2.Egr-1 mediates Si0(2)-driven transcription of membrane type I matrix metalloproteinase in macrophages.

The up-regulation mechanism of membrane type I matrix metalloproteinase (MT1-MMP) in macrophages stimulated by silica in vitro and the contribution of early growth response 1 (Egr-1) transcription factor in the gene expression pathway were investigated. Macrophages stimulated by silica were treated with Egr-1 antibody or Egr-1 decoy oligodeoxynucleotides (ODN). The levels of MT1-MMP proteins were determined by Western blot and the expression of MT1-MMP mRNAs was detected by RT-PCR. The results showed as compared with control macrophages, silica-stimulated group showed up-regulated gene expression of MT1-MMP via Egr-1 (P<0.01). Compared with silica-stimulated macrophages untreated with antibody, the cells treated with 5 microg/mL Egr-1 antibody were associated with reduced expression of MT1-MMP protein (P<0.01) and mRNA (P<0.01). Compared with silica-stimulated untransfected group, the Egr-1 "decoy" ODN group was associated with reduction in the expression of MT1-MMP protein and mRNA (P<0.01). It was concluded gene expression of MT1-MMP which may play a critical role in silicosis was up-regulated by silica in macrophages. Egr-1 participated in the expression of MT1-MMP and positively regulated the expression. Both Egr-1 antibody and Egr-1 decoy ODN suppressed the expression of MT1-MMP through the Egr-1 pathway and may become a potential therapeutic tool in the management of silicosis in the future.

4.Effect of Hydrogen Peroxide on Microstructure of Mice Kidney

Objective To study the effect of hydrogen peroxide on microstructure of mice kidney and discuss the toxic effect on mice kidney.Methods Thirty healthy male mice of Kunming Genus were divided into 3 groups at random:control group and two experimental groups. Running water was fed to control group for 10 days while 0.3,3 g/L hydrogen peroxide running water readily prepared was fed to the experimental groups for 10 days. On the 10th day,the kidneys were taken out,and fixed in the fixation solutions,conventionally produced and stained.Finally,they were studied under the optical microscope.Results Experimental groups:in the kidney tissue cytoplasm of proximal convoluted tubule showed hydropic degeneration and vacuolation which depend on dose of hydrogen peroxide.Conclusion Toxic effect on mice kidney can be caused by hydrogen peroxide.
5.Effect of moxibustion at sanyinjiao (SP 6) on the process of childbirth: a randomized controlled study.

OBJECTIVETo explore the effect of moxibustion at Sanyinjiao (SP 6) on normal process of childbirth and safety of mothers and infants.
METHODSOne hundred and seventy-four cases of singleton primiparas, fetal head position were selected as the subjects and divided into three groups by single-blind randomized method: an acupoint group (n = 59), a non-acupoint group (n = 57) and a blank group (n = 58). When the cervix was dilated by 2 to 3 cm, DAJ-23 type multi-function moxibustion apparatus was used at Sanyinjiao (SP 6) in acupoint group, and at non-acupoint spot (the midpoint from the radial arterial pulse throbbing place on the radial side of the transverse crease of the wrist to the radial end of biceps tendon of the elbow) in non-acupoint group, 30 min for each one. The conventional labor care was adopted in blank group. The differences were observed in the first active stage, the second stage and the third stage of the labor as well as the safety of mothers and infants among three groups.
RESULTSThe durations of the first active stage of labor [(123.05 +/- 96.401) min] and the second stage [(37.407 +/- 22.390) min] in acupoint group were shorter than those [(171.40 +/- 94.339) min, (49.517 +/- 25.373) min] in non-acupoint group, as well as those [(178.79 +/- 97.231) min, (53.684 +/- 20.087) min] in blank group separately (all P < 0.05). It was discovered that the progressions of the first active stage and the second stage of labor were accelerated in acupoint group, and there was no statistically significant difference in the third stage of labor among the three groups (all P > 0.05). Bleeding amount after childbirth in acupoint group was less than those in non-acupoint group and blank group (both P < 0.05), and Apgar scores of newborns in acupoint group and non-acupoint group were higher than that in blank group (both P < 0.05).
CONCLUSIONMoxibustion at Sanyinjiao (SP 6) can shorten the process of childbirth, reduce postpartum bleeding and has no harmful impact on mothers and infants.

6.Blood-enriching Effects of Different Processed Radix Polygoni Multiflori Slices on Blood Deficiency Rats

This study was aimed to investigate the blood-enriching effect of processed Radix Polygoni Multiflori on blood deficiency rats. A total of 140 SD rats were randomly divided into 14 groups, which were the blank control group, blood deficiency model group, positive control group and 11 test groups. The blank control group and model control group were treated with distilled water and the positive control group was given X ue b ao Buxue soluble granules. The test groups were treated with 11 different kinds of processed Radix Polygoni Multiflori sam-ples at a dosage of 2 g?kg-1 respectively. All rats were given drugs once a day. After 10 days, blood samples were collected and the amount of red blood cell (RBC), hemoglobin (Hb), haematocrit (HCT), whole blood viscos-ity and plasma viscosity, prothrombin time were estimated. The results showed that 9 processed Radix Polygoni Multiflori groups can increase the number of RBC and Hb significantly ( P < 0 . 05 or P < 0 . 01 ) in blood deficien-cy rat models. They were the sample processed with decoction of black soybean (DBS) for 24 h, with wine and DBS combination for 24 h , steamed for 4 h and 8 h under high pressure , with DBS for 6 h and 8 h , with wine for 6 h, with wine and DBS combination under high pressure, respectively. The samples processed with DBS un-der high pressure for 8 h increased RBC and Hb were more obviously ( P < 0 . 01 ) . The 8 groups can improve the situation of functional disorder of blood rheology significantly ( P < 0 . 05 or P < 0 . 01 ) . They were the unprocessed samples, processed with DBS and wine for 24 h respectively, with DBS and wine combination for 24 h, steamed for 4 h and 8 h or steamed with DBS for 6 h and 8 h or with wine for 6 h under high pressure respectively. It was concluded that the blood-enriching effect was improved while the R adix Polygoni Multiflori was processed with DBS or wine and under high pressure for 6-8 h .
7.Arthroscopically minimal invasion asisted percutaneous cannulated screw fixation of longitudinal fracture of the patella

Objective To evaluate the effectiveness of arthroscopically assisted percutaneous reduction and internal fixation with eannulated screws.Methods The fracture was reduced by closed manipulation or percutaneous leverage force by using the Kirschner wire.Then the patella was temporarily fixed by using a large size towel clamp or Kirschner wires.Under the guidance of knee arthroscopy,a micro-incision was made at the size of cannulated screw placement,the pilot holes were drilled at a proper depth,and the thread was configured.Two titanium screws were inserted parallelly.Results Following-up chekups for 4~24 months in 18 cases showed a satisfactory recovery of knee functions.According to the Bostman' standard,excellent effects were obtained in 16 cases and good effects in 2 cases.Conclusion Treatment of patellar fractures by percutaneous cannulated screw fixation under arrhroscope of- fered advantages of minimal invasion,accurate reduction,reliable fixation,and excellent recovery of joint functions.
8.Study on the ecological association between natural radioactivity and childhood leukemia in Guangdong province

Objective This study was undertaken to evaluate the ecological association between terrestrial natural radionuclide,indoor radon concentration,natural radioactivity and leukemia incidence among children under 18 years of age.Methods Data were gathered from the disease surveillance program and literature reading while software SPSS 13.0 was used to calculate the Spearman's correlation.Results The incidence rates of childhood(0-18 year)leukemia showed significant differences in different places with the highest as 3.13/105in Jiangmen area and the lowest as 0.42/105 in Maoming area.The incidence in Jiangmen was 7.45 times higher than that in Maoming.There was a rank correlation between the incidence of childhood leukemia and the mean concentrations of natural radio-nuclides in soll(226Ra and 232Th),with a Positive correlation observed for overall leukemia(rs=0.70,P=0.011;rs=0.66,P=0.02 for226 Raand 232Th respectively)and acute lymphoblastic leukemia(ALL)(rs=0.66,P=0.019;rs=0.64,P=0.025 for 226 Ra and 232Th respectively).Associations between the incidence of childhood leukemia and the indoor γ radiation dose rate,the total annual average effective dose equivalent from natural background radiation were also analyzed(both rs=0.59,P=0.042).Conclusion The natural radioactivity was likely to be a causative factor for childhood leukemia in Guangdong.
9.Expression of caudal related homeodomain transcription 2 in colon cancer and its relationship with metas-tasis

Objective To study the expression of caudal related homeodomain transcription 2 ( CDX2 ) protein in human colon cancer , and study the relationship between protein expression and colon cancer metasta -sis.Methods With immunohistochemical technique in peroxidase notation , pathological specimens of 80 cases of colon cancer were selected , including 47 cases of distal metastasis , and 33 cases without cancer metastasis . CDX2 protein expression in cancer tissue and the related data were statistically analyzed .Results Patients'sex, age and location of colon tumors had no statistical relation with CDX 2 protein expression(P>0.05).CDX2 pro-tein was statistically different in well-differentiated(100.00%), moderately-differentiated(80.00%), and poor-ly-differentiated(33.33%) tumor tissues.The difference had statistical significance (P<0.05), The expression of CDX2 protein reduced with the reduction in the degree of colorectal adenocarcinoma differentiation , rs =0.217, indicating that CDX2 protein expression was positively correlated with the malignant degree of colon canc -er.Positive CDX2 protein expression was significantly lower in metastatic colon cancer tissues (72.3%) than in non-metastatic tissues ( 90.9%) , and the difference had statistical significance .Lymph node metastasis could down regulate CDX2 protein expression in colorectal cancer .Conclusion CDX2 protein is positively correlated with the differentiation degree of colon cancer and distal metastasis can down regulate CDX 2 protein expression .
10.Repair of rabbit knee articular cartilage defect by the injectable chitosan/beta-glycerophosphate gel encapsulating allograft chondrocytes and the intervention of Weilingxian

BACKGROUND: At present, studies on repair of cartilage defect have been focused on tissue engineering technique. Growth factors are one of the most important parts. However, the effect and security of growth factors have not been confirmed. Studies have shown that Weilingxian can maintain and promote the synthesis of proteoglycan, collagen Ⅱof chondrocyte, and it also can promote proliferation of chondrocyte and expression of transforming growth factor (TGF)-β1 mRNA.OBJEGTIVE: To investigate the feasibility of injectable chitosan/β-glycerol phosphate (C/β-GP) encapsulating allograft chondrocytes on the repair of articular cartilage defects and the intervention effect and possible mechanisms of Weilingxian.METHODS: A 0.4-mm defect was established on knee articular cartilage. Expeirmental New Zealand rabbits were randomly divided into three groups: Weilingxian, common culture media, and model groups. In the common culture media group, the samples were treated with C/β-GP and chondrocyte suspension (1 mL); at 2 days after gel injection, Weilingxian or common culture media (1 mL) were respectively given into joint cavity, once a day, for 7 successive days. The samples in the model group were not treated. Gross, histological (HE staining, TB staining), type Ⅱ collagen immunohistochemical, and Wakitani score examinations were performed on 6 and 12 weeks after surgery.RESULTS AND CONCLUSION: Defects of articular surface were well filled in Weilingxian and common culture media groups, and hyaline cartilage-like structure was formed. The surface flatness and degree of integration with surrounding tissue of Weilingxian group was better than common culture media group. Formation of cartilage-like and secretion of cartilage matrix and specificity of collagen type Ⅱ were found in histological slices. Defects in the model group were not repaired, while tissue proliferative degeneration was observed. Integration of.repair tissue with surrounding tissue, histology and amount of type Ⅱ collagen secretion in Weilingxian group were better than common culture media group. Wakitani scores of Weilingxian group and common culture media group were significantly lower than model group (P < 0.01), andscores of Weilingxian group was significantly lower than common culture media group (P<0.05). Injectable chitosan/β-glycerolphosphate gel encapsulating allograft chondrocytes could repair articular cartilage defects, and Weilingxian was able to promote the process of it, this manifested the role like growth factor in tissue engineering technique repairing articular cartilage defects.
