1. Evaluation on efficacy of Huaiqihuang granules in treatment of children with asthma at non-acute attack stage

Objective: To explore the efficacy of Huaiqihuang Granules in the treatment of the children with asthma at non-acute attack stage and its effect on the immune function indexes of body. Methods: A total of 180 children with asthma at non-acute attack stage were divided into Huaiqihuang Granules treatment group (n=92, treated with traditional Chinese medicine Huaiqihuang Granules along with oral montelukast and inhaled corticosteroid, ICS+MK+H group) and routine treatment group (n=88, only given oral montelukast and inhaled corticosteroid, ICS+MK group). The indexes of asthma attack degrees, traditional Chinese medicine (TCM) syndrome indexes, blood immune function indexes and pulmonary function indexes of the children in two groups were detected. Results: There was no difference in the disease degrees of the children between two groups before treatment (P>0. 05). Compared with routine treatment group, the score of acute attack of asthma of the children in Huaihuang Granules treatment group was decreased after treatment (P<0. 05). Compared with routine treatment group, the scores of dry / loose stools, low-grade fever, cough/expectoration, spiritlessness/anorexia and white tongue fur of the children in Huaiqihuang Granules treatment group were significantly decreased (P< 0. 01), and the scores of perspire during sleep and dry mouth with desire for drinking were decreased (P<0. 05). Compared with routine treatment group, the levels of immunoglobulins (serum IgA, lgG and lgM) of the children in Huaiqihuang Granules treatment group were increased after treatment (P<0. 05) Compared with routine treatment group, the percentages of T cells, Th cells and NK cells of the children in Huaiqihuang Granules treatment group were increased (P<0. 05), the percentage of B cells was decreased (P<0. 05), and there was no difference in the percentages of serum Ts cells of the children between two groups (P>0. 05). Compared with routine treatment group, the peak expiratory flow (PEF) and the ratio value of forced expiratory volume in the first second /forced vital capacity (FEV1/FVC) of the children in Huaiqihuang Granules treatment group were increased (P<0. 05), but there was no difference in FEV1 of the children between two groups (P>0. 05). Conclusion: On the basis of routine treatment, Huaiqihuang Granules can reduce the condition of children with asthma at non-acute attack stage and improve the TCM syndrome and immune function.
2.Clinical observation on Qidi Yiqi Yangyin Huoxue Recipe in treating diabetic nephropathy at stage III and IV.

OBJECTIVETo observe the clinical effect of Qidi Yiqi Yangyin Huoxue Recipe (QYYHR) on diabetic nephropathy (DN) at stage III and IV.
METHODSOne hundred type 2 DN patients of qi-yin deficiency with blood-stasis syndrome were randomly divided into two groups, all were treated with basal hypoglycemic treatment but with QYYHR given to the treatment group additionally for 3 months. The symptoms were observed, blood glucose, urinary albumin excretion rate (UAER), blood urea nitrogen (BUN), creatinine (Cr) and hemorheological parameters were detected before and after treatment.
RESULTSThe total effective rate was higher in the treatment group than that in the control group (89.6% vs. 68.1%, P < 0.01), and the improvement of UAER, BUN, Cr and hemorheological parameters was also significantly better in the former than in the latter (P < 0.05).
CONCLUSIONQYYHR has favorable effects on type 2 DN of qi-yin deficiency with blood-stasis syndrome.

3. Establishment and identification of humanized mouse models with human immune system and allogeneic human melanoma growth

Objective: To construct a humanized mouse models with human immune system and human tumor growth, and to provide the basis for human tumor immunological research and evaluation on the efficacy of tumor immunotherapeutic durgs. Methods: The humanized mice with human immune system reconstruction were established by intravenous injection of human CD34 fetal liver cells combined with implantation of human fetal thymic tissue under the renal capsule of NOD/SCID IL 2rg -/- after sub-lethal total body irradiation. The peripheral blood was collected, and the immune cell reconstitution was monitored every 3 weeks from the 3th week after humanization, including the ratios of CD45+, CD33, CD3, CD3 CD4+, CD3+ CD8, and CD56 + cells. The human A375 melanoma cells were subcutaneously inoculated at the 15th week after humanization, and the tumor volume was measured every 5 d.. Six weeks after tumor inoculation, the mice were sacrificed and the composition of human immune cells as well as their phenotypes in peripheral blood, bone marrow, lymph node, spleen and tumor tissue were measured by flow cytometry. The composition of human immune cells as well as their phenotypes in tumor tissue were measured by immunohistochemistry. Results: At the 15th week after humanization, the ratio of human T cells was (33. 53 ± 8. 57) % and the ratio of human B cells was (43. 33 ± 10. 05) % in the peripheral blood of the humanized mice. The human melanoma grew slowly during the first 3 weeks, and the human tumor sizes expanded rapidly and reached (1 564 ± 334. 3) mm3 at the 6th week after tumor inoculation. The immunohistochemical and flow cytometry analysis results showed that a large number of human T cells, which mainly composed by CD8+ T subsets, infiltrated in the human melanoma. The ratio of CD8/CD4 in tumor tissue was significantly higher than those in the peripheral blood, bone marrows, lymph nodes and spleen tissue (P< 0. 01). The levels of PD-1 molecules expressed in tumor-infiltrated human CD4 T and CD8 T cells were higher than those in the peripheral blood and immune organs. Conclusion: A humanized mouse models with human immune system and allogeneic human melanoma is successfully established. The high levels of human immune cells, including human T cells and B cells, can be detected in the peripheral blood, lymph organs, and human tumor tissue in this mouse model.
4.Value of serum SDF-1a half-life in predicting postoperative recurrence and metastasis in patients of breast cancer

Objective To investigate the plasma level of SDF-1a in patients with breast cancer after modified radical mastectomy and chemotherapy, and to evaluate the value of serum SDF-1a half-life for predicting breast cancer recurrence and metastasis. The correlation of SDF-1 a half-life and breast cancer recurrence and metastasis after treatment were retrospectively analyzed. Methods Serum chemokine SDF-1a of 112 cases of breast cancer were detected before and after modified radical surgery, and 1 day beforeeach chemotherapy session. SDF-1a levels and the dynamic changes in the process were observed and calculated. Results In 85 cases with no recurrence and metastasis the plasma level of SDF-1 a decreased rapidly to normal level ,while that in 27 cases with recurrence and metastasis was on high level with the halflife of SDF-1a being longer than that in no recurrence group(P <0. 01 ). Taking SDF-1a half-life ≥14 d as cut off point to predict breast cancer recurrence and metastasis after treatment, the sensitivity is 81.5%,specificity is 70. 6 %, and accuracy is 73.2%. Conclusions Serum SDF-1a half-life is a valuable marker in predicting postoperative breast cancer recurrence and metastasis.
5.Celecoxib Inhibits Proliferation and Induces Apoptosis via Cyclooxygen-ase-2 Pathway in Human Pancreatic Carcinoma Cells

In order to evaluate the effects and mechanisms of celecoxib in inhibiting proliferation and inducing apoptosis on human pancreatic carcinoma cells, the anti-proliferative effect was measured by using methabenzthiazuron (MTT) assay. Cell cycle and apoptosis were analyzed by using flow cytometry (FCM), and the PGE2 levels in the supernatant of cultured pancreatic carcinoma cells were quantitated by enzyme-linked immunoabsordent assay (ELISA). Our results showed that celecoxib suppressed the production of PGE2 and inhibited the growth of JF-305 cells, and the anti-proliferative effect of celecoxib could be abolished by addition of PGE2. FCM revealed that celecoxib could inhibit proliferation and induce apoptosis by G1-S cell cycle arrest. It was concluded that cyclooxygenase-2 specific inhibitor celecoxib could inhibit proliferation and induced apoptosis of human pancreatic carcinoma cells via suppression of PGE2 production in vitro.

8.Development and validation of a prediction model for treatment failure in peritoneal dialysis-associated peritonitis patients: a multicenter study.

OBJECTIVE:
To develop and validate a risk prediction model of treatment failure in patients with peritoneal dialysis-associated peritonitis (PDAP).
METHODS:
We retrospectively analyzed the data of patients undergoing peritoneal dialysis (PD) in 3 dialysis centers in Jilin Province who developed PDAP between January 1, 2013 and December 31, 2019. The data collected from the Second Hospital of Jilin University and Second Division of First Hospital of Jilin University) were used as the training dataset and those from Jilin Central Hospital as the validation dataset. We developed a nomogram for predicting treatment failure using a logistic regression model with backward elimination. The performance of the nomogram was assessed by analyzing the C-statistic and the calibration plots. We also plotted decision curves to evaluate the clinical efficacy of the nomogram.
RESULTS:
A total of 977 episodes of PDAP were included in the analysis (625 episodes in the training dataset and 352 episodes in the validation dataset). During follow-up, 78 treatment failures occurred in the training dataset and 35 in the validation dataset. A multivariable logistic regression prediction model was established, and the predictors in the final nomogram model included serum albumin, peritoneal dialysate white cell count on day 5, PD duration, and type of causative organisms. The nomogram showed a good performance in predicting treatment failure, with a C-statistic of 0.827 (95% CI: 0.784-0.871) in the training dataset and of 0.825 (95% CI: 0.743-0.908) in the validation dataset. The nomogram also performed well in calibration in both the training and validation datasets.
CONCLUSION
The established nomogram has a good accuracy in estimating the risk of treatment failure in PDAP patients.

9.Design of new gradient scaffolds based on triply periodic minimal surfaces and study on its mechanical, permeability and tissue differentiation characteristics.

In order to establish a bone scaffold with good biological properties, two kinds of new gradient triply periodic minimal surfaces (TPMS) scaffolds, i.e., two-way linear gradient G scaffolds (L-G) and D, G fusion scaffold (N-G) were designed based on the gyroid (G) and diamond (D)-type TPMS in this study. The structural mechanical parameters of the two kinds of scaffolds were obtained through the compressive simulation. The flow property parameters were also obtained through the computational fluid dynamics (CFD) simulation in this study, and the permeability of the two kinds of scaffolds were calculated by Darcy's law. The tissue differentiation areas of the two kinds of scaffolds were calculated based on the tissue differentiation theory. The results show that L-G scaffold has a better mechanical property than the N-G scaffold. However, N-G scaffold is better than the L-G scaffold in biological properties such as permeability and cartilage differentiation areas. The modeling processes of L-G and N-G scaffolds provide a new insight for the design of bone scaffold. The simulation in this study can also give reference for the prediction of osseointegration after the implantation of scaffold in the human body.
