1.Effects of preconditioning with emulsified isoflurane on Inflammatory response to myocardial ischemiareperfusion injury in rats

Objective To investigate the effects of preconditioning with emulsified isoflurane(eISO)on inflammatory response to myocardial ischemia-reperfusion(I/R)injury in rats.Methods Forty SD rats of both sexes weighing 250-280 g were randomly allocated into 4 groups(n =10 each):groupⅠ sham operation(S);group Ⅱ myocardial I/R + normal saline(NS); group Ⅲ I/R + eISO and group Ⅳ I/R + 30% intralipid(Ⅱ.)(vehicle for eISO).Myocardial ischemia was induced by occlusion of anterior descending branch of left coronary artery for 30 min followed by 180 min reperfusion.NS,30% intralipid and elSO 2 ml/kg were infused iv over 30 min at 30 min before myocardial ischemia in groups Ⅱ,Ⅲ and Ⅳ respectively.The animals were killed at the end of 180 min repeffusion.Their hearts were removed for determination of infarct size and myocardial NF-κB p65 and ICAM-1 expression(by immuno-histochemistry)and plasma concentration of TNF-t(by radioimmunoassay).Results Myocardial I/R induced myocardial infarct and significantly increased plasina TNF-a concentration and myocardial ICAM-1 and NF-κB p65 expression in gro up Ⅱ,Ⅲ and Ⅳ as compared with'sham operation group (Ⅰ).Plasma TNF-a concentration and myocardial ICAM-1 and NF-kB p65 expression were significantly lower in group Ⅲ(eISO)than in group Ⅱ and Ⅳ.Conclusion Down-regulation of myocardial NF-kB and ICAM-1 expression and inhihition of inflammatoy response are involved in the mechanism by which preconditioning with iv elso protects against myocardial I/R injury.
2.Safety of delivering nitric oxide through nitrous oxide path in anesthesia machine

To evaluate the feasibility of delivering NO through N_2O path in anesthesia machine,NO at 800 ppm was delivered through nitrous oxide path in anesthesia machine SIEMENS Ventilator 710 or NARKOMED 2B,to ventilate a test lung. The minute volume of ventilation (MV) was set from 2 to 6L/min, and NO concentration from 10 to 80 ppm. NO and NO_2 concentrations were measured by hemiluminescence and electro-chemical fuel cell technique. SIEMENS Ventilator 710:Non-rebreathing model,its inspiratory limb was prolonged to 7 meters,the NO level was adjusted at 80ppm before passing through soda lime, NO and NO_2 samples were taken before absorber,after absorber, 1, 2,4 and 6 meter site in inspiratory limb. NARKOMED 2B:Oxygen flows were at the rates epual to the MV mentioned above, NO concentration was at 10,20,40,60 and 80 ppm,the inspiratory gas mixture was sampled immediately passing through absorber, NO_2 levels were less than 5.0ppm in all conditions listed above. With SIEMENS Ventilator 710, the highest level of NO_2 was 3.45 ppm before absorber and 2.43 ppm after absorber,with NARKOMED 2B,that was 3.6 ppm. 3.The pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats

Objective To explore the pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats. Methods Ninety SD rats were randomly divided into three experimental groups:sham operation group (group A), warm hepatic ischemia/reperfusion group(group B and group C). Group C was given ischemic preconditioning treatment. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were detected 0 h, 2 h, 6 h, 12 h and 24 h after ischemia reperfusion injury. Levels of TNF-α and IL-1β were tested detected by ELISA. Levels of malondialdehyde (MDA) and superoxide dismutase (SOD) of hepatocytes were detected at the same time points. Mitochondrial membrane potential was examined to assess ischemia reperfusion injury of hepatocytes in rats using chart of intensity of JC-1 in mitochondria. Results The serum levels of ALT, TNF-α, IL-1β, and MDA were significantly higher in hepatic warm ischemia reperfusion group and ischemic preconditioning group than those in sham operation group (P<0.05). Values of prothrombin activity and cholinesterase were significantly lower in liver warm ischemia reperfusion group and ischemic preconditioning group than those of sham operation group (P<0.05). The SOD level of liver was significantly lower in warm ischemia reperfusion group and ischemic preconditioning group than that in sham operation group. The indexes were better in ischemic preconditioning group than those of warm ischemia reperfusion group (P<0.05). The mitochondrial membrane potential level of liver cells reached the lowest value 0 hours after ischemia and reperfusion, and then increased gradually within 24 hours (P<0.05). And the level of mitochondrial membrane potential of liver cells was significantly higher in ischemic preconditioning group than that in warm ischemia reperfusion group (P<0.05). Conclusion Ischemic preconditioning may play a protective role in warm ischemia-reperfusion injury in rats. Ischemic preconditioning may significantly decrease the levels of ALT, AST, TNF-α, IL-1βand MDA, and increase the SOD activity in hepatocytes. Thedamage of mitochondrial membrane potential is decreased after ischemic preconditioning, which might be the pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats.
5.Expression of human mannose-binding lectin in mice liver

Objective:To amplify and clone the human mannose-binding lectin(MBL) gene and to express its recombinant protein in mouse liver tissue.Methods:After the human MBL cDNA was amplified by PCR and identified by sequencing and restriction mapping,it was inserted into eukaryotic expression vector pcDNA3.The recombinant plasmid pcDNA3-MBL was injected into mice in large quantity and large volume in a short time through tail vein.These mice were sacrificed 8 h after the injection.MBL were examined in serum and hepatic tissue with Western blot and immunohistochemistry.Results:After the human MBL gene was amplified and sequenced correctly,it was successfully inserted in the eukaryotic expression vector pcDNA3.Eight hours after the pcDNA3-MBL plasmid were injected into mice through tail veins,human MBL could be found both in serum and hepatic tissue with Western blot and immunohistochemistry examination.Conclusion:Systemic injection of pcDNA-MBL can result in human MBL protein expression in mice liver and secretion into blood.This result may provide a new idea to treat the congenital MBL insufficient patients,who have the predisposition of infectious diseases.
6.Double-equilibration technique for measuring liquid/gas partition coefficients of isoflurane

This study was designed to establish methods to measure partition coefficients of inhaled anesthetics at 1 atmosphere and 37 C by using syringe-flask double equilibration technique. Olive oil/gas (O/G), saline/gas (S/G), and water/gas (W/G) partition coefficients of isoflurane were 90.20?2.01, 0.6256?0.109, and 0. 741?0.025, respectively. Even though O/G was 150 folds of S/G,coefficients of variation for both O/G and S/G were less than 3%. The results indicate that the established double-equilibration technique has a very high repeatability and is applicable to wide ranges of inhaled anesthetics' solubilities.
7.The influence of high-voltage electrical burn on the thrombomodulin, protein C, protein S, and D-Dimer in SD rats

Objective To investigate the influence of high-voltage electrical burn on the throm-bomodulin (TM), protein C (PC), protein S (PS) and D-Dimer (D-D) in SD rats. Methods One hundred and twenty healthy SD rats were divided into the fake high-voltage electrical burn groups (FHEB), high-voltage electrical burn groups (HEB) according to the random number table, with 60 rats in each group. Ten rats were taken from each group at 15 minutes before injury. Plasma were collected from heart blood. Fifty SD rats of HEB group with voltage regulator and experimental transformer. The remaining fifty SD rats of FHEB group were sham injured with the same devices without electric current. At 5 minutes and 1, 2, 4, 8 hour (s) post injury, 10 rats of every group were randomly chosen at each time point for observation of the concentrations of TM, PC, PS and D-D. Plasma were collected from heart blood. Data were processed with analysis of variance of factorial design and LSD test. Results Compared with the FHEB group, the concentration of TM from 5 minutes to 8 hours post injury in HEB group was higher significantly (P < 0.05). Exception of the concentrations of PC and PS at 15 minutes before injury, the concentrations of PC and PS were lower than those of FHEB group (P < 0.05). The concentration of D-D in HEB group peaked at 8 hours post injury in (173.05 ± 4.08) ng/mL. Conclusion High-voltage electrical burn at early stage can increase the concentrations of TM, D-D, as well as decrease the concentrations of PC and PS, which are not only causing the vascular endothelium damage but also possessing serious effect on the thromboplastin function of SD rats.

The aim of the present paper intends to explore the international experiences of healthcare profes-sional organizations and its enlightenment for China. The functions of health professional organizations in advanced countries include certification and accreditation, self-discipline, academic exchanges, political participation, service for industry, member rights protection and so on. The legal authorization, comprehensive supervision and competitive market mechanisms which make sure these functions perform well are worth learning for China.
9.Analysis on the development of healthcare professional organizations in China

This article is based on a questionnaire survey of healthcare professional organizations and interview leaders of them. The results of this investigation show that the number of staff is small, and lack of highly qualified human resources. The organizations rely on donation for income and some of them run at a deficit. The functions such as professional self-discipline are not well performed. The analysis reveals four reasons why functions are not well per-formed:the bureaucratic style of the professional organizations and unclear legal status;the limited authorization from legal and administrative bodies;the defects in the governance system and the lack of capacity. Four aspects of policy recommendations are were proposed: to clearly define the role of the professional organizations and authorization through legal and administrative procedures; to strengthen support of professional organizations through various poli-cies;to improve governance and build comprehensive regulation mechanisms;to enhance the capacity of these organi-zations and so on.
10.Analysis of the role and function of professional organizations in health governance in China

The characteristics of health care require that health care professional organizations play an impor-tant role in health governance. The role of health care professional organizations in China has not been clearly defined and their functions have not been fully developed. This paper explores the nature, and the functions of health care professional associations in China. Based on the analysis of health governance characteristics in China, the role, and functions of the health care professional organizations in China’s health governance were defined and relevant policy recommendations are were proposed.
