1.Clinical research of thalidomide in corporation with NP in treatment of Ⅲ/Ⅳ lung cancer

Objective To study the curative effect and side effect of thalidomide incorporation with NP in treatment of Ⅲ/Ⅳ lung cancer. Methods 36 lung cancer patients were randomly divided into three groups. The patients in experimental group were treated by NP plus thalidomide while without thalidomide in the control group. The difference between the first experimental group and second group is that the above used DDP followed with lobaplatin. The beginning dose of thalidomide was 100 mg/d, increase 50 mg/d every week till 400 mg/d, maintain for at least three months. Results The first experimental group had 4 partial relief cases (34.0 %), 5 improved cases(33 %), total efficacy rate was 34.4 %(4/15), clinical benefice rate 49 %(7/ 15), and the second group was 38 %(4/11), 27 %(3/11), 38 %(4/11), 55 %(6/11). All without significant difference. Conclusion It would be valuable to do clinical research further and widespread popularization study for evaluation of Thalidomide incorporation with NP in treatment of Ⅲ/Ⅳ lung cancer.

In this paper, the content of moisture, ethanol-soluble extractives, total saponins and polysaccharide of different tuber samples of Hemsleya zhejiangensis, from different localities, years and seasons, were detected based upon Chinese Pharmacopoeia 2010 version. The samples of roots, stems and leaves in summer were detected as well. The results are mainly as follows. (1)With tuber quality increasing, the content of total saponins increased and then decreased. The individual quality of tubers getting 594.06 g, the content of total saponins reached the peak. (2) The content of active ingredients in different localities was significantly different, and the population of Wuyanling had the maximum content of total saponins and polysaccharide. (3) The content of active ingredients revealed stability between the years 2012 and 2013, but the content of polysaccharide was significantly different. The content in 2012 was higher than that of 2013. (4) The content of active ingredients reached the peak in autumn, which was the best harvest season. (5) Among different component content detection of nutritional organs, tubers had the maximum content of ethanol-soluble extractives, total saponins and polysaccharide. Leaves also contained higher content of ethanol-soluble extractives and total saponins than roots and stems. All of these provide theoretical basis for plant, harvest and production of H. zhejiangensis, which is an endemic, rare, and endangered medicinal plants.

5.The basic strategies and research advances in the studies on glycosyltransferases involved in ginsenoside biosynthesis.

Traditional herbal medicines, Panax ginseng, Panax quinquefolium and Panax notoginseng, attract our attention for their extensive and powerful pharmacological activities. Ginsenosides are the main active constituents of these medicinal herbs. The related glycosyltransferases involved in ginsenoside biosynthesis are the key enzymes which catalyze the last important step. Modification of ginsenoside aglycones by glycosyltransferases produces the complexity and diversity of ginsenosides, which have more extensive pharmacological activity. At present, ginsenoside aglycones and compound K have been obtained by synthetic biology. As the last step of ginsenoside biosynthesis, glycosylation of ginsenoside aglycones has been studied intensively in recent years. This review summarizes the basic strategies and research advances in studies on glycosyltransferases involved in ginsenoside biosynthesis, which is expected to lay the theoretical foundation for the in-depth research of biosynthetic pathway of ginsenosides and their production by synthetic biology.

6.Research advances of the influence factors of high level expression of recombinant protein in Pichia pastoris.

Pichia pastoris is one of the most important systems used in the field of molecular biology for the expression of recombinant proteins. The system has advantages of high expression, high stability, high secretion, easy high-density fermentation and low cost. Many factors affect the expression of recombinant protein, such as gene copy number, codon usage preference, type of promoter, molecular chaperones, glycosylation, signal peptide and fermentation process. In this review, research advances of the above aspects are summarized, which lay a foundation for improving the expression of recombinant proteins in P. pastoris.

7.Construction of Saccharomyces cerevisiae haploid mutant deficient in lanosterol synthase gene.

Lanosterol synthase is encoded by the erg7 gene and catalyzes the cyclization of 2, 3-oxidosqualene, which is a rate-limiting step of the inherent mevalonate (MVA)/ergosterol metabolic pathway in Saccharomyces cerevisiae. The intermediate 2, 3-oxidosqualene is also the precursor of triterpenoids. Therefore, the cyclization of 2, 3-oxidosqualene is the key branch point of ergosterol and triterpenoids biosynthesis. Down-regulation of 2, 3-oxidosqualene metabolic flux to ergosterol in S. cerevisiae may redirect the metabolic flux toward the triterpenoid synthetic pathway reconstructed by the synthetic biology approach. To construct erg7 knockout cassette harboring the loxP-Marker-loxP element, long primers were designed, which were homologous to the sequences of both erg7 ORF and plasmid pUG66. The cassette was transformed into diploid wild strain INVSc1 by LiAc/SS Carrier DNA/PEG method and then erg7 gene haploid deficient mutant was obtained by homologous recombination. The results of semiquantitative PCR and real-time quantitative PCR revealed that erg7 expression level in erg7 gene haploid deficient mutant is one time lower than that in wild strain. The results of TLC and HPLC showed that the ergosterol content in deficient mutant decreased to 42% of that in wild strain.

8.Treatment of HBeAg positive chronic hepatitis B by xiaoyao powder combined with interferon-alpha: a clinical observation.

OBJECTIVETo study the efficacy of Xiaoyao Powder (XYP) combined with interferon alpha (IFN-alpha) in treating HBeAg positive chronic hepatitis B (CHB) patients and the effect on their quality of life (QOL).
METHODSTotally 193 patients with HBeAg-positive CHB confirmed by liver biopsy were randomly assigned to 2 groups, Group A (94 cases) and Group B (99 cases). IFN-alpha1b was subcutaneously injected to patients in Group A at the dose of 50 microg, thrice per week. Those in Group B additionally took XYP. The therapeutic course for all was 24 weeks. Clinical efficacy was observed by assessing ALT restoration rate, HBeAg negative rate, HBeAg conversion rate, HBV DNA negative rate, complete response rate, partial response rate, and symptoms integral. The evaluation of QOL was performed by using chronic liver disease questionnaire (CLDQ) score. Adverse reaction occurrence rate was observed in the two groups.
RESULTSBetter effects were obtained in Group A on ALT restoration rate, HBeAg negative rate, HBV DNA negative rate, complete response rate, partial response rate, TCM symptoms integral, the total effective rate of TCM sysmptoms, CLDQ score, and adverse reaction rates, showing statistical difference when compared with Group B (P < 0.05, P < 0.01).
CONCLUSIONXYP could elevate the efficacy of TCM symptoms of HBeAg-positive CHB patients and anti-viral effect, improve their QOL, and reduce adverse reaction of IFN-alpha.

9.Seasonal variations in the incidence of deep vein thrombosis following total knee or hip arthroplasty

Objective To explore the difference in the incidence of deep vein thrombosis (DVT) following total knee arthroplasty (TKA) or total hip arthroplasty (THA) between different seasons.Methods The present retrospective study examined 2 363 patients undergoing TKA or THA from August 2008 to February 2016.There were 653 male and 1 710 female with the average age of 64.44±13.03 years old (17-91 years old),including 954 patients in TKA and 1 409 in THA.All of the patients received anticoagulant drugs and were compressed with a pressure pump starting on the first postoperative night.Routine venography of the bilateral lower limbs was performed at 3-5 days after the operation.The incidence of DVT in different seasons and different age groups (≥65 years old and <65 years old) were counted.Results The present study suggested that the probabilities of DVT after arthroplasty in spring,summer,fall and winter were 15.85% (116/732),14.92% (71/476),17.88% (108/604),22.50% (124/551) respectively with significantly difference (P<0.05).The incidence of DVT in winter was higher than that in spring and summer (P< 0.017).The probabilities of DVT after TKA in spring,summer,fall and winter were 19.09% (59/309),15.67% (37/236),18.11% (44/243),27.71% (46/166) respectively with significantly difference (P<0.05).The incidence of DVT in winter was higher than that in summer (P<0.017).The probabilities of DVT after THA in spring,summer,fall and winter were 13.48% (57/423),14.17% (34/240),17.73% (64/361),20.26% (78/385) respectively with significantly difference (P<0.05),and that in winter was higher than in spring (P<0.017).The symptomatic DVT after arthroplasty in spring,summer,fall and winter were 3.55% (26/732),3.78% (18/476),4.97% (30/604),6.90% (38/551) with significantly difference in different seasons (P<0.05),and that in winter was higher than in spring (P<0.017).The symptomatic DVT after TKA in spring,summer,fall and winter were 3.88% (12/309),4.24% (10/236),4.94% (12/243),9.64% (16/166) with significantly difference in different seasons (P<0.05).The symptomatic DVT in winter was higher than that in spring (P<0.017).The symptomatic DVT after THA in spring,summer,fall and winter were 3.31% (14/423),3.33% (8/240),4.99% (18/361),5.71% (22/385) respectively (P>0.05).The probability of DVT following arthroplasty in older patients (age≥65 years old) in spring,summer,fall and winter were 18.49% (76/411),16.61% (45/271),22.07% (81/367),28.05% (99/353) with significantly difference among the groups (P<0.05),while symptomatic DVT in older patients (≥65 years old) in spring,summer,fall and winter were 4.38% (18/411),4.43% (12/271),5.72% (21/367),8.78% (31/353) respectively with significantly difference (P<0.05).The probability of DVT in four seasons were 12.46% (40/321),12.68% (26/205),11.39% (27/237),12.63% (25/198) in younger patients (<65 years old).However,the difference was not detected among different seasons (P>0.05).The probability of symptomatic DVT in four seasons were 2.49% (8/321),2.93% (6/205),3.80% (9/237),3.54% (7/198) in younger patients (<65 years old) without significant difference (P>0.05).Conclusion Seasonal variations could place an important effect on the incidence of DVT following TKA or THA,especially for the old patients with age ≥65 years.
10.Construction and verification of replication-deficient recombinant adenovirus to silence CTGF gene by AdEasy system

Objective:To design and construct the replication-deficient recombinant adenovirus Ad-siCTGF which can silence the expression of connective tissue growth factor (CTGF) by RNA interference and verified its function. Methods:A specific sequence, which was verified to be able to silence CTGF gene with high efficiency, was cloned into pSES-HUS vector to produce the shuttle plasmid pSES-siCTGF. The plasmid after Pme Ⅰ linearization was cotransduced with pAdEasy into BJ5183 E.coli strains to construct recombinant vector Ad-siCTGF. After linearization treatment with Pac Ⅰ enzyme digestion Ad-siCTGF was transfected into HEK293 cells via liposome mediation. The recombinant adenovirus was packaged. The titer of the Ad-siCTGF was increased after three times of cross-infection. 4T1 cells were infected with the adenovirus. The silencing efficiency was tested by real-time fluorescence quantitative (RFQ)-PCR and Western blotting.Results:Pac Ⅰ enzyme digestion electrophoresis indentified that recombinant adenovirus was successfully constructed. The titer of the recombinant adenovirus Ad-siCTGF was 2.6×10~(10) pfu/mL after amplification and purification. The CTGF mRNA and protein expression in 4T1 cells were decreased by 36.27% and 31.56%, respectively, compared with the control groups.Conclusion:The recombinant adenovirus which can silence the expression of CTGF was successfully constructed. It laid a good foundation for further investigation of the action mechanism of CTGF in tumor cells.
