1.Determination of Taurine in Mactra Veneriformis by Pre-column Derivatization RP-HPLC

Objective To determine the content of taurine in Mactra veneriformis by pre-column derivation HPLC.Methods Ultrasound method was used for the extraction of taurine from Mactra veneriformis.In alkaline condition,amino acids reacted with phenyl isothiocyanate and resulted in the derivatives of benzene isothiocyanate-amino acid complex.Kromasil C18 column(4.6 mm ?250 mm,5 ?m) was used and the absorbance at 254 nm was measured.The mobile phase consisted of sodium acetate solution,water and methanol.Results Corresponding chromatographic peaks of taurine were obtained in extract sample and the control.The linear range of taurine was from 10.04 ?g?mL-1 to 50.20 ?g?mL-1,and the recovery was 97.95 %(RSD=1.41%,n=9).Conclusion This method is simple,accurate and reproducible.The seasonal variation range of taurine level is from 10.56 mg/g to 20.91 mg/g,and the taurine level arrived to the peak in June.
2.The characteristics of speech,phone and cognition development of children with mild hearing loss and the effect of early intervention

Objective To explore the characteristics of speech,phone and cognition development of infants and toddlers with mild hearing loss and to study the effect of early intervention.Methods Speech and cognition abilities of 59 children with mild hearing loss,aged 8 months to 2 years and 10 months old were assessed and 34 normal hearing children,aged 8 months to 2 years and 9 months old were compared.Eighteen children with hearing loss were fitted with hearing aids and received speech therapy for 6 months.The participants' speech and cognition development were evaluated with Gesell developmental scale.The phone test was adopted to assess the speech sound development in 59 children with mild hearing loss.Results Of the 59 children with mild hearing loss,37 were judged with normal speech development quotients(DQ),8 with abnormal speech DQ,14 suspect of abnormal speech DQ.The hearing loss in cases with abnormal speech DQ was more serious than that in cases with normal speech DQ(P<0.05).The speech DQ and personal-social behavior DQ of children with mild hearing loss were significantly lower than normal children(P<0.01).The speech DQ and personal-social DQ on Gesell developmental scales were significantly higher in 18 children received early intervention,compared with those who did not receive intervention (P<0.01).Six cases with mild hearing loss demonstrated phone errors,but no abnormal manifestations in tone.After early intervention the phone of those with mild hearing loss improved significantly.No significant difference in gross motor,fine motor and adaptive ability was observed between the children with mild hearing loss and those with normal hearing(P>0.05).Conclusion The speech ability of children with mild hearing loss is significantly lower than those with normal hearing.Some mild hearing loss children show phone errors.Mild hearing loss affect the adaptation and communication abilities of mild hearing loss children significantly.Early intervention can significantly improve the speech,phone,adaptation and communication abilities of children with mild hearing loss.
3.Progress in study of a novel protein Nischarin

Nischarin is a novel protein interacting with integrin ?5 subunit.A series of studies have indicated that Nischarin plays an important role in inhibiting tumor cells migration and invasion.Meanwhile it may also serve as the functional I1-imidazoline receptor.To study this protein will be in favour of understanding the biological mechanisms of inhibiting cell migration and developing novel drugs such as inhibitors for tumor invasion.The article mainly reviews the recent progress in studies on Nischarin.
4.Effects of psychological stress on pathogenetic condition and rapeutic effect of ulcerative colitis

AIM: To investigate the effects of psychological stress on ulcerative colitis rats and to explore the mechanisms of this;meanwhile to investigate the effects of anxiolytic drug on symptoms of ulcerative colitis rats under psychological stress and the mechanisms of it.METHODS: eighty male Wistar rats were randomly divided into four groups(n=20): UC control group;UC stress group;UC+clonazepam group;UC stess+clonazepam group.After UC model was established with immunization,these rats were fightened by cat to set up psychological stress model.During establishing stress model,each groups were respectively received physiologic saline or clonazepam by intragastric administration.After 15d treatment,all rats were sacrified simultaneously.Colon mucosal inflammationand damage were assessed by measuring colon mass,morphologic damage score,colonic MPO and NO activity levels.Morphologic damage score was examined under microscope.Colonic MPO and NO was measured by spectrophotometric method.Serum hydrocortisone was determined by radioimmunoassay(RIA).RESULTS: Morphologic damage score,colonic MPO and NO,serum hydrocortisone of UC stress group were significantly increased compared with those of UC control group;morphologic damage score,colonic MPO and NO,serum hydrocortisone of UC+clonazepam group were decreased markedly vs UC stress group;Little difference was observed between UC+clonazepam group and UC control group.CONCLUSION: Psychological stress can aggravate symptoms of UC.Although benzodiazepine have no anti-inflammatory action,it relieve effects on internal organs of psychological stress and aggravation of symptoms of UC by to effect limbic system and formatio reticularis.
5.Value of hyomental distance measured when the hyoid is located using ultrasound in predicting difficult airway

Objective To evaluate the value of hyomental distance (HMD) measured when the byoid was located using ultrasound in predicting the difficult airway.Methods Two hundred and thirteen patients of both sexes,aged 18-64 yr,with body mass index of 14-34 kg/m2,with interincisor distance >3 cm,with atlanto-axial joint mobility > 90°,of Mallampati class Ⅰ-Ⅳ,of American Society of Anesthesiologists physical status Ⅰ-Ⅲ,scheduled for elective surgery requiring endotracheal intubation under general anesthesia,were enrolled in the study.The HMD was measured by fingers (finger breaths in width,HMDf) and by rulers (HMDr) when the hyoid was located artificially and when the hyoid was viewed on ultrasound (HMDu) before anesthesia induction.After anesthesia induction,the difficult airway was defined as Cormack-Lehane grade 3 or 4 assessed under direct laryngoscope during intubation.The hyoid viewed on ultrasound was considered as the criterion,and the error rate was calculated when the hyoid was located artificially.Youden Index was used to determine the optimal value in predicting difficult airway,and the specificity and sensitivity of HMDf,HMDr and HMDu in predicting difficult airway were calculated based on this criterion.Results The error rate was 18.8% when the hyoid was located artificially.The specificity and sensitivity of HMDu in predicting difficult airway were 64.6% and 91.5%,respectively.The specificity and sensitivity of HMDr in predicting difficult airway were 62.5% and 84.8%,respectively.The specificity and sensitivity of HMDf in predicting difficult airway was 29.2% and 98.2%,respectively.Conclusion The HMD measured when the hyoid is located using ultrasound can accurately predict the difficult airway.
6.Immunogenic cell death of human osteosarcoma cells MG-63 induced by capsaicin

Objective To investigate the effect of capsaicin and cisplatin on the proliferation and immunogenic cell death of human osteosarcoma cells.Methods MTT assay was used to examine the growth inhibiting effects of capsacin and cisplation on MG-63 cells;Mitochondrial membrane potential (MMP) was used to investigate the apoptosis;flow cytometry was used to detect the expression of calreticulin (CRT) on the cell membrane,fluorescein enzymatic method was used to detect the release of ATP,ELISA was used to detect the secretion of high mobility group B1 (HMGB1).Results Capsaicin and cisplatin can inhibit MG-63 cells proliferation in the dose-dependent manner and induce MG-63 apoptosis(P<0.01).Only capsaicin can induce translocation of CRT from endoplasmic reticulum to the cell surface and release of extracellular ATP and HMGB1 (P<0.01).Conclusions Capsaicin can induce human osteosarcoma cells apoptosis and immunogenic cell death.
7.Effect of combined clopidogrel-aspirin treatment for acute cerebral ischemic infarction on a correlation between cerebral microbleeds and hemorrhagic transformation

Objective To investigate effects of combined clopidogrel-aspirin treatment for acute cerebral ischemie infarction on a correlation between cerebral microbleeds (CMBs)and hemorrhagic transformation(HT),so as to provide a new evidence for acute phase treatment of ischemic stroke with CMBs.Methods One hundred and forty-eighty patients with acute cerebral infarction meeting the inclusion criteria were consecutively admitted to our hospitals.All patients underwent susceptibility weighted imaging(SWI) to detect CMBs.Patients were classed into two groups:with and without CMBs and subdivided into brain lobe group,deep group and mixed group.The influence of CMBs or not and CMBs different positions on the post-infarction HT was compared.Logistic regression analysis was used to assess the relationship between HT and the related risk factors.Results The 142 patients finally were included in the study,with 64 patients without CMBs and 78 with CMBs.The detection rates of CMBs were 54.9%.Hypertensive prevalence rate(x2 =6.96,P =0.010)and the levels of uric acid (t =2.04,P =0.040) were higher in CMBs group than group without CMBs.The incidence rate of HT was 12.5 % (8 cases)in no CMBs group,and 21.8%(17 cases)in the CMBs group(x2 =2.09,P=0.150).6 in 15 patients(40.0%)patients experienced HT in lobar CMBs group;6 patients (12.5 %)experienced HT in 48 patients with deep CMBs group;5 patients(33.3%)experienced HT in 15 patients with mixed CMBs group.There was statistically significant difference in HT incidence rate(x2 =6.52,P=0.038)among the 3 groups.Lobar CMBs are more vulnerable for HT.Logistic regression analysis showed that atrial fibrillation(OR=6.48,95 % CI:2.45-17.19,P =0.000) and hyperglycemia (OR =1.02,95 % CI:1.43 1.94,P =0.020) were risk factors for HT,instead of CMBs(OR=1.95,95%CI:0.78-4.87,P=0.150).Conclusions CMBs do not increase the risk of hemorrhage transformation in cerebral ischemic infarction patients at acute stage with combined antithrombotic treatment.While,the double antithrombotic treatment used in patients with the lobar CMBs should be careful.
8.Neuroprotective Effect of Ginsenoside Rg1 on Oxidative Damage Induced by Oxygen-glucose Deprivation and Reperfusion in Cultured Hippocampal Cells

Objective To explore the effect of Ginsenoside Rgl on glutathion (GSH) level and glutathion peroxidase (GPx) activity after oxygen-glucose deprivation/reperfusion in cultured hippocampal cells. Methods The model of oxygen-glucose deprivation and reperfusion were established with the hippocampal neurons of rats. They were randomly divided into control group, model group and Ginsenoside Rgl treatment groups (5 μmol/L, 20 μmol/L, 60 μmol/L). The GSH level and GPx activity were measured 6 h after reperfusion. The apoptosis and the metabolic rate of methyl thiazolyl tetrazolium (MTT) were detected 24 h after reperfusion. Results Compared with model group, the GSH level, GPx activity, and metabolic rate of MTT improved (P<0.001), and the apoptosis decreased in the Ginsenoside Rgl groups (P<0.001) except with the dosage of 5 μmol/L (P>0.05). Conclusion Ginsenoside Rgl can protect the brain from ischemia by increasing the GSH level and GPx activity.
9.The effect of NP regimen plus concurrent irradiation on cancer cell survival of human lung adenocarcinoma

Objective To study the effect of NP regimen plus concurrent irradiation on cancer cell survival of human lung adenocarcinoma.Methods A549 and H1299 cells were served as the experimental objects and they were divided into irradiation group (A group),and vinorelbine tartrate injection plus cisplatin with concurrent radiotherapy group (B group).According to the different irradiation doses A group and B group were divided into A-0,A-4,A-8 group and B-0,B-4,B-8 group.MTT was performed to measure the effect of NP regimen plus concurrent irradiation on the cell proliferation,and FACS was used to detect the effect of NP regimen plus concurrent irradiation on apoptosis and P-gp expression.Results Except the cell inhibition rate of B-0 group and A-0 group had no significant difference,the cell inhibition rate of B-0,B-4 and B-8 group at 6,12,24 and 48 h were significantly lower than those of A-0,A-4 and A-8 group.The apoptosis rate of A-8 group [(45.34 ±4.77)%] was significantly higher than that of A-4 group [(20.19 ± 3.76)%] (t =7.17,P =0.006),the apoptosis rate of A-4 group was significantly higher than that of A-0 group [(4.01 ± 0.95)%] (t =7.23,P=0.019).The apoptosis rate ofB-8 group [(51.12 ± 3.37)%]was significantly higher than that of B-4 group[(36.18 ± 4.73)%] (t =3.59,P =0.037),the apoptosis rate of B-4 group was significantly higher than that of B-0 group [(14.78 ± 2.37)%] (t =9.00,P =0.003).The apoptosis rate of B-0 group [(14.78 ± 2.37)%] was significantly higher than that of A-0 group [(4.01±0.95)%] (t =7.31,P =0.018),the apoptosis rate of B-4 group was significantly higher than that of A-4 group (t =4.58,P=0.020),but there was no significant difference between B-8 group and A-8 group (t =1.71,P =0.185).The S cycle percentage of cells in B-0,B-4 and B-8 groups were significantly higher than those of A-0,A-4 and A-8 group (P <0.05).There were no significant differences among the P-gp fluorescence intensity of A-0,A-4 and A-8 group (P > 0.05).The P-gp fluorescence intensity of B-0 group was significantly lower than that of A-0 group (t =-3.45,P =0.041),the P-tp fluorescence intensity of B-4 group was significantly lower than that of A-4 group(t =-5.85,P =0.010).The P-gp fluorescence intensity of B-8 group was significantly lower than that of A-8 group (t =-8.07,P =0.015).The P-gp fluorescence intensity of B-4 group was significantly lower than that of B-0 group (t =-8.07,P=0.015),and there was no significant difference between the mean fluorescence intensity of B-4 group and B-8 group (t =-3.56,P =0.071).Conclusion NP regimen plus concurrent irradiation could significantly inhibit the proliferation and apoptosis of human lung cancer cells,and decrease the expression of P-gp in lung cancer cells.
10.Analysis and identification of water soluble components of water buffalo horn.

This study is to analyze and identify the water soluble components of water buffalo horn (Bubali Cornu, WBH), and also establish a method for investigating these components. Shotgun proteomic analysis identified proteins in WBH aqueous extraction: keratin, collagen, desmoglein, etc. Ultrafiltration and LC-MS/MS were used to separate and identify the peptides in WBH aqueous extract, as a result, identified peptides were mainly derived from nonspecific degradation products of keratin and collagen, which including C-terminal peptides and non C-terminal peptides. Hypoxanthine, uridine, guanosine, and adenosine were identified by comparing with the standards. The strategy in present study could be used in analyzing water soluble components of animal horn derived TCM. It provides a reference for investigation of the material basis of animal horn derived TCM.
