Objective:To provide a basis for the choicing of Semen Cuscuta baked with stir frying with yellow wine.Methods: The orthogonal design was adopted to optimize the processing methods. The content of total flavonoids determined was as a marker to evaluate the different processes. Other three comparative experiments with different auxiliary materials were observed. Results: the optimum processing method with yellow wine was A 2B 3C 1D 2, and the contents of total flavonoids of test sample was highest in the experiments.Conclusions: The optimum processing method with yellow wine is feasible.
2.Progress on preparation methods of animal model of deep venous thrombosis.

Deep venous thrombosis is a common and serious complication after orthopedics operation, with the characteristics of high incidence rate and death rate, its formation mechanism and the treatment is becoming more and more attention of scholars. Establishment of animal model of deep venous thrombosis can further explore the pathological process of thrombosis or dissolution, is an important means to research of thrombosis mechanism and evaluation of therapeutic method. This review discussed the basic principle of deep venous thrombosis, the selection of experimental animals and making method of animal models.

4.Relationship between muscarinic receptor subtypes and detrusor hyperreflexia in spinal cord transected rat

Objective To investigate the relationship between muscarinic receptor subtypes and detrusor hyperreflexia. Methods Spinal cord was transected in male Wistar rats at L 1~2 level, changes of urodynamics were monitored 3 weeks later by cystometric studies.Spontaneous contraction frequency and strength of detrusor strip mustarded by carbachol (muscarinic receptor agonist), atropine (nonselective M antagonist), methoctramin (M 2 receptor selected antagonist) and 4 DAMP (M 3 receptor selected antagonist) were measured. Results Bladders after SCI showed littler capacity, lower compliance and hyperreflexia. Detrusor of all groups contracted at a definite tension. The contraction frequency in the hyperreflexia group (4.797?0.257 times/min) was higher than in the control's(3.227?0.173 times/min).There were no obvious changes while detrusor strips were mustarded by different muscarinic receptor antagonists.Carbachol could increase detrusor contraction frequency( 6.316 ?0.409 times/min) and the effect could be blocked by atropine(5.304?0.095 times/min) and 4 DAMP(5.046?0.089 times/min). Conclusions Detrusor is a rythmatic tissue at a difinite tension.Excitability rise of detrusor is the main cause of hyperrefexia. M 3 receptor can enhance the excitability of detrusor causing direct contraction.The function of M 2 receptor is still unclear.
5.Clinical significance of detrusor leak point pressure study in evaluating damage of the upper urinary tract secondary to neurogenic voiding dysfunction

Objective To evaluate clinical significance of detrusor leak point pressure (DLPP) in the damage of the upper urinary tract secondary to neurogenic voiding dysfunction. Methods 38 cases of voiding dysfunction received the urodynamic test,especially detecting of detrusor leak point pressure. Results 38 cases were divided into 2 groups by DLPP of 4.0 kPa,26 in high DLPP group and 12 in low DLPP group.The bladder capacity of the high pressure group is (422.95?183.27)ml,significantly less than the low pressure group (464.83?106.43)ml,and its compliance is lower as well as with DSD.Clinical results of B ultrasound,IVU,BUN,Cr show that the high pressure group has more serious than that in the low pressure group. Conclusions DLPP is valuable in evaluating the damage of upper urinary tract secondary to neurogenic voiding dysfunction.
6.The influence of pre-injury stressful life events on the life quality and mood symptoms following cerebral concussion

Objective To test the reliability and validity of Chinese version of Stressful Life Events Questionnaire and the influence of preinjury stressful life events on the life quality and mood symptoms of the patents following cerebral concussion.Methods 285 patients with cerebral concussion were assessed by Stressful Life Events Questionnaire(SLESQ),PTSD Checklist civilian version(PCL-C),Thirty-Six-Item Short-Form Health Survey (SF-36),Hamilton Depression Rating Scale(HDRS),Hamilton Anxiety Scale(HAMA) at baseline.The patients had been reassessed with the HDRS,HAMA,SF-36 at 3 months after cerebral concussion.The hierarchical regression analyses were used to determine the influence of Stressful Life Events for the life quality and mood symptoms of the patient with cerebral concussion.Results The incidence of stressful life events was 42.8%.The multiple regression analysis showed the experience of stressful life events were the influence factor of scores on the SF-36,HDRS,HAMA after adjusted by the age,gender,education,income,social support,the scores of anxiety and depression before injury and the R 2 value of SLESQ to SF-36 physical health factor,mental health factor,HDRS,HAMA was 0.064,0.111,0.145,0.028,respectively.Conclusion The history of stressful events is the influence factor of the patient with cerebral concussion to have poor life quality and mood disorder.
7.Quality control of Longshen Buyi Granule

AIM: To establish a quality control for Longshen Buyi Granule(Radix et Rhizoma Ginseng,Radix Panacis Quinquefolii,Radix Astragali,etc.. METHODS: Ingredients of Radix Ginseng,Radix Panacis quinquefolii,Fructus Corni,Fructus Ligustri lucidi,Radix Ophiopogonis,Pericarpium Cirri reticulatae,Radix Glycyrrhizae were identified by TLC,and astragaloside IV in the granule was determined by HPLC-ELSD. RESULTS: The spots in the TLC were clear and distinguished well.The linear range was within(0.333 8)-10.680 0 ?g(R=0.999 1).The average recovery was 99.42%,and the RSD was 2.00%. CONCLUSION: This method can be used for quality control of Longshen Buyi Granule.
8.Comparison of anatomical locking plate and Gamma nail for the treatment of intertrochanteric fracture with external wall fractures.

OBJECTIVETo compare clinical efficacy of anatomic locking plate and Gamma nail in treating unstable femoral intertrochanteric fractures with external wall fractures.
METHODSFrom June 2010 to June 2014,clinical data of 44 patients with intertroehanteric fractures associated with lateral wall fractures (type 31A2.2-3.3) followed more than 12 months,which treated with Gamma nail or anatomic locking plate,were retrospective analyzed. Sixteen patients were treated with anatomic locking plate, including 6 males and 10 females aged from 32 to 83 years old with an average of 56.5 years old. Twenty-eight patients were treated with Gamma nail including 17 males and 11 females aged from 26 to 87 years old with an average of 60.4 years old. Operative time, intraoperative fluoroscopy times, blood loss (intraoperative and hidden blood loss), hospital stays were observed and compared. PPMS and HHS scoring were used to evaluate postoperative clinical effect.
RESULTSAll patients were followed up from 12 to 24 months with an average of 16.2 months. Operative time in Gamma nail was shorter than anatomic locking plate; while blood loss( intraoperative and hidden blood loss) and intraoperative fluoroscopy times in anatomic locking plate were less than that of in Gamma nail. There was no significant meaning in hospital stays between two groups. Postoperative full weight-bearing time in anatomic locking plate was prolonged than Gamma nail. At the final following-up, PPMS in Gamma nail was 7.50 ± 1.78 and 6.82 ± 1.38 in anatomic locking plate, and there was no obvious meaning between two groups (t = 2.341, P = 0.132); there was no significant differences in HHS score between Gamma nail (83.25 ± 11.18) and anatomic locking plate (86.14 ± 12.36) (t = 1.923, P = 0.243). The incidence of complications in Gamma nail was less than anatomic lock-ing plate (P = 0.005).
CONCLUSIONAnatomic locking plate for intertrochanteric fractures with external wall fractures could avoid re-injury of external wall, especially for severe comminuted fractures, difficult for intramedullary nailing, and there was no significant meaning in hip joint function compared with Gamma nail, while postoperative incidence of complications was higher than Gamma nail, so early weight-bearing was not stress.

9.Syngeneic and allogeneic fetal liver stem cell transplantation in the treatment of mouse hepatic cirrhosis

BACKGROUND:Immunity of fetal liver stem cel transplantation is rarely reported, syngeneic and al ogeneic fetal liver stem cel transplantation in the treatment of hepatic cirrhosis is stil unclear.
OBJECTIVE:To observe the therapeutic effects of syngeneic and al ogeneic fetal liver stem cel transplantation on hepatic cirrhosis as wel as immune rejections during the therapeutic process.
METHODS:The fetal liver stem/progenitor cel s from BALB/c and C57BL/6 mice were isolated and purified by the type IV col agen enzyme digestion method. A total of 104 healthy BALB/c mice were randomly assigned to four groups. Normal control group:no treatment;Hepatic cirrhosis group, syngeneic transplantation group and al ogeneic transplantation group:16 weeks after hepatic cirrhosis models of mice were developed by intraperitoneal injection with carbon tetrachloride, physiological saline, syngeneic fetal liver stem cel s and al ogeneic fetal liver stem cel s were injected via the caudal vein. Final y, the survival statuses, liver function, hepatic fibrosis index, the number and ratio of immune cel s (CD4+T, CD8+T, NK, NKT) and histopathologic examinations were compared in each group after transplantation 4 weeks.
RESULTS AND CONCLUSION:The survival rates in the two transplantation groups were both 100%, which was significantly higher than that in the hepatic cirrhosis group (67%, P<0.05). The liver function and liver fibrosis index in each group did not show statistical differences (P>0.05). Immunological tests showed no difference between groups (P>0.05). Pathohistology examination of hepatic tissue repair:Al ogeneic transplantation group>syngeneic transplantation group>hepatic cirrhosis group. Hence, fetal liver stem cel transplantation via the caudal vein could elevate the survival rate of hepatic cirrhosis mice, al eviate the degree of hepatocyte necrosis. There is no immunologic rejection during syngeneic and al ogeneic fetal liver stem cel transplantation that could help to treat hepatic cirrhosis in mice.
10.Effect of calcium-sensing receptor in cardiac hypertrophy induced by angiotensin Ⅱ in cultured neonatal rat cardiomyocytes

Objective To explore the roles and possible mechanism of calcium-sensing receptor(CaSR) in cell cardiac hypertrophy model using angiotensin Ⅱ (Ang Ⅱ ).Methods The cultured neonatal rat ventricular myocytes were treated with Ang Ⅱ as cell cardiac hypertrophy model.Hypertrophic neonatal rat cardiomyocytes were treated with GdCl3(a specific agonist of CaSR) and/or with Ro318220(a specific inhibitor of PKC pathway).To evaluate the status of cardiac hypertrophy,cell diameter was observed by HE dyeing,and protein content was determined through coomassie brilliant blue protein kit.The intracellular calcium concentration( [ Ca2+]i) was determined by laser scanning confocal microscope.The protein expression of CaSR and PKC pathway were analyzed using Western blotting.Results ①Compared to the control group(0.1263 ± 0.0443),the protein expression of CaSR was increased in Ang Ⅱ group and in GdCl3 group(0.1963 ± 0.0375,0.2778 ± 0.0564,all P< 0.05).Moreover,compared with Ang Ⅱ alone,the increase was significant in GdCl3 group(P < 0.05).②Compared to control group(222.70 ± 22.09),AngⅡ group(392.16 ± 36.85) remarkably increased [Ca2+]i(P< 0.05),and this increase of [Ca2+]i was further enhanced in GdCl3 group (502.60 ± 44.21) versus Ang Ⅱ group (P < 0.05).③Compared to control group,Ang Ⅱ could induce cardiomyocyte hypertrophy,and GdCl3 enhanced the effect.Moreover,this enhancement was attenuated by Ro318220.④Compared to control group(0.27 ± 0.07,0.69 ± 0.06,0.87 ± 0.04),the protein expression of PKCα,PKCε and PKCδ was increased in Ang Ⅱ group(0.60 ± 0.16,1.02 ± 0.13,1.20 ± 0.18,all P< 0.05) and the protein expression of PKCα,PKCε was increased in GdCl3 group(0.82 ± 0.16,1.34 ± 0.12,all P < 0.05).Moreover,compared with Ang Ⅱ group,the protein expression of PKCα,PKCε was obviously increased in GdCl3 group (all P < 0.05);compared with GdCl3 group,the protein expression of PKCα,PKCε(0.41 ± 0.10,0.85 ± 0.14) was obviously decreased in Ro318220 group(all P < 0.05).Conclusions CaSR is involved in cardiac hypertrophy induced by Ang Ⅱ through PKC pathway in cultured neonatal rat cardiomyocytes.
