1.Logistic regression analysis of risk factors of postpartum hemorrhage of cesarean section

Objective To investigate the risk actors of postpartum hemorrhage of cesarean section,in order to provide the basis for the prevention of hemorrhage after cesarean section.Methods A case-control study was conducted in 100 cases with postpartum hemorrhage of cesarean section and 200 cases without bleeding of cesarean section.The clinical data were compared and analyzed.The single factor and multiple factors of the Logistic analysis was used to analyze the risk factors of postpartum hemorrhage of cesarean section.Results Fetal macrosomia (OR =3.678,95% CI:1.456-5.345),placenta adhesion (OR =4.264,95% CI:1.556-5.646),placenta praevia (OR =2.754,95% CI:1.754-5.456),multifetal pregnancy (OR =3.964,95% CI:1.274-4.453) were the risk factors of postpartum hemorrhage after cesarean section.Conclusion Evaluation of the above risk factors before cesarean section was important for prevention of postpartum hemorrhage.
2.Inhibitory effect of BEZ235 on human prostate carcinoma in vitro

Objective:To determine effects of BEZ235,an inhibitor of phosphoionsitol-3-kinase (PI3K)/ mTOR,on the cell proliferation and migration in human prostate carcinoma lines including RWPE-1,PC3,and DU 145 cells.Methods:Viability of RWPE-1,PC3,and DU145 cells was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay,while cell migration was analyzed by wound healing assay.Western blot and immunofluorescence were used to examine the changes of relevant protein expression.Results:The proliferation of PC3 and DU145 cells was effectively inhibited by BEZ235 (P<0.01),whereas RWPE-1 was not obviously inhibited.Invasion and migration of PC3 and DU145 cells were attenuated by BEZ235 via EMT pathway.Conclusion:The PI3K/mTOR dual inhibitor BEZ235 shows substantial anti-tumor activity in human prostate carcinoma lines of PC3 and DU145 cells,which may be involved in the EMT pathway.
3.Short-term influence of LASIK on anterior and posterior corneal asphericity

Background Laser in situ keratomileusis (LASIK) is frequently performed to reduce or eliminate myopic refractive error.Some patients complain of a loss of visual function after surgery even when they have 20/20 visual acuity.One of the reasons is the change of asphericity of the cornea.Objective This study attempts to investigate the short-term changes of the anterior and posterior corneal asphericity after LASIK.Methods One hundred and seven myopic eyes of 54 subjects with the equivalent spherical diopter of -1.30 to -7.50 D who have received LASIK were enrolled in this prospective study.The Q-values of the posterior corneal surface for different corneal diameters (6mm,7mm,8mm and 9mm) were measured with Pentacam preoperatively and 1 month postoperatively.The correlations between Q-value,Q change (△Q),and the mean preoperative spherical equivalent refraction (SE),central corneal thickness (CCT),central ablation depth (AD) and residual bed thickness were investigated.Written informed consents were obtained from all the subjects prior to the clinical trial.Results The Q-values of the anterior and posterior corneal surfaces gradually decreased to negative values with the increase of corneal diameter in myopic eyes.Weak correlations were found between the asphericity of the anterior and posterior corneal surfaces from diameters of 6mm,7mm,8mm and 9mm (r=0.227,0.288,0.303,0.389;P=0.019,0.003,0.002 and 0.000,respectively).No statistically significant correlation was found between Q-value and the diopter of refractive error (P>0.05).Both the anterior and posterior corneal Q-values varied toward the positive direction except that in the 9mm area of the posterior corneal surface at postoperative 1 month(t=-1.495,P=0.138).The increase of the anterior corneal asphericity (△Q) was more obvious than that of the posterior corneal surface and showed a positive correlation with ablation depth and a negative correlation with residual bed thickness.However,no statistically significant correlation was seen between △Q and these two parameters in the posterior corneal surface (P>0.05).Conclusion The shape of the anterior and posterior corneal surface shows more prolateness as the increase of corneal diameter in myopic eyes.Both the anterior and the posterior corneal surfaces have an oblate shift within the ablation zone 1 month after LASIK.
4.Exploration of the Medication Regularity of Traditional Chinese Medicine for Obesity Based on Text Mining Techniques

This study aimed at analyzing the medication regularity based on differentiation in traditional Chinese medicine (TCM) for losing weight using text mining technique.All the references over losing weight were retrieved in CNKI,Wangfang Database,VIP Database and Pubmed.The drugs from the references were classified in accordance with drug property,drug flavor,channel tropism and drug efficacy.Frequency and constituent ratio of a single drug in TCM prescriptions for losing weight were put into analysis using chi square test and factor analysis to find out the medication regularity.It was found that the properties of TCM drugs in the prescriptions contained both cold and warm,while the flavors of the drugs involved pungent,sweet and light.The channel tropism of the drugs mainly belonged to spleen meridian,liver meridian,stomach meridian and lung meridian.They were mostly tonic,relieving,blood-activating,qi regulating,inhibiting-damp and antipyretic drugs.Through factor analysis we found that the common formula compatibilities were concluded as:cassia seed,lotus leaf,hawthorn,salvia miltiorrhiza,polygonum cuspidatum and radix polygonum multiflorum;capillary artemisia,epimedium herb,stephania tetrandra and ligusticum wallichii;dried tangerine peel,pinellia ternata and poria cocos;plantain seed,pericarpium arecae and selfheal;paeonia lactiflora,angelica sinensis,scutellaria baicalensis and ligusticum wallichii;poria cocos,cassia twig,atractylodes and glycyrrhiza;immature bitter orange and bark of magnolia;radix bupleuri,lycium chinensis and jujube;Chinese yam and coix seed;and astragalus,pueraria lobata and polygonatum.In conclusion,formula compatibility mainly combined syndrome differentiation with disease differentiation for the treatment of obesity in clinic,using the drugs belonging to liver meridian,spleen meridian,stomach meridian and lung meridian with the flavors of sweet,bitterness or pungent and the nature of both warm and cold.
5.The risk predictive value of high sensitivity C-reactive protein level for new hemorrhagic stroke events

Objective To study the risk prediction for new intracerebral hemorrhage (ICH) with high sensitivity C-reactive protein ( hs-CRP) level. Methods In a retrospective, nested, case-controlled study, 323 cases of ICH were identified and matched with 646 controls. The hs-CRP levels at baseline were compared between the two groups. The relevance of different hs-CRP levels and the risk of ICH were analyzed. Results The ICH group had a higher median hs-CRP levels (1.10 mg/L) as compared with the control group (0. 66 mg/L) with significant difference ( P<0.01 ). In addition, the increase of risk associated with hs-CRP levels was primarily observed in the individuals with the highest quartile of hs-CRP levels(>2.12 mg/L). These patients had an increased risk of ICH (OR 2. 58, 95% CI 1. 77 to 3. 76) as compared with those in the lowest quartile(≤=0.30 mg/L). Individuals with basiline hs-CRP levels above the specified cut point of 3 mg/L ormore and those in the 80th percentile were at a markedly increased risk of ICH (for specified cut point of 3 mg/L,0R2.26, 95% CI 1.60-3.20, P<0.01; for 80th percentile, OR 2.24,95% CI 1.60-3.13, P <0.01, respectively). Conclusions Risk of ICH might be predicted with the level of hs-CRP. With the increase of hs-CRP level at baseline, the risk of ICH was increased.

Objective To evaluate the reliability of HARSHAW-3500 thermoluminescence dosimetry system by testing its performances.Methods HARSHAW-3500 thermoluminescence dosimetry system had its performances tested and evaluated according to Verification regulation of thermoluminescence dosimetry systems used in persontal and environmental monitoring forXandgammaradiation(JJG 593-2006),Testingcriteriaofpersonneldosimetryperformanceforexternalexposure (GBZ 207-2016),Specifications for individual monitoring of occupational external exposure (GBZ 128-2016) and Thermoluminescence dosimetry systems for personal and environmental monitoring (GB/T 10264-2014),such as batch homogeneity,repeatability,linearity,incidence angle response,stability,energy response and scale factor,quantity inspection,residual dose,detection limit and etc.Results Testing results of various performance indicators proved to be within the limits according to national and industrial standards.Conclusion HARSHAW-3500 thermoluminescence dosimetry system conforms to the requirements for radiation dose measurement.It is beneficial to the improvement of quality and performance of thermoluminescence dosimetry by performances analysis and evaluation.
8.Expression of HOXC13 in ameloblastoma.

OBJECTIVETo study the expression of HOXC13 mRNA in ameloblastoma (AB), and to investigate its biological significance.
METHODSHOXC13 mRNA was examined in 47 cases of AB (primary AB 29 cases, recurrent AB 14 cases, malignant AB 4 cases). 2 cases of fibrous dysplasia of bone, 10 cases of keratocystic odontogenic tumor (KCOT) and 7 cases of normal oral mucosa were selected as control.
RESULTSThe positive rates of HOXC13 mRNA in AB, KCOT, and normal oral mucosa were 97.9% (46/47), 7/10 and 3/7, respectively. There was a significant difference among AB, OKC and normal mucosa (chi(2) = 21.665, P = 0.001). For HOXC13, the keratinizing cells and granulizing cells in AB were negative, some fibroblasts were positive, 2 cases of fibrous dysplasia of bone were positive.
CONCLUSIONSHOXC13 was highly expressed in AB. The expression of HOXC13 mRNA in AB had heterogeneity, which could improve the epithelial proliferation, and its loss may lead to the cornification and degeneration of epithelial cells.

9.Role of dysregulation of Bim in resistance of melanoma cells to endoplasmic reticulum stress-induced apoptosis.

OBJECTIVETo establish a model of ER stress-induced apoptosis with tunicamycin and to examine whether Bim is dysregulated and its potential role in resistance of melanoma cells to apoptosis under endoplasmic reticulum (ER) stress.
METHODSA model of ER stress-induced apoptosis was established with tunicamycin. Apoptotic cells were quantitated using the annexin V/propidium iodide method by flow cytometry. Hoechst staining was also used to confirm the apoptotic cell death. Western blotting was used to measure the activation of caspase-3 and -9, and the expression of Bim, GRP78, CHOP, and Foxo1 at the protein level. The expression of Bim, CHOP and Foxo1 at the mRNA level was quantitated by qPCR. The siRNA technique was used to inhibit the expression of Bim.
RESULTSTreatment of the melanoma cells with tunicamycin did not induce significant apoptosis and activation of caspase cascade, whereas it caused marked activation of caspase-3 and -9, and apoptosis in HEK293 cells which were used as a control. With exposure to tunicamycin (3 µmol/L) for 12, 24, 36 hours the Bim protein levels were not increased in Mel-RM and MM200 cells. Its mRNA levels were 0.37 ± 0.05, 0.13 ± 0.02 and 0.02 ± 0.01 in Mel-RM cells, while 0.41 ± 0.06, 0.16 ± 0.04 and 0.21 ± 0.03 in MM200 cells, respectively. The expression of Bim mRNA was significantly reduced compared with that in the control groups of the two cell lines (P < 0.01). siRNA knockdown of Bim protected HEK293 cells against activation of caspase-3. The cell apoptosis of Bim siRNA group was (5.69 ± 0.38)%, significantly lower than that of the siRNA control group (40.32 ± 1.64)% and blank control group (35.46 ± 2.01)% (P < 0.01). In the melanoma cells after exposure to tunicamycin (3 µmol/L) for 6, 12, 24, and 36 hours the transcription factor CHOP at mRNA level were significantly increased and the expressions at protein level were also up-regulated. The expressions of another transcription factor Foxo1 at mRNA level significantly decreased and the expressions at protein level were down-regulated, too.
CONCLUSIONSThe lack of Bim up-regulation contributes to the resistance of melanoma cells to ER stress-induced apoptosis and may be a mechanism by which melanoma cells adapt to ER stress conditions. Transcription factors CHOP and Foxo1 may be responsible for the dysregulation of Bim in melanoma cells upon ER stress.

10.The regulating effect of auricle electrical stimulation on the cardiac vagus nerve in rats

Objective To compare the regulating effect of electrically stimulating different parts of the auri-cle on the cardiac vagus nerve in rats, and to explore the basic neural mechanism. Methods The tragus, concha auriculae and helix of 24 male Sprague-Dawley rats were stimulated at different intensities ( 0-16 mA) and with differ-ent durations ( 0-15 min) and any changes in the heart rate were observed. One week later, the rats were randomized into a tragus injection group, a concha auriculae injection group, a helix injection group and a control group, each of 6. The rats of the first three groups were injected with 2 μL of cholera toxin subunit B conjugate AF555 ( CTB-AF555) at the right auricle, while the control group was injected with the same amount of aseptic phosphate-buffered saline at the right tragus. Five days later, all of the rats were sacrificed and their right superior and inferior ganglia and the whole bulbus medullae were resected to observe the fluorescent labeling sites. Results The rats'heart rate declined with longer and more intense stimulation of the tragus or concha auriculae, but not with stimulation of the he-lix. With stimulation of the same duration, a significant decrease was observed in the heart rate when the tragus and concha auriculae were stimulated at 10, 12, 14 or 16 mA compared with when the helix was stimulated at the same intensities. The heart rate when the concha auriculae was stimulated at 12 mA was significantly slower than when the tragus was stimulated at the same intensity. At identical stimulus intensities, the heart rate slowed significantly more when the tragus was stimulated for 6 to 15 minutes and the concha auriculae for 4 to 15 minutes compared with stimu-lating the helix for the same length of time. And compared with stimulating the tragus for 6 to 10 minutes, the heart rate decreased significantly more when the concha auriculae was stimulated for the same length of time. All of the rats in the tragus and concha auriculae injection groups displayed nerve tracer in their superior and inferior ganglia. In the tragus injection group, CTB-AF555 was observed in the nucleus tractus solitarius ( NTS) of 3 of the 6 rats. In the concha auriculae injection group it was observed in 4 of the 6. In the helix injection group, CTB-AF555 was observed in the nucleus of the spinal tract in 5 of the 6 rats, but no nerve tracer was found in their superior or inferior ganglia or in the NTS. Conclusion Electrical stimulation of the tragus and concha auriculae can regulate the functioning of the cardiac vagus nerve, but stimulating the helix cannot. This is partly because the nerve signals in tragus or concha auriculae stimulation and the cardiac sensory nerve signal are integrated in the inferior ganglion and then analyzed and processed in the bulbar center to monitor the heart.
