2.Effect of α-MSH on osteoclast formation

Raw264.7 cells were incubated with receptor activator of NF-kappa B ligand (RANKL) and α-melanocyte stimulating hormone(α-MSH) for6 d.The amount of osteoclast cells were counted by tartrate resistant acid phosphatase staining and the acid phosphatase activity was assayed.The expressions of 5 melanocortin receptors (MCR) in Raw264.7 cells were determined by RT-PCR.The results showed that the number of osteoclasts in RANKL +α-MSH group was significantly increased compared with RANKL group (P < 0.05),but there was no osteoclast formation in α-MSH group.Compared with control group and α-MSH group,the acid phosphatase activities were significantly increased in RANKL group and α-MSH+RANKL group (P<0.05).All five MCRs were expressed in the Raw264.7 cells shown by RT-PCR.These results suggest that α-MSH may promote osteoclasts formation through RANK signaling pathway.
3.Application and nursing of the high-pressure injector on advancement CT scanning

Objective To explore the application and nursing of the high-pressure injector on advancement CT scanning.Methods Using non-ionotropic contrast medium to analyze 3258 patients with different constitutions and puncture methods retrospectively.Results In the process of advancement CT scanning, the high-pressure injector was used according to patients practical needs, and plant parameter and controlling dosages and flow velocity were adjusted to obtain good picture for clinical diagnosis, what's more, incidences like phlebitis, air embolism and cadio-vascular diseases don't took place.Conclusions The proper use of high-pressure injector in the process of advancement CT inspection and clinical inspections can obtain satisfactory effect and prevent side effects.
4.Ion chromatography for determination of ammonia sampled by silica gel in air of workplace.

OBJECTIVETo establish an ion chromatography (IC) method for determination of ammonia in air of workplace.
METHODSAmmonia in workplace air was collected in silica gel tube, desorbed with 10 mmol/L methanesulfonic acid by ultrasonic for 10 min, determined by IC.
RESULTSThe linearity range was 0.02-1.00 microg/ml. The linear equation was Y = 12041X-187 (r = 0.9997). The limit of quantification was 0.13 mg/m3 (the air volume was 1.5 L). Collection efficiency was 100%. Extraction efficiency was 99%. The relative standard deviation was 4.2%-6.3%. The penetration capacity was more than 264 microg. Sample could be stored for 14 days at least by ambient storage.
CONCLUSIONThis method is convenient, applicable and sensitive, suitable to determinate ammonia in air of workplace.

5.Study on accumulation of polysaccharides and alcohol-soluble extracts contents of Dendrobium officinale leaves.

This paper revealed the accumulation regularity of polysaccharides and alcohol-soluble extracts contents of Dendrobium officinale leaves, which have provided basis for the development and utilization of the leaves. The polysaccharides and alcohol-soluble extracts contents of three D. officinale strains leaves collected in different growing periods were determined by phenol-sulfric acid method and hot-dip method respectively. The results showed that the content of polysaccharides in leaves was 4.45% -12.17%, and was about a quarter in stems. The alcohol-soluble extracts content in leaves was 7.45% - 29.34%, and was 1.5 times that of stems. The quality variation of polysaccharides in leaves was closely related to the phenophase. The leaves with lower level of metabolism in three stages: winter, early germination stage and deciduous period, which led to lower content of polysaccharides. The leaves at the vigorous growth stage with higher content of polysaccharides. The alcohol-soluble extracts were closely associated to the formation and germination of buds. The content of alcohol-soluble extracts peaked before sprout, and promoted the growth of new shoots.

6.Application of CEUS quantitative analysis in diagnosis of renal tumors

8.Affection of family cognition of breasffeeding on primiparas' breast feeding behaviors

Objective To describe family cognition of breastfeeding and primiparas' breastfeeding behaviors 4 months postpartum and to explore the relationship between family cognition and new mother' breastfeeding behaviors. Methods A cross-sectional questionnaire survey was conducted to 200 primiparas at outpatient clinic in Beijing in terms of their breastfeeding behaviors and family cognition of breastfeeding. Results The rote of entire breastfeeding was 47.0% ;and the score of family cognition was (23. 13 ±3. 19), which was at a moderate level. The total score of family cognition of primiparas who gave entire breastfeeding was significantly higher than those gave mixed or artificial feeding (F=8.26, df=2, P<0. 01). Conclusions The breasffeeding rate in primiparas was relatively low,primipaaras who insists on giving entire breastfeeding have high level of family cognition. To help people form correct and active family cognition of breastfeeding and improve breastfeeding behavior in primiparas, health education should be launched toward not only primiparas but also their family numbers, especially the main caregiver.

10.Effect of Curcuma aromatica on epithelial-myofibroblast transdifferentiation of NRK-52E cells induced by TGF-beta1.

OBJECTIVETo observe the antagonist effect of Curcuma Aromatica (CA) on renal tubular epithelial-myofibroblast transdifferentiation (EMT) induced by transforming growth factor-beta1 (TGF-beta1).
METHODSNormal renal tubular epithelial NRK-52E cells in vitro cultured were randomly divided into 6 groups, i.e., the normal control group (Group C), the TGF-beta1 induced model group (Group T), the low dose CA treated group (Group E1), the moderate dose CA treated group (Group E2), the high dose CA group (Group E3), and the Benazepril Hydrochloride Tablet treated group (Group Y). Except Group C, corresponding medication (with an action of 48 h) was administered to cells in the rest groups after they were induced by TGF-beta1 for 24 h. The morphological changes were observed by inverted phase contrast microscope. The distribution of beta-actin protein was detected by immunohistochemical assay. The mRNA expressions of alpha-smooth muscle actin (alpha-SMA) and E-cadherin (E-cad) were detected by real-time PCR. The concentration of fibronectin (FN) was detected by ELISA.
RESULTSAfter induced by TGF-beta1 for three days, hypertrophy and elongated cells in fusiform-shape occurred,with increased expressions of beta-actin protein in the cytoskeletal structure (P < 0.05), bundle fibrous structure occurred inside cytoplasm with significantly up-regulated intracellular alpha-SMA mRNA expressions (P < 0.05), E-cad mRNA expression decreased (P < 0.05), the FN content in the supernate increased (P < 0.05) in Group T. Compared with Group T, partial cells in Group E1, E2, and E3 showed fibrous changes, accompanied with decreased expression of beta-actin protein and FN concentration (P < 0.05). The expression of alpha-SMA mRNA increased and the expression E-cad mRNA decreased in Group E2 and E3 (both P < 0.05). But there was no statistical difference in the expression levels of E-cad and alpha-SMA mRNA (P > 0.05). Compared with Group E1, the expression of beta-actin protein and FN concentration decreased in Group E2 and E3 (P < 0.05). The expressions of alpha-SMA mRNA decreased and E-cad mRNA increased in Group E3 (P < 0.05). Compared with Group Y, the expression of beta-actin mRNA and FN concentration increased in Group E1 (P < 0.05); the expression of beta-actin mRNA increased in Group E3 (P < 0.05); the expression of E-cad mRNA decreased in Group E3 (P < 0.05). There was no statistical difference in the expression of alpha-SMA mRNA among Group E1, E2, and E3 (P > 0.05).
CONCLUSIONCA could inhibit the occurrence of TGF-beta1 induced EMT, which could be used as an effective drug for treating chronic renal insufficiency.
