1.The clinical effects of metformin combined with cyproterone therapy on Dolyeystic ovary syndrome

Objective To observe the curative effects of metformin combined with cyproterone therapy on polycystic ovary syndrome(PCOS). Methods Ninety diagnosed PCOS patients were treated. The serum luteinizing hormone(LH), luteinizing hormone and follicule stimulating hormone(LH/FSH) ratio and T, waist hip ratio(WHR) of the patients before therapy were compared with using merformin combined with- cyproterone three cycles. Results A significant reduction(P<0.05 or P<0.01) in LH,LH/FSH,T and WHR of patients after the therapy was observed. Menses became regular after the therapy, and ache and hirsutism mitigated. Conclusion Metformin com- bined with cyproterone is a perfect means to treat polycystic ovary syndrome(PCOS).

Objective To observe the effects of hypoxia preconditioning on the mitochondria ultrastructure of hepatocyte during liver transplantation in rats.Methods A modified orthopotic liver autotransplantation model was used to simulate liver transplantatation.Seventy two Sprague-Dawley rats were randomly divided into the following three groups:the normal control(group NC),the autotransplantation group(group AT),and the hypoxia preconditioning group(group HP),with twenty four rats in each group.Group HP were given an 8% oxygen atmosphere for 90 minutes before the operation.At 1,6,and 24 hours after the operation,the rats were killed and the following tests were conducted:1.The morphology of mitochondria was observed under transmission electron microscopy(TEM);2.Mitochondria were quantitatively analyzed by a MiVnt image analysis system.ResultsHepatic cells in group AT showed typical injury signs under TEM,the appearance of hepatocytes and mitochondria in group HP were much better than the group AT,and the areas,perimeters and diameters of mitochondria in group HP were much smaller than those in group AT by a significantly amount(P
4.Effect of parecoxib on cardiac function after acute myocardial infarction in rats

Objective To investigate the effect of parecoxib on cardiac function after acute myocardial infarction (AMI) in rats. Methods Twenty-four adult male SD rats, weighing 230-250 g, were randomly divided into 3 groups ( n = 8 each): group Ⅰ sham operation (group S), group Ⅱ AMI and group Ⅲ parecoxib (group P). Myocardial infarction was induced by ligation of left anterior descending branch (LAD) of coronary artery in group Ⅱ and Ⅲ . In group S, LAD and cervical sympathetic trunk were exposed but not ligated and transected.Group P received intrperitoneal parecoxib 8 mg/kg once a day for 3 days 24 h after ligation of LAD, while group AMI received normal saline instead. At 4th day after ligation LAD, the left ventricular systolic pressure ( LVSP),left ventricular end-diastolic pressure (LVEDP) and ± dp/dtmax were measured and recorded. Blood samples were taken from common carotid artery to determine the plasma concentrations of TXA2 and PGI2 and PGI2/TXA2 was calculated. Then the animals were sacrificed and hearts removed. Myocardial infarct size of left venicle was calculated. Results Compared with group S, LVSP, ± dp/dtmax, plasma concentrations of PGI2 and PGI2/TXA2 were significantly decreased, while LVEDP and plasma concentrations of TXA2 increased in group AMI and P( P <0.05). Compared with group AMI, LVSP, ± dp/dtmax, plasma concentrations of PGI2 and PGI2/TXA2 were significantly decreased, while LVEDP and plasma concentrations of TXA2 increased in group P ( P < 0.05). There was no significant difference in myocardial infarct size between group AMI and P (P > 0.05). Conclusion Parecoxib can improve cardiac function after AMI in rats and the mechanism is related to regulation of the balance of PGI2/TXA2.
5.Etiology,Clinical and Electroencephalogram Characteristics and Prognosis of Infantile Spasms with Focal Seizures

Objective To explore the characteristics of etiology,clinical,electroencephalogram(EEG) and prognosis of infantile spasms(IS) with focal seizures(FS).Methods The significance of age onset,seizure patterns and atteration,etiology,video-EEG(VEEG) and evolution of FS correlating to epileptic spasms(ES),which occurred in 12 cases with IS by means of clinical observation,cranial CT or MRI,VEEG monitory and follow up were investigated.Twelve cases were divided into group A,B,C according to the stages of FS occurring prece-ding,coinciding and following ES.Results Ten cases with IS were identified from focal cortical dysplasia,tuberous scleroses complex,temporal lobe cyst or scleroses and hypoxic-ischemic encephalopathy and others.Clinical manifestation presented FS at certain times during the course of the disease,and other characteristics of frequent attacks,asymmetric spasms or tonic spasms,some atypical seizures in the eyes and the head,motionless staring and focal motor seizures and other.The correlation of ES to FS occurred during one ictal episode as follows:FS→ES(6 cases),ES→FS→ES(1 case),ES→FS(3 cases).FS appeared more frequently,atypical,predominantly involving ocular,facial,oral movement or generalized convulsion,migrating or alternating seizures,associated with epileptic discharges of posterior parietal-occipital and parietal-temporal-occipital origins in group A and early period of group B;whereas in the late period of group B and group C,FS occurred less frequently and more stable,presenting complex partial seizures or secondarily generalized seizures originated mainly from frontal and surrounding areas in the lateralized or bilateral hemispheres.Regarding the natural evolution and development of IS,2 cases with early FS developed into IS;6 cases with IS evolved into Lennox-Gastaut syndrome or symtomatic generalized epilepsy,4 cases got into FS,1 case maintained in a peculiarly epileptic state with FS as well as ES until the operation at 4.5 years old,and the other case was not identified clearly.Conclusions IS coincided with FS is a special kind of aged-related FS associated with secondarily generalized seizures and epileptic encephalopathy.The multiple etiology,seizure patterns,ictal-interictal EEG,clinical evolution and prognosis of the disorder,indicate a complicated interaction of the immature cortico-subcortical abnormalities in the critical developmental period and thereafter,can be identified as a peculiar form of clinical epileptic syndrome.

The market and literature were studied to understand the existing situation of Magnoliae Officinalis Cortex goods, and the collected samples were analyzed, combined with the actual production, a new standard of Magnoliae Officinalis Cortex commercial specification and grade was drafted. Magnoliae Officinalis Cortex goods was divided into two categories according to the source in the old standard. Then each category was divided into four kinds of specifications according to the site. Each kind of specification was divided into several grades according to the length and weight. To judge the quality of Magnoliae Officinalis Cortex goods was mainly based on the appearance quality. In the new standard, the classification of commercial specification and grade is based on the thickness, magnolol and honokiol content. The goods of Magnoliae Officinalis Cortex can be divided into three specifications: Tongpu, Genpu and Doupu. Tongpu is divided into three grades, the remaining two are not graded.

7.Protective effect and action mechanism of petroleum ether extracts from Saussurea involucrate on brain tissues of hypoxia rats.

OBJECTIVETo investigate the protective effect and action mechanism of petroleum ether extracts from Saussurea involucrate on brain tissues of hypoxia rats under constant pressure and closed conditions.
METHODThe PESI dosage-dependent experiment for hypoxia rats was conducted under constant pressure and closed conditions by intraperitoneally injecting 125, 250, 500 mg x kg(-1) to finalize that the optimum dosage is the high dose of PESI. Afterwards, 90 Wistar rats were randomly divided into the hypoxic model group, the acetazolamide 250 mg x kg(-1) group and the PESI high dose group. Each group was further divided into three subgroups according to different hypoxia times, with 10 rats in each subgroup. Under the same hypoxia and administration conditions, the rats were sacrificed after 0, 3, 6 h respectively. Their brain samples were collected for common pathological observation and immunohistochemical staining of HIF-1alpha. Real-time RT-PCR was used to detect HIF-1alpha, EPO, HO-1 and Caspase-3 gene expressions. And the Western blot assay was adopted to detect HIF-1alpha protein expression.
RESULTThe brain tissues of the hypoxia model group were severely damaged with the increase in the hypoxia time. The acetazolamide group and the PESI high does group were damaged in a much lower degree. According to the gene expression and the Western blot assay, high dose of PESI could inhibit HIF-1alpha expression. According to the pure gene expression test, high dose of PESI could increase EPO and HO-1 mRNA expressions, but inhibit Caspase-3 mRNA expression.
CONCLUSIONPESI's protective mechanism for brain tissues of hypoxia rats under constant pressure and closed conditions may be related to its effects in inhibiting HIF-1alpha expression, increasing EPO expression and resisting cell apoptosis.
