2.STUDY ON THE INHIBITED PROLIFERATION AND G_2/M ARREST OF HUMAN GASTRIC CARCINOMA CELLS INDUCED BY GENISTEIN IN VITRO

Objective: To study the effects of genistein on the proliferation and cell cycle progression of human gastric carcinoma cells. Methods: 3H TdR incorporation test was used to investigate the cell proliferation. Flow cytometry was used to analyze the cell cycle arrest. Immunocytochemistry technique and Western blotting were used to observe the cyclin B and P21 waf1/cip1 protein expression. Results: Genistein inhibited the proliferation of tumor cells significantly, arrested cell cycle progression at G 2/M phase, and enhanced cyclin B and P21 waf1/cip1 protein expression in dose dependent manner. Conclusion:Proliferatory inhibition and G 2/M arrest of human gastric carcinoma cells after treated with genistein may be due to increased stability of cyclin B protein and the expression of P21 waf1/cip1 .
3.Effects of five flavonoids on expression of Bcl-2 family proteins in apoptosis of myocardiocytes

Objective To observe the effects of five flavonoids include rutin(RU),dihydromyricetin(DMY),hesperetin(HP),daidzein(DA)and hydroxysaffor yellow A(HYSA)on myocardiocyte apoptosis induced by H2O2 and to explore their relationships with Keshan disease and the possible mechanism.Methods Primary cultured cardiocytes of neonatal rats were randomly divided into control group,model group,and flavonoids preincubation group.The cardiocyte apoptosis was examined by fluorescent staining,the rates of apoptosis were detected by flow cytometry,the expression of Bcl-2 family proteins associated with apoptosis were observed:by Western blot.Results Compared with model group[(24.33±6.51)%],RU[(13.95±3.80)%],DA[(11.82±3.50)%],HYSA[(12.33±3.78)%]could decreased the rate of apoptosis(P<0.05).The five flavonoids could up-regulate Bcl-2 expression,down-regulate Bax expression,and increase the Bcl-2/Bax ratio[RU(0.989±0.094),DMY(0.931±0.280),HP(0.980±0.095),DA(1.049±0.092),HYSA(1.031±0.039),vs model(0.490±0.046),the difference had statistical significances(P<0.05)],but the Bcl-xl did not significantly changed(P>0.05).Conclusions RU,DMY,HP,DA and HYSA have antiapoptotic effects on cardiomyocyte via regulating Bcl-2 and Bax,which gives us a hint in prevention and treatment of Keshan disease.
4.The correlation between lumbar facet triaxially angle in patients with degenerative lumbar spondylolisthesis

Objective To study the etiological effect between the spatial variation of lumbar facet orientation and the degenerative lumbar spondylolisthesis(DLS)in axial,sagittal and coronal plane.Methods 85 patients with DLS at L4 and 110 controlsreceived X-ray and CT scanning.The facet angle at L4-L5 was measured in axial,sagittal and coronal plane.Results Compared with the control group, the DLS-angles were lesser and the difference between the left and right was greater in axial,coronal plane(P <0.05).But were greater in sagittal plane(P <0.05).Conclusion The DLS-angles are more sagittal in axial plane,more level in sagittal plane and more erect in coronal plane.The change of spatial orientation in the facet joint is one of etiological factors of degenerative lumbar spondylolisthesis.
5.Some Teaching Experience by Using the Mode of Learning Followed by Teaching

A new teaching method was developed in the curriculum of Harmful Microorganisms Control Technology.It is characterized by students’ self-learning followed by student’s instruction.Both students and teacher have succeeded in this model after four stages of practice,in which a pleasant learning atmosphere was created in the classroom.An effective interaction between teacher and students was achieved.Students are viewed as main objects in the classroom and they are encouraged to ask questions,to formulate their own ideas,or to find things out for themselves.Thus,students’ abilities including presentation,communication,competition,and cooperation were enhanced.By adapting their role to the new teaching method,teachers have also improved their teaching skill and strategies.
6.Ultrasonic microbubbles for glioma-targeted drug delivery.

Ultrasonic microbubbles were used to open blood-brain barriers (BBB) with a reversed and limited behavior feature in the study, which could improve the brain-targeted delivery of anti-tumor drugs. The glioma rat model was prepared. Low-frequency ultrasound was combined with microbubbles to affect the permeability of BBB compared with the permeability of independently administered Evans blue (EB) crossing BBB. Time point and length of ultrasound were investigated whether they affect the permeability of BBB and the damage of brain tissue. The effect of the growth time of glioma on BBB permeability was explored. Only glioma had a very little impact on BBB permeability. However, ultrasonic microbubbles opened the BBB with the features of temporary, limited and reversed behavior and improved EB and magnetic resonance imaging contrast agent penetrating BBB. A length of 30 s ultrasound is appropriate for opening BBB and no damage of brain tissue. Drugs should be injected before ultrasound so that they enter into brain as BBB opening. Ultrasonic microbubbles can open BBB effectively and safely, which improve drugs penetrating BBB under proper time point and length.

7.Clinical characteristics and prognosis of large cell lung cancer.

OBJECTIVEThe aim of this study was to explore the clinical characteristics and analyze the prognostic factors of large cell lung cancer (LCLC).
METHODSThe clinical data of 111 LCLC cases were collected and retrospectively analyzed. The prognostic factors were evaluated by univariate and multivariate analyses.
RESULTSAmong the 111 cases, the lesions were in the right lung of 53 patients and 26 of them were located in the superior lobe. The lesions were in the left lung of 58 cases, and 35 of them were in the superior lobe. The lesions were presented as central in 36 cases and peripheral in 75 cases, with a mean diameter of 5.3 cm. All the 111 patients were diagnosed as stage I in 38 cases, stage II in 11 cases, stage III in 45 and stage IV in 17 cases. 60 patients had lymph node metastasis and 17 cases had distant metastasis. The overall 1-, 3- and 5-year survival rates of the LCLC were 54.7%, 30.9% and 20.6%, respectively. Cox univariate analysis revealed that TNM stage (P = 0.000), lymph node metastasis (P = 0.000) and M stage (P = 0.000) are prognostic factors. Cox multivariate analysis indicated that TNM stage (P = 0.000) is an independent prognostic factor.
CONCLUSIONThe prognosis of LCLC is worse than other types of non-small cell lung cancer. Complete surgical resection remains the main therapeutic approach. TNM stage is an independent prognostic factor.

8.Application of 24h ambulatory blood pressure monitoring in aged patients with hypertension

Objective: To explore application of 24h ambulatory blood pressure monitoring (ABPM) in aged patients with hypertension. Methods: A total of 223 aged patients with hypertension treated in our hospital from Feb 2013 to Feb 2015 were selected. According to age, patients were divided into young age group (60~79 years, n=137) and advanced age group (≥80 years, n=86). Another 80 normotensive aged people undergoing physical examination in our hospital simultaneously were regarded as healthy control group. The 24h systolic blood pressure (SBP), diastolic blood pressure (DBP) and pulse pressure (PP) were measured and compared between three groups. Results: Ambulatory blood pressure: compared with healthy control group, there were significant rise in 24h mean blood pressure (BP) [24hSBP: (117. 63±11. 53) mmHg vs. (132. 04±11. 23) mmHg vs. (144. 26±12. 87) mmHg], daytime BP [dSBP: (119. 85±13. 20) mmHg vs. (134. 26±13. 52) mmHg vs. (146. 83±10. 64) mmHg]and nighttime BP [nSBP: (115. 48±9. 74) mmHg vs. (128. 76±10. 85) mmHg vs. (141. 67±13. 42) mmHg]in young age group and advanced age group (P<0. 05 or<0. 01). Compared with young age group, there were significant reductions in 24h DBP, dDBP and nDBP, and significant rise in 24h SBP, dSBP, nSBP, 24h PP, dPP and nPP in advanced age group (P<0. 05 or <0. 01). Conclusion: The 24h ABPM can effectively display the blood pressure abnormal fluctuations in aged patients with hypertension, and the higher age is, the greater blood pressure abnormal fluctuation is.
9.Protection of Tongxinluo Against Myocardial Ischemia/Reperfusion Injury Relates to Enhancing Endothelial Barrier in Diabetic Rats

Objective: To clarify that protection of Tongxinluo against myocardial ischemia/reperfusion injury relates to enhancing endothelial barrier in diabetic rats.
Methods: A total of 32 Zucker diabetic rats were randomized into 4 groups:Sham group, Model group, Insulin group and Tongxinluo group, n=8 in each group. In addition, there was a Control group containing 8 non-diabetic Zucker rats. Myocardial ischemia/reperfusion injury model was established by a 45-min ischemia and 3-h reperfusion protocol. The size of infarction was detected by pathological staining, the microvascular permeability was examined by Miles assay to obtain the lfuorescein isothiocyanate concentration in myocardial tissue, the Triton X-100 soluble and insoluble VE-cadherin was measured by membrane protein extraction and western blot analysis.
Results:The size of infarction in Model group was obviously higher than that in Control group (55.2 ± 1.4)%vs (36.2± 1.3)%,P<0.05 and the lfuorescein isothiocyanate concentration was also higher in Model group than Control group. Both insulin group and Tongxinluo group had the similar size of infarction with Model group (36.8 ± 1.2)%vs (38.7 ± 1.1)%, P>0.05. Both insulin group and Tongxinluo group got lower lfuorescein isothiocyanate concentration (all P<0.01) and VE-cadherin internalization (all P<0.05) than that in Model group.
Conclusion: Protection of Tongxinluo against myocardial ischemia/reperfusion injury in diabetic rats is as effective as
insulin, but the effect is independent of reducing blood glucose and may be related to enhancing endothelial barrier.
10.Analysis of epidemiologic feature and genetic sequence of Sapovirus in China.

To investigate epidemiologic feature and genetic variance of Sapovirus among children in China, fecal specimens were collected from children under 5 years old with acute diarrhea from Feb 2006 to Jan 2007 in nine provinces including Anhui, Fujian et al. A total of 1,110 fecal samples were detected for Sapovirus by reverse transcriptase-PCR (RT-PCR). Ten samples (0.9%) were positive for Sapovirus. The PCR products were then sequenced and analysed by phylogenetic tree. The results indicated that the detected Sapovirus strains were classified into two genogroups and three genotypes, including G I/1, G I/3, G II/3.
