1. Comparative Research on the Properties of Nitrendipine Solid Dispersions Prepared by Different Techniques

OBJECTIVE: To compare the properties of nitrendipine/PVP k25 solid dispersions prepared by three different technologies including spray drying, freeze-drying and co-precipitation technology. METHODS: The characteristics of nitrendipine solid dispersions including micromorphology, crystalline profile and intermolecular interactions were analyzed by scanning electron microscopy (SEM), differential scanning calorimetry (DSC), powder X-ray diffraction (PXRD) and Fourier transform infrared spectroscopy (FT-IR). The dissolution and solubility characteristics were investigated by dissolution apparatus and constant temperature shaker. The crystallization inhibition effect of polymers against the drug in a supersaturated state was investigated by supersaturation crystallization experiment. RESULTS: Nitrendipine existed in an amorphous form in the solid dispersion obtained by spray drying method. Compared with co-precipitation and freeze-drying method, the solid dispersion prepared by spray drying method significantly improved the dissolution and solubility of nitrendipine (P<0.05). The RESULTS: of supersaturation crystallization inhibition test showed that PVP k25 could obviously inhibit the crystallization of nitrendipine in supersaturated state. CONCLUSION: Based on the analysis of the above mentioned properties, nitrendipine solid dispersion prepared by spray drying technology displays the most optimal properties. Spray drying technology is more suitable for the practical production and industrial applications of nitrendipine solid dispersions.
2.Dihydroartemisinin inhibits proliferation of pancreatic cancer JF-305 cells by regulating expression of apoptosis related proteins and production of reactive oxygen species.

To investigate the effect of dihydroartemisinin on apoptosis of human pancreatic cancer cell line JF-305 and the role of reactive oxygen species(ROS) in the apoptosis of JF-305 cells induced by dihydroartemisinin. MTT assays were used to detect effect of different concentrations of dihydroartemisinin on cells proliferation of JF-305 lines. Cell cycle was detected by flow cytometry, and the apoptotic morphology was observed by Hoechst 333258 fluorescence staining. Annexin V fluorescence staining was used to detect the apoptosis changes of JF-305 cells, while DCFH-DA was used to detect the changes of ROS during apoptosis process. Western blot was used to detect the protein expression changes of Bax, Bcl-2, Cleaved caspase-3, Cleaved caspase-9 and Cyto C. As compared with the control group, the JF-305 cells proliferation was inhibited significantly(P<0.05) after treatment with different concentrations of dihydroartemisimin for 48 h; cell cycle was blocked in the G2/M phase; apoptotic morphology of nuclear condensation, aggregation, and fragmentation was found, and the apoptosis ratio was increased(P<0.05). DCFH-DA detection showed that the cell ROS was increased significantly after dihydroartemisinin treatment(P<0.05). Western blot results showed that the expression of Bcl-2 protein was down-regulated; the expression of Bax protein was up-regulated; the ration of Bax/Bcl-2 was increased and the protein expression levels of Cleaved caspase-3, Cleaved caspase-9 and Cyto C were increased after dihydroartemisinin treatment. Therefore, dihydroartemisinin could induce apoptosis of JF-305 cells, and the possible mechanism may be related to the formation and increasing of ROS.
3.Effect of Cancer Target Drug LHRH-PE40 on Elasticity of HeLa Cells

The quantitative analysis of biomechanical profiles at the single-cell level can provide additional information. It is usually not available in traditional cell biology approaches, but may be crucial to assess and understand tumor prognosis and response to chemotherapy. In this study, the on-line changes of cell elastic properties after the addition of cancer target drug LHRH-PE40 were monitored by atomic force microscopy ( AFM) on living HeLa cell surface under physiological condition. The results from AFM based force spectroscopy showed that LHRH-PE40 induced a distinct increase of the cell surface elasticity of HeLa cells. The fluorescence images implied that the target drug LHRH-PE40 would affect the re-organization of cell actions, which led to the increase of the elasticity of HeLa cells.
4.Therapeutic Effect and Mechanism of Flavonoids from Sophora Flavescens Ait. on Heat Stress-induced Reproductive Dysfunction in Mice

Abstract
OBJECTIVE To investigate the effect and mechanism of flavonoids from Sophora flavescens Ait. on testicular tissue damage in male mice induced by local heat stress in the scrotum. METHODS TCMSP database was used to screen the targets of flavonoids in Sophora flavescens Ait., and the bioinformatics analysis was performed on the target. The mouse model of scrotal heat stress was used and the flavonoids of Sophora flavescens Ait. was used for intervention. The sperm density and sperm aberration rate of mice in each group were measured, and the morphological changes of testicular tissue were observed. Tumor necrosis factor-α (TNF-α) and interleukin 17(IL-17) mRNA and protein levels in testicalar were detected of by q-PCR and Western blotting. Na+-K+-ATPase, Mg2+-ATPase, Ca2+-ATPase, lactate dehydrogenase(LDH), sorbitol dehydrogenase(SDH) activity, malondialdehyde (MDA), NO, TNF-α level and the content of testosterone in serum were detected in tissue homogenate. RESULTS Heat stress could lead to the decrease of sperm density and increase of aberrant sperm, the obvious thinning of testicular spermatogenic epithelium, the decrease of cell level and quantity, the significant decrease of ATPase, LDH, SDH activities, and the increase of MDA, NO content, TNF-α and IL-17 expression in testicular tissue. After the intervention with 250, 500 mg·kg-1·d-1 flavonoids of Sophora flavescens Ait., the quality of sperm and the damage of testicular tissue morphology were improved. The level of TNF-α and IL-17 in serum and testicular tissue were decreased, and the activity of ATPase, SDH and the level of testosterone were increased. CONCLUSION The mechanism of flavonoids of Sophora flavescens Ait. is through inhibiting the inflammatory factor TNF-α and IL-17 levels, improve the anti-lipid peroxidation ability and inhibite the role of NO, enhance the activity of energy enzymes in spermatogenesis, improve the level of serum testosterone, and improve the reproductive disorders caused by heat stress.
5.Determination of Triptolide and Triptonide in plasma by HPLC

Objective To establish a method for quantitative determination of triptolide and triptonide in human plasma samples by HPLC.Methods The samples were extracted with Oasis HLB solid phase extraction cartridge and analyzed with reserved phase HPLC coupled with diode array detection.Results Recoveries rates obtained from spiked plasma for the two diterpenoid were higher than 80%.The linearity ranged from 10 ng/ml to 1 000 ng/ml.Detection limits of the method were 3.0ng/ml for triptolide and 4.5ng/ml for triptonide.Conclusion The method is fast and accurate,which is applicable to determination of diterpenoid in cases of suspected poisoning of Triptergium wilfordii Hook.f. in forensic practice.
6.Retrospective analysis on 1563 cases of cervical cancer patients

Objective To analyze the trend of cervical cancer, age structure and influencing factors. Methods Retrospective study and survey. The 1563 patients with cervical cancer and gynecological medical examination of 600 non-cervical cancer patients who took part in Physical examination in the obstetrics and gynecology department received the enclosed analysis questionnaire. Results Cervical cancer is becoming a trend of cervical cancer patients being younger, high risk period at the age of 36 to 55, and the downward trend towards the menopausal women. The number of being pregnant, ma]-sexuality (the first sexual intercourse at a younger age, or many sexual partners), and smoking are the causes which lead to cervical cancer. Conclusion Give great importance to a trend of cervical cancer patients being younger; screening, publicity and education about cervical cancer;, missionaries and strengthen anti-cancer awareness, promote a healthy lifestyle, which is the key to prevention and treatment of cervical cancer.
7.Clinical significance of microRNA-155 peripheral blood and CD4+T cytological test for patients with atopic dermatitis

Objective:To study the expression level of peripheral blood mircoRNA-155 and the variation characteristics of CD4+T cell percentage and to expound the clinical significance of mircoRNA-155 quantitative test and CD 4+T cell percentage cytological test.Methods:Fluorescent quantitative PCR had been used to detect the expression level of mircoRNA -155 in the peripheral blood.Flow cytometry had been used to detect the cell percentage of the peripheral blood T lymphocyte subset CD 3+T cell,CD4+T cell and CD8+T cell.The mircoRNA-155 expression levels and the cell percentages of CD 4+T were tested respectively in the selected 40 patients with atopic dermatitis ,30 patients with skin eczema and 30 healthy controls.Finally,the differences of the above groups were counted and analyzed .Results:The relative expression levels of mircoRNA-155 of the atopic dermatitis group were higher than those of the eczema group and the normal control group ( P<0.05 ).The CD4+T cell percentage of the atopic dermatitis group was higher than those of the eczema group and the normal control group ( P<0.05 ) .Conclusion: The expression level of the peripheral blood mircoRNA-155 in the patients with atopic dermatitis is increased significantly and CD 4+T cell percentage in the patients is raised as well.Consequently ,the clinical detection of miRNA-155 and CD4+T cell shall be of great significance .
8.Screening and sequencing of the novel differential gene in leptospira serovar lai

AIM: To amplify and analyze the differential DNA fragment between pathogenic leptospira serovar lai and nonpathagenic leptospira serovar patoc I. METHODS: The previously subtractive DNA fragment only existing in leptospira serovar Lai was amplified by cassette ligation and semi-nested PCR.The obtained gene was sequenced and searched homologically. In addition, the deduced amino acid was analyzed and the secondary stracture of protein was predicted. RESULTS: The 580 bp DNA fragment, which deposited in GenBank (AF495587), was cloned, and four overlapping open reading frames (ORF) was contained. The high homology with conserved hypothetical protein streptococcus pyogenes was found. CONCLUSION: This study lays foundation for deeply exploring biological actions of new gene and pathogenic mechanism of leptospira serovar lai.
9. Effects of different concentrations of juglone on the proliferation and apoptosis of HeLa cells

Objective To study the effect of different concentrations of juglone on the proliferation and apoptosis of human cervical cancer cells (HeLa), and explore the anti-tumor effect of juglone in vitro. Methods HeLa cells were cultured in vitro for 24 h and then divided into control group and juglone-treated group. The cells in juglone-treated group were cultured again with 10, 20, 50, 100 and 200μmol/L juglone, respectively, for 24h. The morphological changes in HeLa cells were observed with inverted microscope. The proliferation of HeLa cells was assessed detected by methyl thiazolyl tetrazolium (MTT) assay. The cell cycle and apoptosis were observed by flow cytometry (FCM). The expression of caspase-3 in HeLa cells was determined by caspase-3 colorimetric assay kit. Results Under morphological observation, it was found that different concentrations of juglone led to change in cell shape. MTT assay showed that juglone significantly inhibited the growth of HeLa cells in a dose-dependent manner (P<0.05 or P<0.01). FCM assay indicated that the percentage of the cell cycles arresting at G2/M phase after treatment with above concentrations of juglone for 24 h were increased to 12.92%, 16.23%, 23.64%, 34.22% and 52.64% from 7.5%, respectively (P<0.05 or P<0.01). The caspase-3 activity in juglone-treated HeLa cells remarkably increased in a dose-dependent manner as compared with control group. Conclusion Various concentrations of juglone can inhibit the proliferation and induce the apoptosis of HeLa cells in a dose-dependent manner, implying that juglone may have anti-tumor effect.
10. Eye injury after non-ocular surgery in patients underwent general anesthesia: A report of 2 cases and literature review

Objective: To study the pathogenesis and the risk factors of eye injury after non-ocular surgery in the patients underwent general anesthesia∗ and to provide the reference for its prevention, diagnosis and treatment. Methods: The clinical materials of two patients with eye injury after non-ocular surgery underwent general anesthesia were analyzed and the related literatures were reviewed. Results: A young woman patient underwent laparoscopic hysterectomy with general anesthesia while positioned Trendelenburg, the eyes were being closed naturally without protection, and corneal abrasion of both eyes occurred after operation. The lesion had completely resolved with no sequelae after treatment. An old man underwent cervical posterior laminoplasty with general anesthesia in prone position developed ischemic optic neuropathy (ION) after operation. The vision of the patients partly recovered after symptomatic treatment. Conclusion: Corneal abrasion is the most frequent ophthalmologic complication during general anesthesia, most of the patients have good prognosis. Postoperative visual loss (POVL) is the most severe ophthalmologic complication without effective treatment available, the delicate reasons and mechanisms are not totally clear, prevention outweighs treatment.
