1.Effect of propofol and monosialoterahexosyl ganglioside on cognitive function and the expression of caspase-3 in immature rats

Objective To investigate the effects of multiple doses of propofol or/and monosialotetrahexosyl ganglioside (GM1) on cognitive function and caspase-3 expression in immature rats.Methods Seventy-seven immature SD rats,17-18-day old and weighted 33-40g,were randomly divided into 7 groups (11 each):control group (NS),propofol 50mg/kg group (P50),propofol 100mg/kg group (P_ 100 ),propofol 200mg/kg group (P_ 200 ),GM1 10mg/kg group (G),propofol 100mg/kg and GM1 10mg/kg group (P_ 100 G),and propofol 200mg/kg and GM1 10mg/kg group (P_ 200 G).Each group received normal saline,propofol 50mg/(kg?d),100 mg/(kg?d),200mg/(kg?d),GM1 10mg/(kg?d),propofol 100mg/(kg?d) and GM1 10mg/(kg?d),or propofol 200 mg/(kg?d) and GM1 10mg/(kg?d) intraperitoneally in a bolus or divided doses,respectively,for 6 consecutive days.The tests of learning and memory were performed in Morris water maze everyday at the 3rd hour after the rats waked up from anesthesia.The animals were sacrificed on the 6th day after the Morris water maze test to obtain the brain specimen,and the expression of caspase-3 was detected by immunohistochemistry and Western blotting.Results Latency period of water maze test was significantly longer in groups P_ 100,P_ 200 and P_ 200 G than in group NS,and the frequency of crossing platform were fewer in P_ 100,P_ 200 and P_ 200 G groups (P
2.Antimicrobial Resistance of Bacteria Isolated from a Hospital 2007-2008

OBJECTIVE To investigate the resistance of clinical isolated strains to the commonly used antibacterials in our hospital 2007-2008.METHODS Clinical isolated strains and sensitivity of drugs were detected by ATB system.The result of drug sensitivity was judged by CLSI standard and analyzed with statistical software WHONET5.3.RESULTS Altogether 3150 strains bacteria were isolated,17.4% were Gram-positive strains and 82.6% were Gram-negative strains,and the top five isolates were Pseudomonas aeruginosa,Escherichia coli,Klebsiella pneumoniae,Acinetobacter baumannii,and Staphylococcus aureus.The reasistance rate of Gram-positive strains to minocycline was 15.4%.Five VRE strains were isolated.Various Enterobacteriaceae bacteria were sensitive to imipenem meropenem,cefoperazone-sulbactam and piperacillin-tazobactam,and their rate was 86.5% to 97.7%.Some of Acinetobacter and Stenotrophomonas maltophilia were multidrug resistant.CONCLUSIONS It is serous that multidrug resistance of isolated strains of the patients exists in our hospital.
3.Expression of bmp3,bmp4 and bmp7 during dog permanent tooth root formation

Objective:To evaluate the physiological roles of bmp3,bmp4 and bmp7 during the formation of permanent tooth roots in dog. Methods:The expression of bmp3,bmp4 and bmp7 mRNA at different stages of the development of permanent tooth roots was examined by in situ hybridization in 3 dogs aged 12-18 weeks. Results:bmp3 was found in dental sac surrounding the germs at the early stage of tooth root development, and in cementoblasts and periodontal cells at the later stage. bmp4 was found in odontoblasts, dental papilla and osteoblasts. bmp7 positive signals was found only in epithelial cells of root sheath around cervical circulus at early stage, then located in cementoblasts and odontoblasts at later stage. Conclusion:The spatiotemporal expressions of bmp3,bmp4 and bmp7 are widely diverse, indicating that they participate in the regulation of tooth root development.
4.Survival analysis of unresectable advanced pancreatic cancer after high intensity focused ultrasound therapy

Objective To observe the 2-year survival situation of high intensity focused ultrasound(HIFU) treatment in unr esectable advanced pancreatic carcinoma.Methods Thirty-eight patients with unresectable pancreatic cancer received HIFU treatment.After treatment,the changes of laboratory tumor markers examination results,pain score,life quality score and survival situation were recorded.Results Among 35 patients with pain symptom before HIFU treatment,pain was relieved after HIFU treatment in 28 cases,the remission rate was 80.0%.The CA19-9 and CEA levels after HIFU therapy were obviously reduced compared with before treatment.The imaging examination showed the coagulation necrosis in HIFU-treated area.It was found the tumor volume was obviously shrunk during follow-up period.The median survival period was extended to (12.9 ± 6.6) months.Conclusion The HIFU treatment can effectively improve the life quality in the patients with unresectable pancreatic cancer and extends their survival period.
5.Apoptosis Induction by Sijunzi Decoction in Human Gastric Cancer Xenografts in Nude Mice

Objective: The current study was designed to examine whether Sijunzi decoction could induce apoptosis in human gastric cancer xenografts in nude mice. Methods: A human gastric adenocarcinoma cell line SGC-7901 was grafted into 30 nude mice. The animals in two experimental groups received either Sijunzi decoction for 40 days or 5-fluorouracil (5-FU) for 6 days beginning 2 days after grafting. The control animals received saline according to an identical schedule. The animals were sacrified 41 days after grafting. The therapeutic effect was determined by measuring of tumor size and tumor weight. Apoptotic indices were examined with terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate fluorescence nick end labeling (TUNEL) method and flow cytometry analysis. Morphological changes were observed by electron microscopy. Results: Comparing with controls, tumor growth was significantly inhibited by Sijunzi decoction ( 34.33% , P<0.05) or 5 FU( 50.00% , P< 0.05) . Apoptotic index(AI) was significantly higher(16.24% ± 3.21% as determined by TUNEL and 11.38% ± 6.46% by FACScan) in Sijunzi decoction treatment group than that in the control group (TUNEL:2.63% ± 1.03% ,P< 0.01; FACScan:7.15% ± 1.32% ,P< 0.05); while there was no significant increase of AI in 5 FU treatment group. Cell shrinkage, nuclear chromatin condensation, formation of membrane blebs and apoptotic bodies were frequently observed on tumor sections in Sijunzi decoction group, while only few apoptotic cells/bodies and necrotic cells were found in the 5-FU treatment group, and in control group cancer cells were in proliferation. Conclusions: The data demonstrated enhanced apoptosis in human gastric cancer xenografts in nude mice after treatment with Sijunzi decoction, suggesting that Sijunzi decoction may be a potential anticancer agent for gastric cancer treatment.
6.Clinical Study on Combined Phacoemulsification and Intraocular Lens Implantation/Trabeculectomy with Mitomycin C.

This study was undertaken to evaluate the outcome of combined phacoemulsification, intraocular lens implanation, and trabeculectomy with Mitomycin C. We reviewed 39 eyes in 36 consecutive cases who had been undergone triple procedure for coexisting cataract and glaucoma from January 1993 to October 1995 at Chonnam university hospital. The mean follow-up period was 8. 8 months. Preoperatively, most patients had a visual acuity of worse than 0.4, but the visual acuity improved postoperatively in 20 eyes (51.0%) achieving the visual acuity better than 0.5 at 6 months after the surgery. The mean preoperative intraocular pressure was 31.1 +/- 12.9mmHg, and the mean postoperative IOP was 16.5 +/- 5.4mmHg at 6 months follow-up. The mean number of antiglaucoma medication was 1.87 preoperatively, and 0.28 postoperatively. The postoperative complications were posterior capsular opacity (7 eyes), hyphema (2 eyes), shallow anterior chamber (3 eyes), transient hypotony without macular edema (2 eyes). There was no statistically significant difference in intraocular pressure control at different concentration of Mitomycin C (0.2 and 0.4mg/ml). We conclude that the use of Mitomycin C in combined procedure provides good short-term(6 months) results regarding intraocular pressure control and visual recovery.

8.Comparison of the Hoffer Q and SRK-II fomulas in IOL Power Calculation.

Although most available IOL power calculation formulas perform accurately for eyes of average axial length, they have been shown to be inaccurate for eyes that have unusually short and long axial length. To compare the prediction accuracy of new Hoffer Q with SRK-II formula, we reviewed, retrospectively, a series of 225 ECCE with PCL implantation cases, including 106 Hoffer Q group and 119 SRK-II group, each of which was further divided into subgroups based on axial length. The Hoffer Q formula overestimated the refraction and resulted in more myopic shift and was less accurate than SRK-II in all length eyes, except short length eyes. In short length eyes, both formulas had similar mean error and mean absolute error but Hoffer Q formula was more accurate than the SRK-II in +/- 0.5D prediction proportion.

9.Clinical Study on Primary Acute Angle Closure Glaucoma.

In order to study the clinical characteristics of primary acute angle closure glaucoma, we analyzed the data of 72 eyes from 63 patients with the disease from January 1, 1990 to December 31, 1993 to the Department of Ophthalmology of Chonnam University Hospital. The mean age of the patients was 63.3 years and female and male ratio was 5.9:1. And the mean IOP at first visit was 53.2 mmHg. There was no significant difference of rop between right and left eyes, and 11 patients had the disease in both eyes. 38% of the patients developed the disease in winter time. The development of the disease was closely associated with physical illness and emotional stress. 39 eyes(49.3%) of them was required filtering surgery because medication and laser treatment was insufficient to lower IOP.

10.Isolation and purification of alpha-glycerophosphate oxidase in a polyethylene glycol/(NH4 )2SO4 aqueous two-phase system.

Alpha-glycerophosphate oxidase (alpha-GPO) from Enterococcus casseli flavus was successfully isolated and purified by using polyethylene glycol (PEG)/(NH4)2SO4 aqueous two-phase system (ATPS). The results showed that the chosen PEG/(NH4)2SO4 ATPS could be affected by PEG molecular weight, pH, concentration of PEG and (NH4)2SO4, and inorganic salt as well as additional amount of crude enzyme. After evaluating these influencing factors, the final optimum purification strategy was formed by 16.5% (m/m) PEG2000, 13.2% (m/m) (NH4)2SO4, pH 7.5 and 30% (m/m) additive crude enzyme, respectively. The NaCl was a negative influencing factor which would lead to lower purification fold and activity recovery. These conditions eventually resulted in the activity recovery of 89% (m/m), distribution coefficient of 1.2 and purification fold of 7.0.
