1.An experimental study on the relationship between the expression of GFAP and iNOS and the post-traumatic intervals after brain contusion in rats

Objective To observe the relationship between the expression of GFAP and iNOS the post-traumatic intervals after experimental cerebral contusion,in order to attempt to find out a method of timing wound age of brain contusion in the forensic prediction of injury time.Methods After setting up an model of experimental focal cerebral contusion in rats,immunohistochemical method was applied to observing the changes of inducible nitric oxide synthase (iNOS) and glial fibrillary acidic protein (GFAP) staining combined with histology method in rats.The immunostaining results were measured quantitatively with computer imaging analysis system.Results (1) At 3h after injury,GFAP-positive astrocytes around the wounded area were definitely detected.The intensity and the area of GFAP-positive cells increased following the post-traumatic intervals.The intensity peaked dually at 1d and 5d,respectively,followed by reduction at 3d,which revealed two-peak waveforms,The area of GFAP positive cells enhanced significantly at 3h and gradually increased with prolonging survival time,and reached a peak at 7d after injury.(2) At 12h after injury,the iNOS isoform was present and iNOS positive staining cells were noted.The intensity and the area of iNOS positive cells increased following the post-traumatic intervals.The area of positive reached maximal level at 1-3days,gradually subsided after 5d,and maintain a higher lever up to 7d post-injury;but the intensity of individual positive cell gradually increased with prolonging survival time,reached a peak at 5d and gradually subsided after 5d,but the level was still higher than the initial interval.Conclusions Expression of GFAP and iNOS is correlated with post-traumatic intervals after cerebral contusion in rats,suggesting that the expression of iNOS and GFAP may be served as the markers for timing of brain contusion in forensic practice.
2.Surgical Treatment of Primary Gallbladder Carcinoma

Objective To explore how to improve the diagnosis and surgical treatment of primary gallbladder carcinoma. Methods The clinical data of 59 patients with primary gallbladder carcinoma underwent operative treatment in our hospital from January 1985 to December 1999 were retrospectively analyzed. Results There were 22 males and 37 females in this series (M:F=1:1.68). 59%(35/59) patients were complicated by gallstone. Right upper quadrant abdominal pain, jaundice and abdominal mass were the major clinical manifestations. 34 cases underwent cholecystectomy with 57%(34/59) resection rate. Pathological examination showed that gallbladder carcinomas accounted for 94%. Postoperative 1-,3-,5-year survival rates were 36%(21/59), 20%(12/59)and 10%(6/59), respectively. Tumor chemoprevention and treatment are two approaches aimed at improving the survival of patients with cancers. An ideal anti-tumor drug is that which not only kills tumor cells but also alleviates tumor-causing risk factors, such as precancerous lesions, and prevents tumor recurrence. Chinese herbal monomers are considered to be ideal treatment agents due to their multi-target effects. Astragaloside has been shown to possess tumor chemoprevention, direct anti-tumor, and chemotherapeutic drug sensitization effects. In this paper, we review the effects of astragaloside on tumor prevention and treatment and provide directions for further research.

5.Effects of bisphenol-A on blastocyst development and implantation.

OBJECTIVETo determine the effects of bisphenol-A (BPA) on blastocyst development and implantation.
METHODSAccording to completely randomized grouping method, 90 pregnant mice were divided into 100, 300, and 600 mg/(kg·d)BPA groups and control group. BPA-treated pregnant mice were orally administered with BPA at concentrations of 100, 300 and 600 mg/(kg·d) from day 0.5 to day 3.5 of their pregnancy. Blastocyst implantation and development were studied.
RESULTSIn the 300 mg/(kg·d) BPA group, the number of implantation sites and implantation rate were significantly decreased. In the 600 mg/(kg·d) group, no implantation sites were observed among pregnant mice and BPA inhibited embryo implantation. Blastocyst development on day 4 was examined, and findings showed that the development rate and total numbers of blastocysts in BPA treatment groups had no significant difference from the control group. However, BPA at 300 and 600 mg/(kg·d) significantly reduced blastocyst hatching rate and dramatically increased the number of blastocyst apoptotic cells when compared with those in the control group.
CONCLUSIONBPA at a high concentration damages the blastocyst development before implantation and inhibits embryo implantation.

6.Precision diagnosis of hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the most common type of primary hepatocellular carcinoma (PHC). Early diagnosis of HCC remains the key to improve the prognosis. In recent years, with the promotion of the concept of precision medicine and more in-depth analysis of the biological mechanism underlying HCC, new diagnostic methods, including emerging serum markers, liquid biopsies, molecular diagnosis, and advances in imaging (novel contrast agents and radiomics), have emerged one after another. Herein, we reviewed and analyzed scientific advances in the early diagnosis of HCC and discussed their application and shortcomings. This review aimed to provide a reference for scientific research and clinical practice of HCC.

7.Effects of Tongxinluo on MMP-3, 9 and PPARgamma expressions in atherosclerotic rabbits.

OBJECTIVETo study the effects of Tongxinluo on the expressions of matrix metalloproteinase-3 (MMP-3), MMP-9 and peroxisome proliferators-activated receptor gamma (PPARgamma) in atherosclerotic rabbits and explore the mechanism of its anti-atherosclerotic effect.
METHODSTwenty-four rabbits were randomized equally into control group, atherosclerotic model group (fed with high-fat diet for 14 weeks) and Tongxinluo group. The expressions of MMP-3, 9 and PPARgamma in the 3 groups were observed by means of immunohistochemistry.
RESULTSThe expressions of MMP-3, 9 and PPARgamma in the model group and Tongxinluo group were significantly higher than those in the control group (P<0.01). After high-fat diet feeding for 14 weeks, Tongxinluo group showed significantly lower expressions of MMP-3 and 9 but higher expression of PPARgamma than the model group (P<0.01).
CONCLUSIONTongxinluo can inhibit the expression of MMP-3 and 9 and increase the expression of PPARgamma, which might be the mechanism of its anti-atherosclerotic effect.

8.Determination of cholesteryl palmitate in Oviductus Ranae by HPLC.

OBJECTIVETo develop a new method for the determination of cholesteryl palmitate in Oviductus Ranae.
METHODA HPLC method was set up, using Zorbax Silica column and cyclohexane-diethyl ether (40:1) as mobile phase with a flow rate of 1.0 mL x min(-1), and the UV detection wavelength was 203 nm.
RESULTThe calibration curve was linear over the range of 0.60-8.92 microg (r = 0.9997), the average recovery of the method was 98.4%. RSD 1.8% (n = 6).
CONCLUSIONThe results showed that method was reliable and accurate.

9.Value of serum GP73, AFP, and AFP-L3 in diagnosis of liver cancer and recurrence monitoring after radiofrequency ablation

ObjectiveTo explore the clinical value of three serum tumor markers, Golgi protein 73 (GP73), alpha-fetoprotein (AFP), and AFP-L3, in the diagnosis of liver cancer and recurrence monitoring after radio frequency ablation. MethodsA total of 174 patients who visited our hospital from July 2012 to October 2013 were included in the study, consisting of 86 patients with newly diagnosed liver cancer, 39 with liver cirrhosis, 29 with hepatitis, and 20 healthy controls. Among the patients with newly diagnosed liver cancer, 37 were followed up for three months after the radiofrequency ablation. Serum levels of GP73, AFP, and AFP-L3 were measured by ELISA, electrochemiluminescence, and affinity adsorption chromatography, respectively. Nonparametric tests were performed on the results of serum samples from the four groups which showed skewed distribution and were represented by median (quartile interval) [M(P25-P75)]. Overall comparison was made by Kruskal-Wallis H test, and comparison between groups was made by Mann-Whitney U test. Pair-matching rank-sum test was performed using Wilcoxon Signed Ranks, and categorical data were analyzed by χ2 test. ResultsThe levels of GP73, AFP, and AFP-L3 in the liver cancer group were significantly higher than those in other groups (all P<0.05). The positive rates of GP73 and AFP-L3 in the liver cancer group were significantly higher than those in other groups (all P<0.05), and the positive rates of the two markers were significantly higher than that of AFP among patients with liver cancer (P<0.05). Thirty-seven patients with newly diagnosed liver cancer were reexamined three months after radiofrequency ablation, and the preoperational AFP-L3 level in the patients who had recurrence was significantly higher than that in the patients without recurrence (P<0.05). ConclusionSerum GP73, AFP, and AFP-L3 show great values in the diagnosis of liver cancer. AFP-L3 can be used as an indicator for the identification of benign or malignant liver disease and risk of recurrence after radiofrequency ablation.
10.A COMPREHENSIVE STUDY ABOUT THE INFLUENCE OF SOYBEAN ON BIOAVAILABILITY OF NON-HEME IRON IN DIET

A series of research results on the influence of soybean to the bioavai?lability of non-heme iron in diet are reported. It is proved that Soybean and some of its products decrease the bioavailability of non-heme iron in diet by human observation and animal experiments. This result is due to insufficient control over the inhibitors and utilization of the enhancers of the bioavailability of iron in soybean itself.Experiments show that through some special treatment processes such as sprouting, fermenting and making bean curd, the above disadvantages may be effectively reduced and remarkably increase the bioavailability of the non-heme iron. These conclusions possess both theoretical and practical meaning for full utilization of soybean protein and effective prevention of iron deficiency anemia.
