3.Efficacy analysis of bortezomib-based chemotherapy for 28 cases with multiple myeloma

Objective To examine the efficacy and adverse reactions of bortezomib-based chemotherapy in treatment of multiple myeloma (MM).Methods Twenty-eight patients with multiple myeloma received a joint chemotherapy containing bortezomib.The efficacy was determined according to EBMT criteria.Results 28 patients received the chemotherapy,20 patients were newly diagnosed and 8 patients were relapsed or refractory.25 patients can be evaluateed efficacy.The total response rate was 100 % (25/25),consisting of 5 patients with complete response (CR),10 patients with almost complete remission (nCR),10 patients with partial remission (PR).The main side effects include peripheral neuropathy,thrombocytopenia,gastrointestinal disorders and viral infections.These side effects were improved by symptomatic treatment and generally did not affect the treatment.Conclusion For the newly diagnosed and relapsed or refractory multiple myeloma cases,bortezomib-based chemotherapy is a safe and effective therapeutic drug with rapid onset,high treatment response rate,and adverse reaction can be tolerated.
4.Treatment of 57 Headache Cases by Needling Point Xingjian (LR 2) Through Taichong(LR 3)

Needling point Xingjian (LR 2 ) through Taichong (LR 3) was used as main therapy to treat 57 headache cases by syndromic differentiation, which was compared with the treatment of another 60 cases with conventional points. The curative rate was 100% and 91.7% separately in one of the two groups. There was a significant difference in curative effect (P <0.01 ). The effect of the point-through-point method was superior to that of the conventional point method.
5.Observation on Therapeutic Effect in Treatment of Hypoxemic Infantile Encephalopathy by Acupuncture plus Hyperbaric Oxygen

Objective: To investigate the therapeutic effect of acupuncture plus hyperbaric oxygen in treating hypoxemic infantile encephalopathy (HIE). Methods: Fifty-nine HIE children were divided into treatment (33 cases) and control (26 cases) groups. Both groups were treated by basic therapy of removing acidosis, controlling cerebral edema and convulsion, and intravenous drip of cerebrolysin or citicoline. The treatment group was treated by acupuncture plus hyperbaric oxygen and the control group was treated by hyperbaric oxygen only. Results: The total effective rate was 97.0% in the treatment group and 73.1% in the control group. Statistical analysis showed a highly significant difference (P<0.01). Conclusion: Acupuncture plus hyperbaric oxygen was better than simple hyperbaric oxygen in treating HIE. Its main manifestations were shortening the course of disease, increasing cure rate, decreasing death rate and reducing the occurrence of sequelae.HIE children should be treated as early as possible.
6.Application of subaxillary vertical small incision in lung operations

Objective To explore the improvement of traditional incision in thoracotomy and the application of subaxillary vertical small incision in lung operations. Methods We carried out a retrospective analysis on documents of 680 cases of subaxillary vertical small incision pneumonectomy conducted in this hospital between December 1996 and June 2003. Results The length of incision was 8~13 cm (mean, 11 2 cm). The time for thoracotomy was 4 5~10 min (mean, 6 min) and the operation time was 50~ 170 min (mean, 135 min). The intraoperative blood loss was 100~1200 ml (mean, 350 ml), the postoperative drainage volume was 120~800 ml (mean, 300 ml), and the perioperative blood transfusion, 0~1400 ml (mean, 300 ml). Postoperative pain classification results revealed that 585 cases of grade 1, 60 cases of grade 2 and 35 cases of grade 3 severity were observed. The postoperative hospital stay was 10~21 days (mean, 14 days). No surgical death occurred. Conclusions Subaxillary vertical small incision can be simply made. It offers minimal invasion, less blood loss, mild postoperative pain, quick recovery and good cosmetic results, being a viable option in most operations of pneumonectomy.
7.Diagnosis and surgical treatment of pulmonary sclerosing hemangioma: Report of 33 cases

Objective To discuss the diagnosis and surgical management of pulmonary sclerosing hemangioma(PSH).Methods Clinical records of 33 cases of PSH(34 lesions) in this hospital from 1987 to 2002 were reviewed.There were 6 male cases and 27 female cases,with a mean age of 41.3 years(range,24~57 years).Twenty-three cases had an initial symptom of bloody sputum,while 10 cases were disclosed by physical examinations.Image findings revealed solitary mass of the lung in all the cases,in 5 of which a "crescent sign" was presented.Results Preoperatively,28 cases were diagnosed as having pulmonary benign tumors(including 5 cases of suspected sclerosing hemangioma),1 case was clarified as having PSH by percutaneous needle aspiration cytology,and 4 cases were misdiagnosed as having lung cancer. All the patients received surgical treatment,including 21 cases of unilateral pulmonary lobectomy,1 case of bilateral lobectomy,5 cases of segmentectomy,4 cases of wedge resection(including 1 case of bilateral thoracoscopic wedge resection),and 2 cases of tumor excision.The operation time was 45~210 min(mean,90 min),and the blood loss was 100~600 ml(mean,230 ml).No peri-operative deaths were encountered.Follow-up surveys in 33 cases for(3~)10 years(mean,5.5 years) found no recurrence.Conclusions In middle-aged women,if the sign of bloody sputum develops,and image findings indicate the solitary mass of the lung,especially with a "crescent sign",a diagnosis of PSH is possible.Percutaneous needle aspiration cytology is helpful to make a confirmative diagnosis in some patients.It is advisable to manage the disease surgically.
8.COMPARING CHARACTERISTICS OF ETHANOL PRODUCTION BY RESPIRATORY DEFICIENT MUTANTS AND WILD STRAIN IN SACCHROMYCES CEREVISIAE

In order to compare the characteristics of ethano l production, a serie s respiratory deficient mutants (rho -) of Saccharomyces cerevisiae and the ir wil d parent strain (rho +) were assayed for ethanol yield, ethanol fermentation ki netics, ethanol tolerance, and activities of ethanol-dehydrogenase. The data i nd icated that: Though the average amount of ethanol produced by mutants rho - a nd by wild rho + strains were similar, some mutants produced a little more amount of ethanol than wild type. The kinetics curve of ethanol fermentation suggested t h at rho - mutants had a little higher ethanol fermenting speeds than wild type . Th e activity of ethanol-dehydrogenase per unit weight of protein of rho - mutan ts c ells were higher than that of wild strain rho +; the activity of ethanol-dehyd rogenase per unit volume of fermentation medium of rho + were higher than that rho - mutants,mainly due to the higher biomass of rho + were higher than that of rho - mutan ts. Some rho - mutants could tolerate higher ethanol concentration than wild rh o + stra in, but some not. If higher ethanol tolerance and higher ethanol yield rho - mu tants were used in fermenting process,higher ethanol yield would be expected,but some problems such as that rho - mutants grew slowly should be considered.
9.Up-regulating effect of hepatitis C virus core protein on inducible nitric oxide synthase gene promoter

Objective To investigate the transactivating effect of hepatitis C virus(HCV)core protein on inducible nitric oxide synthase(iNOS)gene promoter and the molecular biological mecha- nisms of HCV pathogenesis.Methods Polymerase chain reaction(PCR)technique was employed to amplify the sequence of iNOS promoter by using HepG2 genomic DNA as template,and the product was cloned into pGEM-T vector.The iNOSp gene was cut from T-iNOSp by KpnⅠand XhoⅠ,and then was cloned into pCAT3-Basic,the constructed vector was named as pCAT3-iNOSp,pCAT3-iN- OSp was transfected into the LO_2 cell line.LO_2 cell was also cotransfected with pcDNA3.1(-)-core and pCAT3-iNOSp by FuGENE 6 transfection reagents.The LO_2 cells transfected with pCAT3-Basic was used as negative control.The activity of CAT in LO_2 cells was detected by an ELISA kit after 48 hours,which reflected the transactivating function of HCV core protein to iNOS gene promoter.Re- sults The expressive vector pcDNA3.1(-)-core and report vector pCAT3-iNOSp had been construc- ted and confirmed by restriction enzyme digestion and sequencing.The expression of CAT in LO_2 cells transfected with pCAT3-iNOSp and peDNA3,1(-)-core was 11 times as higher as that of pCAT3-bas- ic,and 6 times as higher as that of pCAT3-iNOSp.Conclusion It is suggested that HCV core protein can transactivate iNOS gene promoter.
