BACKGROUND: Numerous studies have shown that nuclear factor-KB (NF-kB) was positively correlated with vascular endothelial growth factor (VEGF) in the cerebral hypoxia region. Thus, we assumed whether NF-kB in the pathway of hypoxia-inducible factor-1 α (HIF-1α) upregulating VEGF and plays a bridge effect.OBJECTIVE: To study the effects of NF-kB in HIF-1α pathway of increasing expression of VEGF using HIF-1α-modified neural stem cells (NSCs) as vectors.DESIGN, TIME AND SETTING: In vitro observation of cytology was conducted at the Neuroscience Institute, Jiamusi University from March to December 2008.MATERIALS: Wistar rats aged less than 24 hours of both genders were used.METHODS: NSCs were transfected after amplification of adenovirus vector HIF-1α (AdHIF-ia)-green fluorescent protein (GFP).Fluorescence detection was used to determine HIF-1α-GFP and blank vector Ad-GFP expression in NSCs. Protein was extracted from transfected NSCs, blank vector NSCs and normal NSCs, separately. Subsequently, NF-kB specific inhibitor pyrrolidine dithiocarbamate (PDTC) (50,150, 300 μmol/L) was added in HIF-1α-modified NSCs. Western blot analysis was used to determine changes in VEGF expression.MAIN OUTCOME MEASURES: The following parameters were measured: expression of HIF-1α, VEGF and NF-kB in NSCs in each group; VEGF expression in NSCs following treatment of NF-kB specific inhibitor.RESULTS: Gene expression was associated with MOI and transfected time following AdHIF-1α-GFP transfected with NSCs. After transfected AdHIF-1α-GFP in NSCs, HIF-1α, VEGF and NF-kB expression was positively correlated. Expression of VEGF was reduced in AdHIF-1α-GFP-modified NSCs following treatment of NF-kB inhibitor PDTC in a concentration-dependent fashion. There were significant differences in VEGF expression between each concentration group (P < 0.05-0.01).CONCLUSION: NF-kB in signaling pathway of HIF-1α-upregulated VEGF expression and played a bridging effect.

Objective To investigate the sensitivity and accuracy of monitoring oxygen saturation of arterial blood flowing through descending aorta with a probe placed in the lower segment of esophagus ( SeO2). Methods This study was approved by the ethics committee of our hospital and written consent was obtained. Forty-five ASA Ⅰ-Ⅲ patients (34 males, 11 females) aged 14-79, weighing 40-90 kg undergoing elective (36 patients) or emergency (9 patients) surgery were studied. After induction of anesthesia a probe we made (patent number ZL 200320115080.2) was placed in the lower segment of esophagus and connected to a monitor (Philips-150A) . The depth of insertion and the direction of the probe were adjusted until the same waveform as SpO2 was obtained. Once the best waveform was obtained, 10-15 min stabilization was allowed. The ventilator was then disconnected. When SeO, decreased from 100% to 90% mechanical ventilation was started again. The following parameters were recorded:(1) the onset time (the time from disconnection of ventilator to the time when SeO2 and SpO2 started to decrease), (2) the time needed for SeO2 and SpO2 to decrease from 100% to 95% (T95) and 90% (T90), and (3) the recovery time (the time needed for SeO2 and SpDO2 to return to 100% after mechanical ventilation was started again. Arterial blood samples were taken for blood gas analysis when Se02 and SpO2 were 90% , 95% and 100% . Results SeO2 , SpO, and SaO2 were 100% when the patients were being ventilated with 100% oxygen. When the ventilator was disconnected SeO, dropped much faster than SpO2. It took (286?15)s for SeO2 to drop from 100% to 95% while for SpO2 it took (398 ?28)s. The most readings of SeO2 were in the range of 2 standard deviation from SaO2. SeO2 correlated well with SaO2 and SpO2. r was 0.9754 and 0.9656 respectively(P

Objective To determine the influence of ginsenoside Rg1 on angiogenesis and the expression of vascular endothelial growth factor (VEGF) and hypoxia inducible factor-1? (HIF-1?) in rat acute myocardium ischemia (AMI) in rat. Methods A murine model of AMI was reproduced. Wistar rats (n=104) were randomly divided into shamoperation group, AMI control group, low ginsenoside Rg1 group (1mg/kg) and high ginsenoside Rg1 group (5mg/kg). Microvascular density, VEGF protein, VEGF and HIF-1? mRNA were assessed at 3, 7, 10 and 14 days after operation. Results The values of VEGF and HIF-1? mRNA were increasing with the duration of ischemia and hypoxia, and positive relationship was found between the time of AMI and the expression of HIF-1? mRNA. Ginsenoside Rg1 enhanced the formation of angiogenesis (P

Objective To study the clinical appilication of detachable balloon embolization techenique in carotid cavernous fistula. Methods 27 cases consisted of 26 cases of typical TCCF and 1 case of CCF caused by left anterio cerebral artery(A1) aneurysm ruptured . 27 cases were treated by endovascular embolization with detachable balloon. Transcranial Doppler ultrasound(TCD) was used before and after embolization. All the patients were followed-up for more than 6 months. Results 22 cases of TCCF were cured by detachable embolization and their internal carotid artery(ICA) remained patent. The first segement of left anteriol cerebral artery was embolized by two detachable balloons in the patient with CCF caused by ruptured aneurysm. Internal carotid artery was embolized by two balloons in 4 cases. There was no severe complication in this group. Conclusions Detachable balloon embolization is a safe and effective method for internal carotid cavernous fistula.

To standardize the harvest ways of Dendrobium officinale and improve the quality and yield of D. officinale, a field experiment was carried out to study the effect of two kinds of harvest ways, which were keeping some of the axial shoot and harvesting all of the shoot by the end of the year. Then, the agronomic traits and yield were measured and the contents of polysaccharides and extractum were determined. The results showed that the harvest ways significantly affected the growth of D. officinale. Keeping some of the axial shoot could significantly improved the number of sprout, stem length, internode number and the internodal length, which also triggered increase the weight of fresh stems, leaves and the total of them and dry stems in per unit area, but it could not promote the stem diameter and the polysaccharide content in stems. Keeping some of the axial shoot moderately was conducive to the improvement of the production of medicinal materials in the process of harvesting by promoting the germination and growth of new buds, and to ensure the polysaccharide content by regulating the illumination and the density of cultivation.

Objective To study the characteristics of its pathologic and neurophysiologic changes of a ani-mal model of cervical spinal cord compression, and to explore the relationship between the severity of spinal cord inju-ry and its functions. Methods Thirty-two rabbits were divided into 4 groups randomly. A catheter was inserted intothe vertebral canal and a balloon was inflated to compress spinal cord in epidural space. According to the diameter ofballoon, the animals were divided into 4 groups (A, B, C, D group), Each group consisted of 8 rabbits. The corti-cal somatosensory evoked potential (CSEP) and motor evoked potential (MEP) were used to assess neurologicalfunction. Motor function behavior was scored before and on 1, 3, 7 and 14 days after surgery. Histological observa-tions were performed, pathological changes were observed by light and electron microscope. Results Spinal cordcompression resulted in a gradual increase of the peak latency and significant decrease of the peak amplitude. The la-tency and amplitude of MEP were changed more dramatically than those of CSEP. The result indicated that MEP wasmore sensitive than CSEP to the compression. Analysis also revealed that the severer the pathologic changes, the lon-ger the latency and the lower the amplitude of the evoked potentials. Conclusion The animal model presented inthis paper was simple and standardized. Evoked potentials as a noninvasive technique have great value in monitoringspinal cord function. The variance of amplitude and the latency in significantly correlate with the degree of compres-sion of the spinal cord.
