3.Rapid N-glycosylation profiling of glycoprotein based on ultrafiltration-devices.

With development of bio-technique, more and more proteins were applied as clinical approaches. However, the protein homogeneity, especially the N-glycosylation limited the further research and application of these protein drugs. The analysis method for N-glycans is believed to be critical in protein drugs development. To enhance the N-glycans isolation efficiency and accelerate the pretreatment, a new strategy was built on ultrafiltration-devices. New methods increased the isolation efficiency of N-glycans containing N-acetylglucosa mine with 10%-20%. The degrading of N-glycans containing sialic acids was also minimized with this method. 20%-100% more N-glycans with sialic acids were isolated. The pretreatment was finished within 30 min. Coupled with HPLC-HRMS, an effective and reliable strategy designed for protein drugs N-glycans analysis were developed.

4.Effect of cisplatin combined with docetaxel concurrent radiotherapy on advanced esophageal carcinoma and the effect on immune status

Objective To investigate the clinical effect and patients of docetaxel and cisplatin combined with radiotherapy for advanced esophageal carcinoma of the safety and the effect on the immune status of patients. Methods 104 cases of advanced esophageal cancer patients admitted in Dongyang People's Hospital from January 2011 to June 2014 were divided into the control group and the observation group with 52 cases in each group. The two groups of patients were treated with three-dimensional conformal radiotherapy, total dose of 55~65gy, the course of treatment was six weeks, at the same time, the control group were treated with cisplatin (20mg/m2) combined with 5-fluorouracil (400mg/m2) chemotherapy, the observation group were treated with cisplatin (20mg/m2) combined with docetaxel (120mg/m2) chemotherapy, the course of treatment was three weeks. Compared the two groups of patients with clinical treatment effect, survival rate and the average survival time, adverse reaction rate, and detection of two groups of patients before and after the treatment of immune function index CD3+, CD4+, CD8+ and CD4+/CD8+ levels. Results The total effective rate of observation group was 86.53%, the total effective rate of control group was 69.23%, the difference was statistically significant (P< 0.05), the patients in the observation group two years survival rate and mean survival time were significantly better than the control group, the difference was statistically significant (P< 0.05), the adverse reaction rate of the observation group was 59.62%, the adverse reaction rate of the control group was 65.38%, the difference was statistically significant (P< 0.05), two patients immune function indexes of CD3+, CD4+, CD8+ and CD4+/CD8+ levels had no significant difference before treatment after treatment, patients in the observation group, CD4+, CD3+, CD8+and CD4+/CD8+were significantly better than the control group, the difference was statistically significant (P< 0.05). Conclusion Three-dimensional conformal radiotherapy based on cisplatin combined with docetaxel chemotherapy is superior to cisplatin combined with 5-fluorouracil chemotherapy, and less adverse reaction, while patients with advanced esophageal carcinoma immune function less.
5.MiRNA-106a Induces Multidrug Resistance of Gastric Cancer Cells by Targeting RUNX 3

Objective To explore the effect of miRNA‐106a(miR‐106a) expression on multidrug resistance(MDR)of gastric cancer(GC)cells and the involvement of runt‐related transcription factor 3 RUNX3.Methods The expression of miR‐106a was detected in two human gastric adenocarcinoma cell lines with MDR by immunoblotting and apoptosis assay. The sensitivity of GC cells to anticancer drugs was observed by detecting the expression of miR‐106a by using immunoblotting and PCR ,and the relationship between miR‐106a and RUNX3 was determined by luciferase activity assay.Results miR‐106a was significantly in‐creased in GC cells with MDR ,and it suppressed the sensitivity of GC cells to anticancer drugs. It could modulate MDR by tar‐geting RUNX3.Conclusion miR‐106a can induce the MDR by targeting RUNX3 in GC.
6.Effects of caffeine citrate on apnea and its effect on endothelin, nitric oxide and β-endorphin in premature infants

Objective To analyze the clinical effect of caffeine citrate on apnea and its effect on endothelin, nitric oxide and β-endorphin in premature infants. Methods 118 children with apnea-hypoprenalia were selected and divided into experimental group and control group according to different treatment methods. The control group were treated with aminophylline, and the experimental group were treated with caffeine citrate. The clinical symptoms, endotheliolysin, nitric oxide and β-endorphin in two groups of patients were compared. Results The total effective rate of 84.75% (50/59) in the experimental group was higher than 69.49% (41/59) in the control group(P<0.05). The incidence of abdominal distension, heart rate, feeding intolerance, bronchopulmonary dysplasia and irritability were significantly lower in the experimental group than those in the control group (P <0.05) . After treatment, the levels of endotheliolysin, nitric oxide in the experimental group were significantly higher than those in the control group (P <0.05), and the level of β endorphin in the experimental group was significantly lower than that in the control group (P <0.05). Conclusion Caffeine citrate has a significant clinical effect on apnea in premature infants, and can reduce the incidence of adverse reactions.
7.Behavior change of adriamycin resistant human breast cancer cell line MCF7

Objective To investigate mechanisms of resistance to adriamycin(ADR)by human breast cancer cell line MCF-7 and to find the alteration of features and celluar behavior of MCF-7 after exposure to ADR.MethodsProliferation speed,population doubling time of MCF(wild type),MCF-7/ADR(exposure to adriamycin)and withdrawl group were respectively tested.Cell phenotype alteration was detected using SP immunohiatochemistry methods.Results No significant difference of proliferation speed was found between MCF-7/ADR and MCF cells.As exposure time prolonged,withdrawl group cells grew faster,thus population doubling time shortened.Differentiation of MCF-7/ADR and wthdrawl group was lower than wild group.The expression of drug resistance associated marker of MCF-7/ADR such as Pgp,LRP,GST-pi,TOPOⅡwas higher than that of MCF-7,ER turned to express negatively,and expression of PR gradually decreased as exposure continued.Conclusion MCF-7 cells exposed to ADR got drug resistant,their proliferation was not suppressed by withdrawl of ADR and even grew faster.Drug resistant cells gained dedifferentiation ability.Their heredity and biochemistry features changed,expression of target enzyme also altered and was reversible by drug withdrawl.
