Objective To explore the efficient way to purify and to enrich umbilical cord mesenchymal stem cells which have abilities to differentiate into chondrocytes. Methods Umbilical cord was acquired from new born infants and chopped into 1 mm2 cubics, then they were pretreated with colleganase and trypsin so as to get the mixed cell suspension. CD45(-),CD90(+) cells were acquired from the mixed cell suspension via FCM sorting and were cultured in the medium as umbilical cord mesenchymal stem PO cells. The PO cells were detached and passaged by multiplex collagenase NB4 and the positive rate of CD90 and CD45 of the P1-P3 cells were observeed via FCM ; the P3 cells were induced to differentiated into chondrocytes. Results FCM results showed that the positive rate of CD90 on P1, P2, F3 was (80.86 ± 7.85)%,(95.86± 3.28)%,(97.15± 1.43)%, while the positive rate of CD45 on P1, P2, P3 cells was (2.53 ±0.28)% and (0.97 ± 0.48)% and (0.05 ± 0.01)% respectively; P3 cells had differenciated into chondrocytes. Conclusion FCM presorting and Multiplex collagenase detachment procedures can swiftly acquire highly purified human umbilical cord mesenchymal stem cells which have the ability to differentiate into chondroeytes.

Background Streptozotocin (STZ)-induced type 1 diabetic model is an acceptable model and is often used to the study of diabetic retinopathy (DR).However, the model is often established using retinal digest preparation in vitro in albino rats, and therefore it is difficult to evaluate the change of retinal vessels by fundus fluorescein angiography (FFA) in vivo.Recently, pigmented rats are being used as the DR model. But the study on the comparison between albino rat model and pigmented rat model is seldom. Objective This study was to observe and compare the manifestations of FFA and retinal digest preparation in early diabetic pigmented diabetic rat and albino diabetic rat,and to select the right model of DR using FFA.Methods Type 1 diabetic models were induced in 15 six-week-old health male BN rats and 15 six-week-old health male SD rats by the injection of STZ (55 mg/kg) via tail vein. The models were thought to be successful in the rats with the blood glucose level ≥ 16.7 mmol/L.The right eyes of the rats were extracted 6 weeks after injection of STZ.Lens were examined by slit lamp microscope.Retinal vascular changes were examined by fundus photography,FFA and periodic acid Schiff staining of retinal digest preparation. The manifestations of FFA and retinal digest preparation were contrasted between BN rats and SD rats. The number of eyes with lens opacification was compared by Chi-square test and the ratio of vascular endothelial cells and perithelial cells (E/P) was compared between BN rats and SD rats. The use and care of experimental animals complied the Statement of Ethic Committee of Tianjin Medicine University. Results Six weeks after injection of STZ,11 BN rats and 10SD rats were included in this study. The blood levels were (24.73±2.98) mmol/L and (22.36±3.65) mmol/L in BN rats and SD rats, respectively, without significant difference between the 2 types of rats (t =7.873,P>0.05).Lens opacification occurred in 6 BN rats and in 5 SD rats (P=0.717).FFA showed the clear retinal vascular under the brown background.Evident vessel disorder and fluorescence leakage like background stage of DR were seen in 9 eyes. However,in the SD rats, retinal vessel abnormality could not exhibited owing to the interference of choroid fluorescent light from choroidal vessels and vortex vein.Retinal digest preparation exhibited that unevenness of vessel diameter, decrease of perithelial cells and increase of endothelial cells in both types of rats. The E/P values were 11.50±3.68in the BN rats and 12.86±3.94 in the SD rats, without significant difference between them (t=9.785,P>0.05).Conclusions The abnormality of fundus vascular morphology can be better displayed in pigmented diabetic rats than albino rats by FFA in vivo.

Objective To investigate the changes of plasma adrenodullin(ADM) and C-type natriuretic peptide(CNP) level in children with chronic heart failure(CHF) and its clinical implications. Methods Forty-two children with CHF were collected. The patients suffered from dilated cardiomyopathy, congenital heart defects, and other heart diseases.According to a modified scoring system described by Ross and Reithman,16 patients were classified as class Ⅱ,14 as class Ⅲ,and 12 as class Ⅳ.Plasma levels of ADM and CNP were measured by radioimmunoassay assay in these patients and 11 healthy children.Echocardiography was performed to measure left ventricular function and the ratio of E/A.Results Plasma ADM and CNP levels of CHF patients were significantly elevated as compared with those of the control subjects [(218.27?106.53) ng/L vs(74.39?53.99) ng/L,P=0;(190.27?108.38) ng/L vs(92.59?(59.46) ng/L),P0.05).ADM levels were elevated with the advancing severity of CHF determined by a modified scoring system described by Ross. However, the plasma CNP levels in the normal state wasn′t significantly different from those observed in class Ⅱ.Likewise, the plasma CNP level in the class Ⅲ was not significantly different from that observed in class Ⅱ.Conclusions ADM and CNP might play a compensatory and defensive roles in the pathophysiology of the pediatric CHF.ADM may be a biochemical marker for evaluation the severity of the chronic heart failure in children, but also a new prognostic indicator of this syndrome.

OBJECTIVETo observe the preconditioning cardioprotection of atorvastatin (ATV) in rabbits underwent 40 min ischemia and 240 min reperfusion and to explore related mechanisms.
METHODSThe rabbits were randomized divided into Control group, ATV group (10 mg.kg(-1).d(-1) for 3 days before ischemia), ATV plus iNOS inhibitor S-methylisothiourea sulfate group (ATV + SMT group), SMT group, ATV plus mito K(ATP) channel blocker 5-hydroxydecanoate group (ATV + 5-HD group) and 5-HD group (n = 16 each group). The infarction size, CK-MB, LDH-1, nitric oxide synthase and mitochondrial ATP synthesization capacity ([ATP] m) were determined at the end of reperfusion.
RESULTSInfarction size, CK-MB, LDH-1 were decreased by 26.3%, 31.4%, 19.1% and iNOS, [ATP] m increased by 102.6%, 46.8% post ATV compared to control group (all P < 0.05) and these effects could be blocked by cotreatment with SMT and 5-HD except the iNOS was not affected by 5-HD.
CONCLUSIONThe atorvastatin preconditioning exerted cardioprotection by upregulating iNOS and activating mito K(ATP).

Objective To investigate the diagnostic value of serum ferritin in children with systemic onset juvenile idiopathic arthritis (SO-JIA). Methods Fifty-seven patients with fever of unknown origin (rectal temperature>38.5 ℃ ) over two weeks and hospitalized in our general medicine ward longer than one week were enrolled in this study. Patients were recorded the course of fever, elinieal symptoms and signs including rash and swollen joints/arthralgia, laboratory tests including complete blood cell count, C-reactive protein, erythroeyte sedimentation rate, lactate dehydrogenase and the level of serum ferritin. SPSS 10.0 was used for statistical analysis. Results Two of 57 patients could not be diagnosed before discharge. The other 55 patients whose diagnosis was confirmed were divided into four groups. Twenty-five patients were SO-JIA,12 patients had hematologic or oncological diseases, 12 patients had infectious diseases and 6 patients had other rheumatic diseases. The level of serum ferritin was significantly higher (P<0.01) in SO-JIA group than in other groups. Moreover, the levels of serum ferritin in SO-JIA group were all higher than normal and the levels of serum ferritin in 76% of SO-JIA group were more than five-fold elevation. Four cut-off levels of serum ferritin level for the diagnosis of SO-JIA were selected based on clinical practice and ROC curve. When the cut-off levels of serum ferritin were 328.25, 529.50, 731.05 ng/ml and 1121.10 ng/ml, the sensitivity for the diagnosis were 100%, 88%, 72%, and 64% respectively, the specificity were 77%, 87%, 90% and 100%,respectively. Conclusion Serum ferritin is valuable for the diagnosis of SO-JIA and 529.50 ng/ml may be a good cut-off level
