2.Determination of ganciclover in human plasma by reversed-phase high performance liquid chromatography

Objective To develop a method for the determination of ganciclover in human plasma by RPHPLC.Methods Plasma containing ganciclover was extracted with methanol and methylene chloride,qualitative and quantitative analysis was carried out directly.Working curve,linear range,recovery,precision and so on was obtained according to the sample pre-processing method and analysis state.The HPLC method has been taken to investigate the plasma concentration of ganciclover for 12 volunteers.Results The relationship of the peak area of ganciclover concentration in plasma linear within the range of 0.05 μg/mL~1.60 μg/mL(r=0.9999).The lowest detection limit was 0.01 μg/mL(S/N≥13).The intra and inter-day RSD were less than 5.1%respectively.The recovery is about 90.0%~95.4%.Conclusion The established method in the article was shown to be sensitive,accurate and simple for the determination of ganciclover level.It is suitable for clinical detection of ganciclover and forensic medicine and toxicology analysis.
4.Progress in study of a novel protein Nischarin

Nischarin is a novel protein interacting with integrin ?5 subunit.A series of studies have indicated that Nischarin plays an important role in inhibiting tumor cells migration and invasion.Meanwhile it may also serve as the functional I1-imidazoline receptor.To study this protein will be in favour of understanding the biological mechanisms of inhibiting cell migration and developing novel drugs such as inhibitors for tumor invasion.The article mainly reviews the recent progress in studies on Nischarin.
6.Serum levels of interleukin-36 and its receptor antagonist in patients with psoriasis and their correlations with disease severity

Objective To evaluate the relationship of serum levels of interleukin (IL)-36α,IL-36β and IL-36γas well as their receptor antagonist IL-36Ra with disease severity in patients with psoriasis.Methods Venous blood samples were collected from 45 patients with generalized pustular psoriasis (GPP),34 patients with psoriasis vulgaris (PV),and 37 healthy human controls.Enzyme-linked immunosorbent assay (ELISA) was performed to determine the serum levels of IL-36α,IL-36β,IL-36γ and IL-36Ra.Nonparametric Mann-Whitney test was conducted to compare the levels of IL-36 and IL-36Ra among these groups,and Spearman's rank correlation analysis to assess the relationship between the serum levels of IL-36 and IL-36Ra and disease severity.Results No statistical difference was observed in the serum levels of IL-36 or IL-36Ra among the patients with GPP,patients with PV,and healthy human controls.The serum levels of IL-36β and IL-36γ (given as the median ± interquartile range) were significantly higher in 27 patients with GPP during episodes of pustules ((12.101 ± 11.315) ng/L and (34.541 ± 15.580) ng/L respectively) and in 7 patients with severe GPP ((11.218 ± 9.318) ng/L and (38.536 ± 17.332) ng/L respectively) than in the healthy human controls ((5.355 ± 9.020) ng/L and (23.052 ± 22.410) ng/L respectively,P < 0.05 or 0.01).The serum level of IL-36γwas positively correlated with that of IL-36β in patients with GPP,patients with PV,and the healthy human controls (r =0.85,0.86,0.91,respectively,all P < 0.01),and both IL-36β and IL-36γserum levels were lowly and positively correlated with the severity of GPP (r =0.33,0.41,respectively,both P < 0.05).A positive correlation was also observed between the serum level of IL-36β and psoriasis area and severity index (PASI) scores in patients with PV (r =0.54,P < 0.01).Conclusions The serum levels of IL-363 and IL-36γ are lowly and positively correlated with disease severity in patients with GPP,suggesting that IL-36β and IL-36γplay an important role in the pathogenesis of GPP.
7.Effects of endoscopic ultrasonography combined with fine needle aspiration biopsy on pancreatic cystic lesions

Objective To evaluate the effects of endoscopic ultrasonography ( EUS) combined with cyst fluid analysis, cytology, histopathology based on endoscopic ultrasound guided fine needle aspiration ( EUS?FNA) on pancreatic cystic lesions ( PCLs) . Methods Clinical data of 45 patients were analyzed ret?rospectively from January 2006 to April 2015 including MRI, EUS?FNA, MRCP. The results of conventional imaging and EUS?FNA were compared based on postoperative pathology. Results There were 8 cases of non tumor pancreatic cystic lesions, 4 cases of serous cystadenoma (SCN), 11 cases of mucinouscystadenoma ( MCN) , 8 cases of intraductal papillary mucinous tumor ( IPMN ) , 5 cases of solid pseudopapillary papilloma ( SPN) , 9 cases of non pancreatic cystic lesions ( nPCLs) . The diagnostic accuracy rate of tradi?tional imaging tests ( B?ultrasound, CT, MRI, MRCP ) and EUS?FNA were 42?2% ( 19/45 ) and 77?8%(35/45) (P<0?05). The diagnostic value of EUS?FNA of PCLs was higher than that of traditional imaging tests, with specificity, sensitivity, positive predictive value ( PPV) and negative predictive value ( NPV) of 58?3% ( 7/12 ) , 97?0% ( 32/33 ) , 86?5%( 32/37 ) , 87?5% ( 7/8 ) and 41?7% ( 5/12 ) , 87?9%(29/33), 80?6% (29/36), 55?6% (5/9),respectively. Conclusion EUS?FNA is more valuable than traditional imaging tests for the diagnosis of PCLs.
8.Detection and Drug Resistance Analysis of Ureaplasma urealyticum and Mycoplasma hominis of Urogenital Tract

OBJECTIVE To find out the status about detection and drug resistance of Ureaplasma urealyticum(Uu) and Mycoplasma hominis(Mh) of urogenital tract infection in Shaoxing area of Zhejiang Province.METHODS The detections and drug sensitive tests of Uu and Mh adopting the reagent boxes produced by bioMerieux Company in France were made.RESULTS In 2678 samples of suspicious urogenital tract infection patients,980 samples cultured of mycoplasma were positive,and the positive rate was 36.6%.Among these cases,763 samples(28.5%) were Uu positive;116 samples(4.3%) were Mh positive.In the drug sensitive test,Uu and Mh were most sensitive to PRI,JOS,DOX and TET.To CFLX,OFL,ERY,CM and AM,they had the highest level of drug resistance.CONCLUSIONS The clinical therapy of mycoplasma infection ought to choose the appropriate antibiotics based on the results of the drug sensitive test of the laboratory.
9.Preparation and Quality Evaluation of Xiaochuan cataplasm

Objective To study the influence of the preparation and quality control of Xiaochuan cataplasm on its drug action. Methods Xiaochuan cataplasm was prepared with hydrosoluble polymers matrix. A sequence of tests such as adhesive force test, recontour test, paste-content test, cold-resistance test, heat-resistance test, toxicity and pharmacodynamic action tests were performed on this drug. Results Xiaochuan cataplasm contains a sound volume of medicine, and shows good adhesive property, no sensibilization and stimulation, and notable drug action. Conclusion The results demonstrate that the preparation craft of Xiao-chuan cataplasm is reasonable and practicable., the quality is controllable and the method is a safe and effective transdermal drug delivery system.
10.Effects of thalidomide on the expression and secretion of vascular endothelial growth factor and tumor necrosis factor-alpha by a human keratinocyte cell line HaCaT

Objective To investigate the effect of thalidomide on the proliferation of as well as the expression and secretion of vascular endothelial growth factor (VEGF) and tumor necrosis factor-alpha (TNF-α) by the human keratinocyte cell line HaCaT.Methods Cultured HaCaT cells were divided into several groups to be treated with dimethyl sulfoxide (negative control group) and various concentrations (0.01nmol/L-100 μmol/L) of thalidomide (experimental groups) respectively for 20 to 24 hours.Subsequently,water soluble tetrazolium-1(WST-1) assay was performed to estimate cellular proliferation,real time quantitative PCR to detect the mRNA expression of VEGF and TNF-α in HaCaT cells,and enzyme-linked immunosorbent assay (ELISA) to quantify the protein expressions of VEGF and TNF-α in the culture supernatants of HaCaT cells.Statistical analysis was done by one-way analysis of variance with least significant difference post hoc test.Results The survival rate of HaCaT cells was 74.3%,82.9% and 90.8% after 24-hour treatment with thalidomide of 100,10 and 1 μmol/L respectively,significantly lower than that in the negative control group (100%,all P <0.01).A significant decrease was induced in the mRNA expression (0.439-to 0.634-fold change,all P <0.01) and supematant level ((0.587-to 0.923-fold change,P <0.05) of VEGF in HaCaT cells by thalidomide of 0.01-100 nmol/L,as well as in the mRNA expression (0.493-to 0.587-fold change,P <0.05) and supernatant level (0.408-to 0.617-fold change,P <0.01) of TNF-α by thalidomide of 0.1-100 nmol/L.Conclusion Within a certain range of concentration,thalidomide could suppress the proliferation of,as well as the expression and secretion of VEGF and TNF-α by,HaCaT cells.
