1.The role of mitochondrial permeability transition pore in the occurrence of septic myocardial depression

Sepsis is defined as life-threatening organ dysfunction caused by a dys-regulated host response to infection and septic myocardial depression (SMD) is a common complication. Pathogenesis of SMD is complicated and there is lack of specific treatment. Mitochondrial damage is an important pathological basis of SMD, and mitochondrial permeability transition pore (MPTP) plays an important role in maintaining the normal structure and function of the mitochondria. The change of MPTP during sepsis is summarized in this review so as to reveal the significant mechanism of MPTP in the occurrence of SMD.
2.Chromatography fingerprint of the constituents of Melia toosendan Sieb

AIM:To establish the chromatography fingerprint of the constituents of Melia toosendan Sieb from different areas in order to provide a base for the identification of its quality. METHODS: A gradient separated method was applied. Column:Inertsil ODS-3 C_ 18 ,mobile phase:acetonitrile-water,detection wavelength:270 nm,flow rate:1.0 mL/min,column temperature:25 ℃. RESULTS: To establish the chromatography fingerprint of the constituents of Melia toosendan Sieb., make the technical parameters for its quality controlling,and mark 39 main peaks as its characteristic fingerprint. CONCLUSION: The distribution of constituents of Melia toosendan Sieb differ a little from different areas, but the propotion of the constituents differ greatly, with the Melia toosendan Sieb.from the same area , the distribution and propotion differ a little. This method is reproducible, simple and easy, and can be use to provide a base for the quality control of Melia toosendan Sieb.

Objective This study examined the effects of Midazolum-ketamine oral solution (MKOS) on the expression of N-methyl-D-aspartate receptor 1 (NMDAR1) and gamma-aminobutyric acid (GABA)A receptors (GABAAR) mRNA in the cerebral cortex of rat, in order to investigate the sedation mechanism of MKOS. Methods Fifty Sprague-Dawley(SD) rats were divided into ten groups according to the observed time after MKOS administration (0,5,10, 15,30,60,90,120,240 and 360 minutes, n =5 each). The 0 minute group(control group) received 0.9% saline instead. Immunohistochemical staining and in situ hybridization were used to detect the expressions of NMDAR1 and GABAAR mRNA in the cerebral cortex. Results Both GABAAR and NMDAR1 all expressed in the glial cells of cerebral cortex. The expression of NMDAR1 in control group was strong. The expression of NMDAR1 became weaker during 15 to 90 minutes after administration of MKOS (P<0.05). The expression of GABAAR mRNA in control group was weaker,while became stronger during 30 to 90 minutes after administration of MKOS (P <0. 05). Conclusion MKOS may play sedation by strengthening the expression of GABAAR and suppressing the expression of NMDAR1 in the cerebral cortex.

6.CT imaging characteristics and comparative analysis of parotid pleomorphic adeonoma and adenolymphoma

Objective To investigate CT image features of pleomorphic adenoma and adenolymphoma,and to improve the diagno-sis accuracy.Methods CT data of 1 3 cases of pleomorphic adenomas and 9 cases of adenolymphomas confirmed by pathology were retrospectively analyzed and compared.Results All 1 3 cases of pleomorphic adeonoma were solitary lesions,in which 1 1 cases were located in the non posterior inferior quadrant,and 1 1 cases showed persistent or delayed enhancement,with “slowly forward and slowly quit”enhancement performance.In all of the 9 cases of adenolymphoma,3 cases were multiple lesions.In total of 1 3 lesions, 9 lesions were located in the posterior inferior quadrant,and 9 lesions were significantly enhanced in the arterial phase and enhance-ment was decreased in the delayed phase,which showed “quickly forward and quickly quit”.Conclusion CT image features of pleo-morphic adenoma and adenolymphoma are different.Most of lesions can be diagnosed correctly before surgery by using image fea-tures with clinical information.
7.The mechanism of Er-xian Decoction in regulating lipid metabolism disorders on bilateral ovariectomized rats based on metabolomics study

Studies have shown that women's menopause caused by permanent cessation of ovarian function is closely related to lipid metabolism disorders. Er-xian Decoction has been used in the clinical treatment for gynecological diseases and has a good effect on diseases related to reduced sex hormone function. In this study, metabolomics was performed on bilateral ovariectomized model rats within 12 weeks after modeling to mimic the physiological state of menopausal women in different menopausal stages and Er-xian Decoction dosed model rats. The results of liver oil red O staining sections showed lipid metabolic disorder of bilateral ovariectomized model rats and the regulating effects of Er-xian Decoction. 46 potential biomarkers (6 steroid hormones, 3 sphingolipids, 11 phospholipids and 26 glycerides) in plasma and 32 potential biomarkers (1 steroid hormones, 20 phospholipids and 11 glycerides) in liver were obtained based on lipidomics analysis. Then, we analyzed the differential metabolic pathways and construct the lipid metabolism network significantly regulated by Er-xian Decoction. The results provided valuable information for in-depth understanding of the gradual changes on lipid metabolism disorders under menopausal conditions and the characteristics and mechanisms of compound Er-xian Decoction's regulatory effects. The study complied with the procedures established by the Animal Experiment Ethics Committee of the Institute of Materia Medica, Chinese Academy of Medical Sciences and passed the animal experiment ethics examine (No. 00000918).

9.Expression and significance of matrix metalloproteinase 2 and its tissue inhibitor in human endometrial carcinoma

Objective To study the express of matrix metalloproteinase(MMP)-2 and tissue inhibitor of metalloproteinase(TIMP)-2 protein in human endometrial carcinoma. Methods The Immunohistochemistry was used to measure the MMP-2, TIMP-2 in endometrial carcinoma tissue of 40 patients, the atypical hyperplasia of 12 samples and the normal endometrium of 12 samples. Results The positive express of MMP-2 and TIMP-2 in the normal endometrium , atypical hyperplasia and endometrial carcinoma was increased gradually. In endometrial carcinoma, with progression of clinical stage and decrease of histological grade as well as deepening of cancer invasion, the ratio of MMP-2/TIMP-2 was increased. Conclusions The expression of MMP-2 and TIMP-2 were connected with endometrial carcinoma. The imbalance of MMP-2/TIMP-2 in the endometrial cancer might be one of the pathogenesis and development.
10.The influence of low-dose aspirin on the platelet aggregation in the elderly patients with ischemic cerebrovascular diseases

Objective To observe the efficacy of low dose aspirin(ASA) on the platelet aggregation in aged patients with ischemic cerebrovascular diseases(ICVD). Methods Platelet aggregation by arachidonic acid(AA, 500 ?g/ml), adenosine diphosphate (ADP,5 ?mol/L), epinephrine(EPI, 5 ?mol/L), and collagen (COL,2 ?g/ml) using Born's method was induced in 70 elderly with ICVD treated with ASA (40 mg/d) and in 30 untreated patients as well as in 50 healthy controls. Results A high coefficient of variation of the maximum aggregation rate induced by AA, COL and EPI were observed in the ASA (40 mg/d) treated group. Twenty-five out of the 70 patients in the ASA treated group (40 mg/d) were found that their platelet aggregation induced by AA were not markedly inhibited. Ten patients were randomly selected from the above 25 patients to receive an increasing dose to 80 mg/d. At repeated tests, 8 of these 10 achieved marked inhibition, and 2 of the 10 who showed no inhibition continued receiving an increasing dose to 100 mg/d and then achieved marked inhibition. Conclusions There is a great individual variation in response to low dose aspirin in aged patients with ICVD, and the laboratory evaluation of aspirin efficacy and its individualization is necessary in clinical practice.
