2.Preventive Analgesic Effect of Preoperative Ketamine in Relieving Postoperative Pain after Laparoscopic Cholecystectomy

Objective To investigate the effects of ketamine in relieving postoperative pain after laparoscopic cholecystectomy(LC).Methods Forty patients undergoing selective LC from July to August 2006 were randomly divided into ketamine group and control group,with 20 patients in each group.Incision pain and non-incision pain was assessed at 1,2,4,6,12,24 h and once each day for the first 3 days postoperatively with visual analogue pain scale(VAS)and sedation scale.Adverse effects and analgesic requirements were recorded.Results Patients in ketamine group had significantly lower scores in incision pain and non-incision pain compared with those of control group(F=22.805,P =0.000;F=18.109,P=0.000).Incidence of postoperative nausea and vomiting was not significantly different(P=1.000)between ketamine(55%)and control group(60%).No significant difference was noticed in analgesic requirements between ketamine group(3 cases)and control group(9 cases)(P=0.082).Conclusions Preoperative administration of ketamine significantly improves postoperative analgesia after laparoscopic cholecystectomy,but does not reduce the incidence of postoperative nausea and vomiting.
3.Pathological Changes in Gastric Polyp Tissues and Its Background Mucosa under Gastroscopy

Objective To investigate the relationship between the pathology of gastric polyp tissues and the precancerous and malignant transformations of its background mucosa by endoscopic biopsy. Methods From 2003 to 2008,1825 patients were diagnosed with gastric polyps in our hospital by gastroscopy and pathological examination. We classified the lesions and then analyzed the relationship between pathological changes of gastric polyp tissues and the precancerous and malignant transformations of its background mucosa by endoscopic biopsy. Results In this study,the detection rate of gastric polyps was 4.3% (1825/42 003). The lesions included fundic gland polyps (FGP,62.5%,1141/1825),hyperplasia polyps (HPP,32.2%,587/1825),adenoma (0.8%,14/1825),inflammatory polyps (IP,1.7%,31/1825),and coincident polyps (two types of polyps were found in one case,2.8%,52/1825). The rates of intestinal metaplasia (IM,5.3%,31/587) and dysplasia (6.8%,40/587) in HPP tissue were higher than those in FGP tissues (0.2%,2/1141 and 0.4%,4/1141,P
4.Discriminating schizophrenia with combination of multiple tests:a pilot study

Objective: The pilot study was to explore the possibility of discriminating schizophrenia from the normal with combination of multiple tests. Methods: Forty medication-naive (25 cases) or medication-free for at least three weeks (15 cases) patients with schizophrenia and 41 healthy normal controls matched with sex, age, headedness, and education, were recruited in the study. The Structured Clinical Interview (SCID) was used to render the DSM-Ⅳ diagnoses for schizophrenia or schizophreniform disorders. The intrahemispheric coherences between the parietal and frontal electrode pairs (left: F3/P3; right: F4/P4) and interhemispheric coherences between frontal leads (F3/F4) and parietal leads (P3/P4) in the wide-band alpha frequency and peak frequency of photic driving were calculated at rest and during 1Hz light stimulation, respectively. Tests of exploratory eye tracking movement test (ETM), Wisconsin Card Sorting Test (WCST), and Continuous Performance Test (CPT) were finished in the same working day. Stepwise discriminant function analyses were chosen to re-classify the patients and controls. Results:In the discriminant analyses, 61.7% of the subjects could be correctly classified into groups of patients or normal controls using the EEG coherence alone, 77.27% with ETM measures alone, and 70.6% when WCST and CPT parameters were combined. The correct discriminate rate increased to 81.5% when all the above variables were taken into consideration.Conclusion:In consideration of the heterogeneity of schizophrenia, it is suggested necessarily to combine multiple tests to make discrimination of schizophrenia.
7.Construction and identification of small hairpin RNA expression plasmid targeting opticin gene in rat

Objective To construct small hairpin RNA (shRNA) expression plasmid targeting rat opticin gene. Methods Four pairs of opticin oligonucleotides were synthesized and inserted into the plasmid vector, resulting into four plasmids: shRNA-1, shRNA-2, shRNA-3 and shRNA-4. Then the four constructed shRNA expression vectors and empty vector were transfected into rat ciliary non-pigment epithelium (NPE) cells by lipofectmaine 2000. Non-transfected NPE cells were set as control group. The expression of opticin mRNA and protein were measured by Reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot respectively. Results The opticin mRNA expression of the shRNA-1,shRNA-2, shRNA-3, shRNA-4 group were decreased compared with the control group (F = 10. 239, P = 0. 000);the inhibitory rate were 85.7% ,62. 87% ,54.87% and 48.77% respectively. The opticin protein expression of the shRNA-1,shRNA-2, shRNA-3, shRNA-4 group were also decreased compared with the control group (F=17.870, P= 0.000);the inhibitory rate were 78.7%, 34.6%, 31.1% and 16.8% respectively.Conclusions The shRNA-1 expression plasmid has most potent inhibitory effect on opticin expression in rat ciliary NPE cells.
8.CLINICAL ANALYSIS OF 104 PATIENTS WITH INFECTIVE ENDOCARDITIS PATIENTS

patients with infective endocarditis were analyzed. Anong them, 75 were patients with congenital heart disease (72 1%), 21 were patients with rheumatic heart disease and 8 were patients without heart disease(7 7%). It is suggested that early diagnoses and treatment of CHD are the main pretective methods for reducing the incidence of IE; the incidence of fever, anaemia, embolism, hepatomegaly and splenomegaly, etc. declines notably; the possibility of excrescence invading aortic valve (44 2%) is higher than that of mitral valve (24 7%), and IE of aortic valve results in heart failure in a short while and the valve replacement should be done as early as possible; positive rate of blood culture declines because of extensive use of antibiotics; appropriate medical treatment combined with active surgical treatment plays an important role in the treatment of IE.
9.Study of chondrocyte on the layered scaffold under minic microgravity culture

[Objective]To evaluate the feasibility of the minic microgravity as a method and the layered cylindric collagen-chitosan-?-tricalcium phosphate composite as a scaffold for the cartilage tissue engineering after an observation of how it absorbs the chondrocytes and affects the cell behavior.[Method]The chondrocytes were isolated and multiplied in vitro,and then the chondrocytes were seeded onto the porous collagen-chitosan-?-tricalcium phosphate composite scaffold and were cultured in both minic microgravity and ordinary environment for 3 weeks.The effects of the composite scaffold on the cell adhesivity,proliferation,morphological changes and synthesis of the extracellularmatrix were observed by the growth curve,phase-contrast microscopy,histology,scanning electron microscopy and immunohistochemistry.[Result]The chondrocytes that adhered to the scaffold increased significantly and secreted extracellular matrix in the center of the porous scaffold around the chondrocytes under minic microgravity compared with ordinary environment.Immunohistochemistry of type Ⅱ collagen was positive.[Conclusion]The minic microgravity environment will be a good method for the cartilage tissue engineering.And the layered cylindric collagen-chitosan-?-tricalcium phosphate composite scaffold has a good cellular compatibility.It will be an ideal scaffold for the cartilage tissue engineering.
10.Effect of Atenolol on the Pharmacokinetics of Nitrendipine in Healthy Volunteers
