1.The dementia models induced by intracerebroventricular infusion of ?-Amyloid peptide in mice

Aim To establish the dementia mice models induced by intracerebroventricular infusion of ?-Amyloid peptide and related measuring index. Methods The mice models were made by intracerebroventricular infusion of long fragment of soluble?-Amyloid peptide with micropump, intracerebroventricular injection of long soluble?-Amyloid peptide fragment and short insoluble ?-Amyloid peptide fragment,and intracerebroventricular injection of ?-Amyloid peptide combining with transforming growth factor. Results Compared with normal control, the learning and memory ability was decreased in all above model mice. the ChAT activity in both hippocampus and cortex of model mice decreased, the hippocampal neuron were depleted and the content of apoptosis associated protein Bal-2?BAX and p53 were increased. Conclusions All above mice models could be choose as the dementia models which were simulating the pathological charactoristics of the Alzheimer's disease.
2.THE INFLUENCE OF JIAOGULAN EXTRACT ON PLATELET AGGREGATION AND ARACHIDONATE METABOLISM IN RABBITS

jiaogulan ( Gynostemma pentaphyllum) extract (0.25~2g/L. final concentration) obviously inhibited the platelet aggregation & TXB2 release induced by arachidonic acid ( AA ) in vitro, and the ID50 of inhibiting TXB2 release was 0.28g/L. With the dose of 35mg/kg i. v. , it evidently inhibited platelet aggregation at 10 & 20 min after injection, and also markedly decreased TXB2 released by platelets during 10~40 min after injection, especialy within 20 min. Jiaogulan extract (0.25~4g/L) did not influence 6-keto-PGF1? release of aorta thoracalis of rabbits in vitro. Enzyme immunoassay was used to measure the content of TXB2 & 6-keto-PGF1? in the experiments. The above results suggest that Jiaogulan extract may decrease TXA2/PGI2 ratio.
3.Detection significance of platelet indexes and pancreatic blood flowparameters in patients with severe acute pancreatitis

Objective To study and analyze the detection significance of platelet indexes and pancreatic blood flow parameters in the patients with severe acute pancreatitis(SAP).Methods Sixty-one patients with SAP in our hospital from June 2014 to June 2016 were selected as the group A,contemporaneous 61 patients with mild acute pancreatitis were selected as group B and 61 persons undergoing the healthy physical examination were selected as the group C.Then the platelet indexes and pancreatic blood flow parameters were detected and compared among 3 groups,and at the same time the platelet indexes and pancreatic blood flow parameters in the group A were compared among different stages of disease course.Results MPV,PDW and platelet activation indexes in the group A were all significantly higher than those in the group B and C,while PCT was lower than that in the group B and group C,the pancreatic head and pancreatic tail blood flow parameters were all worse than those in the group B and C,moreover the pancreatic head and pancreatic tail blood flow parameters in the group B were all worse than those in the group C,and the platelet indexes and pancreatic blood flow parameters in the group A all had significant differences among different stages of disease course (P<0.05),the differences in these indexes were statistically significant.Conclusion The detection significance of platelet indexes and pancreatic blood flow parameters in the patients with SAP is higher,and also has a certain value for the disease course stages in the patients with SAP,so their monitoring has higher value.

5.Effect of collaborative health education by both doctors and nurses on parturition

Objective To explore the effect of collaborative health education by both doctors and nurses on parturition?Methods One hundred and sixty primiparas at uterogestation undergoing analgesic parturition were assigned equally into the control and observation groups according to their admission sequence? The control group received routine nursing intervention and the observation group the collaborative health education by both doctors and nurses? The two groups were compared in terms of analgesic effect,vaginal delivery and post-partum hemorrhage? Result The rates of analgesic delivery and vaginal delivery in the observation group were significantly higher and the volume of postpartum hemorrhage was significantly smaller than those of the control group(all P < 0?05)?Conclusion The collaborative health education may be effective in alleviating mental stress of the preimiparas,enhancing the analgesic effect,increasing the rate of vaginal delivery and decreasing the volume of postpartum hemorrhage?

7.Clinical studies of bacillus of Calmette-Guerin polysaccharide nucleic acid adjunctive therapy in pulmonary tuberculosis

Objective To evaluate the effectiveness, the influence on cellular immune function and the side-effect of bacillus of Calmette-Guerin polysaccharide nucleic acid(BCG-PSN)combined with antituberculous chemotherapy in the treatment of pulmonary tuberculosis. Methods A total of 60 pulmonary tuberculosis patients were divided into treatment group(30 patients)and control group(30 patients)by random digits table. All patients accepted the same standard antituberculous chemotherapy, meanwhile patients in treatment group were injected with BCG-PSN. Observed and compared the clinical symptom,the size of the focas nidus,the change of toxic response and immunity. Results The symptoms were significantly relieved in both groups after treatment(P<0.05), but it was significantly better in treatment group(P<0.05), the effective rate and control rate in treatment group[36.7%(11/30),96.7%(29/30)]were significantly higher than those in control group[23.3%(7/30), 86.7%(26/30)](P < 0.05). After treatment,the levels of CD3,CD4 and IL-2 were higher, and the level of CD8 was lower, but the treatment group improved significantly better than control group(P < 0.05). The rate of leukopenia was lower in treatment group than that in control group[10.0%(3/30)vs. 33.3%(10/30),P <0.05]. As to the safety,no other toxicities were observed in the treatment group. Conclusions BCG-PSN combined with antituberculous chemotherapy in the treatment of pulmonary tuberculosis contributes to relieve the symptom, reduce size of the nidus, decrease leukopenia incidence and enhance the cell immunity. It is safe.
8.Construction of a plant effective expression vector containing the gene of hepatitis B virus surface antigen

OBJECTIVE: To construct a plant effective expression vector driven by a fruit specific promoter for the expression of hepatitis B virus surface antigen (HBsAg), to further improve the expression of exogenous gene in plant, and to prepare for the development of an effective anti-hepatitis vaccine. METHODS: Tomato fruit-specific promoters' gene 2A12 and E8 were respectively introduced to pBPFOmega7 to form pB2A12 and pBE8. The DNA fragment containing HBsAg-s gene from plasmid YEP-HBs was inserted respectively into pB2A12 and pBE8 to form pB2A12-HBs and pBE8-HBs. The fragment containing "p35S+2A12+Omega+HBsAg-s+Tnos" of the pB2A12-HBs was sub-cloned into plasmid pCAMBIA1301 to yield the reconstructed plant binary expression plasmid pCAM2A12-HBs, and the fragment containing "p35S+E8+Omega+HBsAg-s+Tnos" of the pBE8-HBs was sub-cloned into plasmid pCAMBIA1301 to yield the plasmid pCAME8-HBs. The inserted gene HBsAg and fruit-specific promoters in the reconstructed plant binary expression vectors were confirmed by sequencing. Then, pCAM2A12-HBs and pCAME8-HBs were directly introduced into Agrobacterium tumefaciens strain EHA105. RESULTS: Digestion with restriction enzymes proved that all recombinant vectors had the inserts with expected length of the target fragments, and the sequencing results were confirmed correct. CONCLUSION: In this study, plant expression vector containing HBsAg gene driven by fruit specific promoter and CaMV35s promoter was successfully constructed.
9.Investigation on the practice status of health industry standard of Nursing Grading

Objective To investigate the training effect and application development of the health industry standard of Nursing Grading, and to explore it'sclinical applicability. Methods An electronic questionnaire survey was conducted among nurses in 30 three general hospitals of Shandong Province from November 10 to December 10 in 2015. The questionnaire mainly included four parts: basic information, training status, application status and the clinical applicability. All the questionnaires were automatically submitted by the nurse′s cell phone or computer, 3386 valid questionnaires were recovered. Results The status of standard training:93.92%(3180/3386) nurses received a comprehensive training about the standard. The answer rates of 4 theoretical examination questions was 88.95%(3012/3386), 84.49%(2861/3386), 85.17%(2884/3386), 65.71%(2225/3386)respectively. 77.26%of the nurses needed to continue training. Application status: 97.67% (3307/3386) nurses in the hospital has begun to apply Nursing Grading standard, and 97.01% (3208/3307) and 97.76% (3231/3307) of the hospital revised the standard of care systems and work processes; 78.23% (2649/3386) nurses divide the level of care based on both the patient's condition and self-care ability presently. 74.45%(2521/3386) nurses did it as follows,the doctor determine the patient′s condition level, the nurse determine the level of patient′s self-care ability, then the nursing grading was determined according to the both. In the clinical applicability survey, the choice proportion of very agree and agreeofHelping nurses to determine the patient′s level of care and It is beneficial to improve the efficiency of nursing management was 86.30% (2922/3386) and 82.66%(2799/3386);The choice proportion of very agree and agree ofHelp nurses to improve the quality of nursing services, reflecting the value of the nursing work, to meet the needs of patients was 79.36% (2687/3386), 76.56% (2592/3386), 80.98% (2742/3386) respectively. Conclusions The hospital attaches great importance to the standard of Nursing Grading, but the training effect needs to be improved;standard has been widely used in Shandong Province Physician-nurse collaboration has become the decision-making main body of nursing grading;the standard application can not only help the nurses to do decision-making correctly and improve the efficiency. It also can help nurses to improve the quality of nursing service, and enhance patients′satisfaction. So it is very suitable for clinical practice.
10.Effects of Stilbene-glycoside on Learning and Memory and Inflammatory Reaction of Brain in Dementia Mice

Objective To observe the effect of 2,3,5,4'-tetrahydroxy stilbene-2-?-D-glycoside (TSG)on learning and memory and free radicals metabolism in mice with dementia induced by ?-amyloid(A?).Methods The mice models were established by injection of A?1-40 into the right lateral ventricle,and the treatment group was administered with TSG for 8 weeks by gastrogavage.Morris water maze and step-through test were performed in all the mice and then the mice were killed and radioimmunoassay was used to detect the content of interleukin-6(IL-6).Results The learning and memory in model mice were worse and the cortical IL-6 content increased compared to the normal control mice.TSG improved the learning and memory of A?-induced model mice and reduced cortical IL-6 content.Conclusion TSG could improve the learning and memory of dementia mice and decrease cortical IL-6 content,indicating a promising prospect in the treatment of dementia disease such as Alzheimer's disease.
