Chinese Journal of Endocrine Surgery 2021;15(1):71-77
Objective:To investigate the role of TCF7L2 gene polymorphism and related mechanisms in postmenopausal women with type 2 diabetes mellitus (T2DM) who are at high risk for osteoporosis (OP) .Methods:148 postmenopausal women with T2DM from Jan. 2019 to Jun. 2020 were selected as study subjects. Among them, 86 patients combined with OP (T2DM+OP group) and 62 patients without OP (T2DM group) were included, and 100 healthy postmenopausal women who underwent physical examination in our hospital during the same period were included as the control group. Fasting venous blood was extracted to detect clinical and bone metabolism indicators. Two SNP sites of TCF7L2, rs7901695 (T>C) and rs290487 (C>T) , were genotyped by capillary electrophoresis and section analysis (SNaPshot) . The relative mRNA expression of TCF7L2 gene was determined by quantitative real-time fluorescence quantitative PCR assay. To investigate the effects of overexpression and silencing of TCF7L2 on proliferation of rat osteoblasts, lentiviral vectors were constructed. Results:For rs7901695, TT, TC and CC genotypes in T2DM+OP group, T2DM group and control group were significantly different ( χ2=12.545, P=0.014) , and the frequency distribution of T and C alleles was significantly different among the three groups ( χ2=17.089, P<0.001) . For RS290487, CC, CT and TT genotypes of T2DM+OP group, T2DM group and control group were significantly different ( χ2=10.500, P=0.033) , and the frequency distribution of C and T alleles was significantly different among the three groups ( χ2=14.665, P=0.001) . For rs7901695, FPG and TG levels in T2DM+OP patients with CC+TC genotype were significantly higher than those with wild-type TT genotype (FPG: t=2.559, P=0.014; TG: t=2.034, P=0.036) , while serum calcium and 25OHD3 levels were significantly lower than those with Wild-type TT genotype (calcium: t=3.889, P<0.001; 25OHD: t=4.112, P<0.001) . For rs290487 site, serum calcium, BGP and 25OHD3 levels in T2DM+OP patients with TT+CT genotype were significantly lower than those with wild-type CC genotype, and the difference was statistically significant (calcium: t=3.751, P<0.001; BGP: 2.731, P=0.007; 25OHD3: t=3.225, P=0.002) . The relative mRNA expression level of TCF7L2 gene in T2DM+OP patients was significantly lower than that in T2DM group and the control group ( F=5.735, P<0.05) . With the increase of Allele C at rs7901695, the relative mRNA expression level of TCF7L2 gene decreased gradually ( F=5.723, P<0.05) . In addition, the proliferation rate of rat osteoblasts overexpressed by TCF7L2 was significantly increased, while the proliferation level of TCF7L2 silenced osteoblasts was significantly decreased ( F=7.846, P<0.05) . Conclusion:In postmenopausal WOMEN with T2DM, the variation of TCF7L2 gene will lead to the decrease of gene expression level, and the suppression of osteoblasts proliferation, thus participate in the pathogenesis of OP.
3.An Effective Strategy for the Development of Rapeseed Oil-Based Biodiesel Industry

China Biotechnology 2006;0(06):-
Rapeseed oil is one of the major raw materials for biodiesel production. The bottleneck of rapeseed oil-based biodiesel production at present is high cost of raw materials, which generally accounts for about 75% of total production cost. Current status and principal problems concerned in the production of biodiesel from rapeseed oil in China and other countries were introduced, based on which it was proposed to grow high-erucic acid rape in existing winter fallow fields and jointly produce biodiesel, erucic acid and its derivative products, and other highly valuable by-products such as glycerin and sterols from high-erucic acid rapeseed oil. The feasibility of this development strategy was analyzed in terms of product and market demand potential, economic benefits of enterprises, techniques and conditions of production, and source of raw materials. It was believed that the implementation of the strategy in China was of great significance in realizing rise and sustainable development of rapeseed oil-based biodiesel production, improving quality of biodiesel, promoting the development of biochemical engineering and many other industries, as well as in building a socialist new countryside.

OBJECTIVE:To provide reference for the development of pharmaceutical care for periconceptional women. METH-ODS:Probability proportionate to size sampling(PPS)method was adopted to select participants from periconceptional women in Yongchuan District. The participants were investigated by questionnaires about drug use safety knowledge, attitude and behavior. Ep-idata 3.0 software was used to establish database. The data were analyzed with SPSS 15.0 software. RESULTS:Totally 800 ques-tionnaires were distributed and then 779 valid questionnaires were collected with effective rate of 97.38%. The total awareness rate of knowledge about drug use safety was 57.69%. The awareness rate rise with the increase of educational level (P<0.01). The awareness rate was higher in pregnant women than in prepregnant ones, with statistical significance in most of the questions(P<0.05 or P<0.01). For questions about attitude for drug use safety, the positive answer rate was higher in pregnant women than in prepregnant ones, with statistical significance in most of the questions (P<0.01). Among 385 pregnant women, only 56.88% re-ceived folic acid following the WHO recommendation. Of 181 pregnant women who had taken medicine,8.29% indicated that the physicians or pharmacists did not inform that whether the medicine affected pregnancy. CONCLUSIONS:Among periconceptional women, awareness rate of drug use safety is in low level and varied by their educational levels. There are difference in awareness rate of drug use safety and attitude between prepregnant women and pregnant woman. Drug use behavior of pregnant woman is to be modified.

Journal of Medical Postgraduates 2003;0(08):-
Objective: To analyze the aberrant methylation of death-asscciated protein kinase (DAPK) gene in blood from non-small cell lung cancer (NSCLC) patients and to evaluate its clinical significance. Methods: A methylation-specific PCR was performed for the detection of promoter hypermethylation of DAPK gene in blood DNA from 65 cases of NSCLC patients, and the relation of the aberrant methylation of DAPK gene and the clinical pathological data was analyzed. Results: 30.8% (20/65) of the blood from 65 cases of NSCLC patients showed hypermethylation in DAPK promoter, whereas no methylated DAPK gene promoter was found in blood from normal control or patients with benign lung diseases. DAPK gene promoter methylation in blood did not strongly correlate with the pathological class, stage, metastasis and differentiation of NSCLC. Conclusion: Detection of the aberrant methylation of DAPK gene in blood DNA from NSCLC patients might offer an effective means for the earlier auxiliary diagnosis of the malignancy.
