Next generation sequencing technologies,providing high speed and throughput that contribute greatly to DNA sequencing,have many applications in clinical virology,including identification of novel or unsuspected viruses,evaluation of human viromes,investigation of viral genetic diversity and evolution,monitoring of antiviral drug resistance,and analysis of virus-host interaction.Meanwhile,due to the complexity in bioinformatics data processing and lack of quality management standards,further efforts and explorations need to be put in the application and quality management of NGS in clinical laboratory.

4. Clinical analysis of benign paroxysmal positional vertigo in youth

Objective To explore the clinical features of benign paroxysmal positional vertigo (BPPV) in patients of 40 years old or younger. Methods The clinical features of 40 patients (age ≤40 years) with BPPV admitted from August 2009 to July 2011 were retrospectively analyzed, and compared with that of 286 middle and old-aged patients (age >40 years) with BPPV admitted in the same period. Results The incidence of BPPV in young patients showed a relatively higher incidence (12.3%), with the sex ratio of male to female 1:3 and a mean age of 32.1±6.2 years. The mean latent period of vertigo attack was 1.2±1.3 seconds, and 10 patients (25%) showed no obvious latent period. The median duration of vertigo was 10s. The posterior semicircular canal was involved in 27 patients (67.5%). Cupulolithiasis was found in 6 patients, and all the otolithiasis was found to involve the posterior semicircular canal. The proportion of BPPV with no obvious latency was higher in the young patients than in the middle and old-aged patients (25.0% vs 11.5%, χ2=5.554, P=0.018), but no significant difference in sex ratio, the course of disease, the ratio of involved semicircular canal, the latency and the duration of vertigo attack was found between two groups (P>0.05). Conclusions The incidence of BPPV is relatively higher in youth, more common in females, and the posterior semicircular canal is commonly involved. Compared with the middle and old-aged patients with BPPV, the proportion patients with no obvious latency of vertigo attack in young patients is higher, and the incidence of cupulolithiasis involving posterior semicircular canal is higher in the youth.
5.Cognitive impairment after ischemic stroke: pathophysiological mechanisms and predictors

Cognitive impairment caused by ischemic stroke is becoming more and more concerned.It is crucial to explore its pathophysiological mechanism and related clinical predictive indexes for understanding the occurrence,development and treatment of the disease.This article discusses the pathophysiologic mechanism of cognitive impairment after stroke from the aspects of white matter damage,amyloid abnormal deposition,blood-brain barrier damage,and synaptic plasticity injury,and reviews the risk factors,imaging and biological markers that can be used to predict cognitive impairment after stroke.

Methods: Aiming at the use and management of instruments of teaching and researching in universities, this paper expounds how to reasonably purchase and improve the use efficiency of large-scale instruments. Results:Five suggestions were proposed. Conclusion: To give full play to the investment benefit and its important role of large-scale instruments in the teaching and scientific research.
8.Optimum culture media and liquid conditions in shaking flasks for laccase production by Ganoderma lucidum

Objective To study the optimum culture media and liquid conditions in shaking flasks for laccase production by Ganoderma lucidum. Methods Taking S_3 stain of G. lucidum as test materials and laccase activity of G. lucidum as measurement index to optimize the culture media and liquid conditions through orthogonal test. Results The optimum culture components of media were: Glucose 30 g/L, cotton 0.2%, (NH_4)_2HPO_4 0.66 g/L, casein 0.5%, Tween-80 0.15 mL; the optimum conditions were: initial pH value of medium was 5.5, 75 mL medium was in 250 mL-flask, inoculation was 12.5%, mycelium age was 5?24 h for 9?24 h culture. Conclusion Laccase activity of G. lucidum has been improved remarkably in the optimum culture media and liquid conditions.

As many naturally-derived proteins,keratins have intrinsic biological activity,good biocompatibility and are biodegradable.Human hair keratins,as one type of the autogenous proteins,also have low immune rejection.Compared to other autogenous materials,human hair keratins come from a variety of sources and show attraction in field of regenerative medicine.The human hair keratins applied in neural implant materials,wound dressing,cell culture substrates,drug delivery system and soft tissue reconstructions,which are based on hydrogels,scaffolds coating,films and composite materials present huge development potential in regenerative medicine.
