Adiponectin receptor 1 gene rs1139646C/G and rs10920531C/A single nucleotide polymorphisms have nothing to do with Chinese Han PCOS women.Genetic linkage disequilibrium analysis has nothing to do with PCOS.The rs1139646C/G in Chinese Han population is relatively stable site,and no locus mutation is found.
8.The effects of electrical stimulation of the pelvic floor on neurogenic bladder dysfunction after spinal cord injury: A systematic review and meta-analysis of randomized and controlled trials

Objective:To determine the effect of pelvic floor electrical stimulation combined with conventional treatment on neurogenic bladder dysfunction after spinal cord injury.Methods:The Pubmed/Medline, Embase, Cochrane, CINAHL, China National Knowledge Infrastructure, CQVIP and Wanfang databases were electronically searched for reports published before April 2019 of randomized and controlled trials testing the effect of electrical stimulation of the pelvic floor on neurogenic bladder after spinal cord injury. Meta-analysis of all the reports collected was performed.Results:Seven randomized and controlled trials with 319 patients were identified. Together they showed that compared with conventional treatment, pelvic floor muscle stimulation better increased bladder capacity and reduced the volume of residual urine. Average single urination output was increased, while the frequency of urination decreased along with episodes of urinary incontinence. Average lower urinary tract symptoms scores were also significantly better.Conclusions:Electrical stimulation of the pelvic floor combined with conventional therapy may be more effective than conventional therapy alone in alleviating the symptoms of neurogenic bladder.
9.Levothyroxine sodium in treatment of severe pulmonary tuberculosis with euthyroid sick syndrome

Objective To evaluate the efficacy of low-dose levothyroxine sodium in treatment of severe pulmonary tuberculosis with euthyroid sick syndrome(ESS). Methods One hundred and twenty inpatients with severe pulmonary tuberculosis and ESS were randomly divided into treatment group and control group by gender, age, disease duration and severity. Both groups were given anti- tuberculosis, antiinfection treatment and nutritional support for 2 weeks; patients in treatment group were given low-dose levothyroxine sodium additionally. Thyroid function, clinical improvements, increase of albumin, reduction of acid-fast bacilli, improvements on images and the mortality rates were compared between the groups.Results After 2 weeks of treatment, symptoms including fever, cough and night sweats were improved in both groups. Marked improvements were observed in 19 patients(31.7%)of treatment group and 8 patients (13.3%)of control group(χ2 = 5. 73, P < 0.05). Clearance rate of acid-fast bacilli in treatment group was 25.0%(15/60), but that in the control was only 6.7%(4/60)(χ2 = 7. 50, P < 0.01). Serum albumin in the treatment group was increased to(34.2 ±0.4)g/L after the treatment, and that in the control group was(29.1 ±0.6)g/L(t =2.42, P<0.05). T3 and FT3 were significantly increased in both groups, but more significant difference was observed in the treatment group(t = 59. 42 and 50. 66, P < 0. 01). No empty closed after treatment in both groups, but the effective rate in treatment group was significantly higher than that in the control group(93.3% vs. 76.7%, χ2 =6. 54, P<0.05). Two patients in control group died(2/60, 3. 3%), while no death was reported in treatment group. Conclusions Low-dose levothyroxine sodium treatment is effective for ESS in patients with severe pulmonary tuberculosis.Improvement on low T3 syndrome may be an important indicator for the overall improvement or recovery.
10.PHARMACOKINETICS OF SITE-SPECIFIC DELIVERY OF DEXAMETHASONE-DEXTRAN PRODRUG IN RAT GASTROINTESTINAL TRACT

AIM To explore whether dexamethasone-dextran (260 000) has the characteristics of site-specific delivery in rat gastrointestinal tract. METHODS Dexamethasone prodrug and dexamethasone were administered to rat ig at the dose of 5 μmol*kg-1. The distribution of dexamethasone in the contents and mucosa of different parts of the rat GI tract at different time intervals and its concentration in plasma were determined by HPLC. RESULTS Dexamethasone was mainly released in the cecum and colon contents and mucosa after oral administration of dexamethasone prodrug. The absorption was reduced significantly. The peak time of the drug in plasma was 8.1 h, and the peak concentration was 32 μg*L-1. However, free dexamethasone was found mainly in the contents and mucosa of the stomach, proximal and distal small intestine. The peak time of the drug in plasma was 2.2 h, and the peak concentration was 2120 μg*L-1. CONCLUSION Dexamethasone can be specifically delivered to the large intestine by using dexamethasone-dextran (260 000). It appears that the prodrug has a potential in the treatment of inflammatory bowel disease.
