Objective To observe the efficacy of low dose aspirin(ASA) on the platelet aggregation in aged patients with ischemic cerebrovascular diseases(ICVD). Methods Platelet aggregation by arachidonic acid(AA, 500 ?g/ml), adenosine diphosphate (ADP,5 ?mol/L), epinephrine(EPI, 5 ?mol/L), and collagen (COL,2 ?g/ml) using Born's method was induced in 70 elderly with ICVD treated with ASA (40 mg/d) and in 30 untreated patients as well as in 50 healthy controls. Results A high coefficient of variation of the maximum aggregation rate induced by AA, COL and EPI were observed in the ASA (40 mg/d) treated group. Twenty-five out of the 70 patients in the ASA treated group (40 mg/d) were found that their platelet aggregation induced by AA were not markedly inhibited. Ten patients were randomly selected from the above 25 patients to receive an increasing dose to 80 mg/d. At repeated tests, 8 of these 10 achieved marked inhibition, and 2 of the 10 who showed no inhibition continued receiving an increasing dose to 100 mg/d and then achieved marked inhibition. Conclusions There is a great individual variation in response to low dose aspirin in aged patients with ICVD, and the laboratory evaluation of aspirin efficacy and its individualization is necessary in clinical practice.

Objective To determine whether Fudenine is a novel membrane protein. Methods Green fluorescence protein(GFP)was used to localize Fudenine in vivo. GFP, as a control, was targeted to cytoplasm.Epithelial cell, CBRH7919, and non-epithelial cell, L-6TG, were cultured and transiently transfected by using the lipofectamine reagent. After 48 h, intact cells were examined with fluorescence microscope for Fudenine. Results Reporter plasmid pEGFP-N1, as a control , was expressed and localized to cytoplasm. But Fudenine, driven by the cytomegalovirus promoterenhancer contained in the pEGFP-N1 vector, was overexpressed and targeted to cellular membrane. Conclusions Fudenine is a novel membrane protein. It may play the similar role with its homologues AC133 antigen and prominin in human and mouse.respectively. It might be involved in signaling transduction and regulate blood glucose metabolism in vivo.

OBJECTIVE:
To explore the effect of the cerebral thrombin preconditioning on the thrombin-induced brain edema, to detect the expression of tumor necrosis factor-alpha (TNF-alpha), and to analyse the relationship between TNF-alpha and the thrombin-induced brain edema.
METHODS:
Forty SD rats were randomly divided into a ST group and a TT group. The rats received 50 L saline (ST group) or 1 U thrombin infusion (TT group), and received the second infusion (10 U thrombin) 24 h later. The rats were sacrificed at 24 and 72 h after the second infusion in order to examine the changes of brain water and sodium contents as well as the expression of TNF-alpha in the brain.
RESULTS:
The brain water and sodium contents in the ST group were significantly higher than those on the TT group, and those on the 1st day were higher than those on the 3 th day. The positive expression of TNF-alpha and in the change of water content were identical in the TT group and the ST group.
CONCLUSION
Thrombin preconditioning can alleviate the thrombin-induced brain edema. The increase of TNF-alpha expression after thrombin treatment may be related to the thrombin-induced brain edema.

Journal of Zhejiang University. Science. B 2007;8(5):338-344
The fruit shape is important quantitative trait closely related to the fruit quality. However, the genetic model of fruit shapes has not been proposed. Therefore, in the present study, analysis of genetic effects for fruit shape traits (fruit length and fruit perimeter) in sponge gourd was conducted by employing a developmental genetic model including fruit direct effects and maternal effects. Analysis approaches of unconditional and conditional variances were applied to evaluate the genetic behavior of fruit shape traits at economical and physiological maturation times. The results of variance analysis indicated that fruit length and fruit perimeter were simultaneously affected by fruit direct genetic effects and maternal effects. Fruit direct genetic effects were relatively more important for fruit shape traits at whole developmental period. The gene expression was most active at the economical maturation stage (1 approximately 12 d after flowering) for two shape traits, and the activation of gene was mostly due to direct dominance effects at physiological maturation stage (13 approximately 60 d after flowering). The coefficients due to different genetic effects, as well as the phenotypic correlation coefficients, varied significantly between fruit shape traits themselves at various maturation stages. The results showed that it was relatively easy to improve fruit shape traits for industrial purpose by carefully selecting the parents at economical maturation stage instead of that at physiological maturation stage.
