1.Effect of curcumin on PI3K/AKT/mTOR in rats with cerebral ischemia-reperfusion injury

Objective To explore the effect and mechanism of curcumin on rat cerebral ischemia reperfusion injury.Methods The rat model of cerebral ischemia reperfusion injury was constructed by the suture-occluded method.The effects of curcumin on cerebral infarction range,cerebral water content,neurological symptoms,cerebral histopathological morphology and expressions of PI3K,AKT,p-AKT,m-TOR,MDA,CAT,GPX,SOD,Bcl-2,Bax,Caspase-3 and Cleavage-Caspase-3 were evaluated.Results Cur-cumin had the protective effect on cerebral ischemia reperfusion injury,could alleviate the neurological symptoms,decreased the cer-ebral tissue pathological morphological changes and cerebral water content,in addition,which could alleviate the expressions of MDA,Bax,Cleavage-Caspase-3,IL-6,MCP-1 and TNF-αand increased the expressions of PI3K,p-AKT,mTOR,Bcl-2,Caspase-3, CAT,GPX and SOD.Conclusion The curcumin pretreatment has the significantly protective effect on cerebral ischemia-reperfusion injury,which may be associated with activating PI3K/AKT/mTOR signal pathway,while suppressing inflammation,apoptosis and oxidative stress.
2.Optimum culture media and liquid conditions in shaking flasks for laccase production by Ganoderma lucidum

Objective To study the optimum culture media and liquid conditions in shaking flasks for laccase production by Ganoderma lucidum. Methods Taking S_3 stain of G. lucidum as test materials and laccase activity of G. lucidum as measurement index to optimize the culture media and liquid conditions through orthogonal test. Results The optimum culture components of media were: Glucose 30 g/L, cotton 0.2%, (NH_4)_2HPO_4 0.66 g/L, casein 0.5%, Tween-80 0.15 mL; the optimum conditions were: initial pH value of medium was 5.5, 75 mL medium was in 250 mL-flask, inoculation was 12.5%, mycelium age was 5?24 h for 9?24 h culture. Conclusion Laccase activity of G. lucidum has been improved remarkably in the optimum culture media and liquid conditions.

4.lnfluences of Rhodospirillaceae on immune function of mice

Objective To investigate the influences of Rhodospirillaceae on immune function of mice.Methods Four tests were performed in mice after 4 w medication with different doses of Rhodospirillaceae ig,including:1.The transformation of spleen lymphocyte(MTT method);2.The phagocytosis of macrophagocyte(semi-vivo method);3.The value of HC50; 4.The activity of NK cell(LDH method).Results1.The transformation of spleen lymphocyte;2.The phagocytosis of macrophagocyte,the Value of HC50 and the activity of NK cell in test groups were all raised,the differences were significant and very significant when compared with the control group.Couclusions Rhodospirillaceae could enhance the cellular and humoral immunity of mice.
5.The Clinical Features of the Sick Sinus Syndrome in Elderly Patients

Objective To Study the clinical characteristics of the :sick sinus syndrome,(SSS) in elderly patients.Methods The disease onset age and causes, natural course and symptoms,dynamic electrocardiography(DCG) and cardiac pacing occasion were reviewed and analyzed. in 50 elderly patients had the diagnosis of SSS.Results The morbidity of SSS had increased significantly in elderly men over 70 years. The ordinary causes were coronary heart disease (CHD),specific SSS and the hypertensive heart disease,cardiomyopathy; and degenerative valvular heart disease etc.The mean natural course were about 10 years.The course of DCG type III was the longest one and the type IV was the shortest. The early symptoms were palpitation, dizzy and weakness,and the late symptoms were transient amaurosis, syncope and the worse cardiac angina and heart failure .The cardiac pacing was predicated by the clinical symptom's severity, which was determinated by both of the severity of bradycardia or sinus arrest and the patient's ability of compensation and adaptation.DCG was the commonly used diagnostic method for SSS in elderly patients.Conclusions The SSS has high incidence in elderly men over 70 and is caused by kinds of deasese,such as CHD and specific SSS,etc.The course is frequently intermittent,hide and very long the disease course. The early symptoms and DCG are atypical. The late symptoms commonly are transient amaurosis and syncope .and the late DCG has four types.The cardiac pacing was predicated by the clinical symptom's severity and DCG was the commonly used to diagnose SSS in elderly patients.
6.Experience on the treatment of 62 cases of endoscopic thyroidectomy via breast areola approach

ObjectiveTo study the therapeutic effect and feasibility of endoscopic thyroidectomy via breast areola approach. MethodEndoscopic thyroidectomy via breast areola approach was performed in 62 patients, including 16 cases of adenoma, 43 cases of nodular goiter, and 3 cases of thyroid carcinoma.ResultsEndoscopic thyroidectomy was succeeded in 62 patients, 2 cases of cutaneous emphysema, 1 case of light skin bum which diameter was 0.5 cm, 1 case of transient hoarse voice,and 1 case of puncture sinus tract bleeding. There was no converted to open surgery,no hypercapnia, no damage of recurrent laryngeal nerve or parathyriod glands. ConclusionEndoscopic thyroidectomy via breast areola approach is safe and effective,and has good cosmetic results.

8.Analysis of laparoscopic examination in diagnosis of aseites of unknown origin

Objective To investigate the diagnosis value of laparosco-pie examination in ascites.Methods The data of twenty-three cases with ascites of unknown origin undergoing laparescopie examination and pathologic examination were retrospectively analyzed from May 2004 to August 2008.Results Twenty-two eases were definitely diagnosed by laparoscopie exploration,the positive rate was 95.6%.One ease re-mained unknown.Among them,18 eases were tuberculous peritonitis; 3 eases were metastatic adenocarcino-ma; one ease was peritoneal mesothelioma.No obvious postoperative complications occurred.Conclusion Laparoscopie examination is a safe method with hige definite diagnostic rate,which has important values in diagnosis of ascites of unknown origin.
10.Rapid N-glycosylation profiling of glycoprotein based on ultrafiltration-devices.

With development of bio-technique, more and more proteins were applied as clinical approaches. However, the protein homogeneity, especially the N-glycosylation limited the further research and application of these protein drugs. The analysis method for N-glycans is believed to be critical in protein drugs development. To enhance the N-glycans isolation efficiency and accelerate the pretreatment, a new strategy was built on ultrafiltration-devices. New methods increased the isolation efficiency of N-glycans containing N-acetylglucosa mine with 10%-20%. The degrading of N-glycans containing sialic acids was also minimized with this method. 20%-100% more N-glycans with sialic acids were isolated. The pretreatment was finished within 30 min. Coupled with HPLC-HRMS, an effective and reliable strategy designed for protein drugs N-glycans analysis were developed.
