1.Pentamidine sensitizes leukemia K562 cells to TRAIL-induced apoptosis

Background and purpose:Tumor necrosis factor-related apoptosis inducing ligand(TRAIL)is a successful clinical anti-tumor agent.However,not all tumor cells are sensitive to TRAIL-mediated apoptosis.In this study,we examined the TRAIL-induced apoptosis after pretreatment with pentamidine in K562 cells.Methods:When K562 cells were pretreated with pentamidine followed by TRAIL,apoptosis of cells was analyzed by cellular morphology via viewing live cells under a light microscope and annexin V-FITC/propidium iodine FACS.Western blot was performed to examine the cleavage of caspase-3,-8 and poly(ADP-ribose)polymerase(PARR)and protein levels of X-linked inhibitor of apoptosis protein(XIAP).Results:Apoptotic cell death occurred and cleavage of caspase-3,-8 and PARR appeared when K562 cells were pretreated with 10?g/ml of pentamidine followed by 200 ng/ml of TRAIL.In addition,pentamidine decreased protein levels of XIAP.Conclusions:Combined treatment with pentamidine and TRAIL may become a new strategy for cancer therapy.
4.Chemical constituents of basidiomycete Hydnum repandum

Objective To study the chemical constituents of the fruiting bodies of Hydnum repandum.Methods Separation and purification were performed on silica gel, Sephadex LH-20 and ODS CC. Their sturctures were established by 2D-NMR (1H-1HCOSY, HMQC, HMBC, and NOESY), MS, HR-MS spectra, and ORD data. Results Eleven compounds were obtained and identified as sarcodonin A ( Ⅰ ),scabronine B (Ⅱ), 3β-hydroxy-5α, 8α-epidioxyergosta-6, 22-dien (Ⅲ), (22E, 24R)-ergosta-7, 22-diene3β, 5α, 6β- triol (Ⅳ), (22E, 24R)-ergosta-7, 22-diene-3β-ol (Ⅴ), benzoic acid (Ⅵ), 4-hydroxylbenzaldehyde (Ⅶ), 4-monopropanoylbenzenediol (Ⅷ), ethyl-β-D-glucopyranoside (Ⅸ), thioacetic anhydride ( Ⅹ ), (2S, 2'R, 3S, 4R)-2-(2-hydroxyoctadecanoylamino) docosane-1, 3, 4-triol (Ⅺ). Conclusion All of the compounds are isolated from this fungus for the first time.
5.Comparative analysis of the effect of simple high cut of fibula and arthroscopic lavage debridement combined with fibular osteotomy in the treatment of knee osteoarthritis

Objective To compare the short and long term effect of knee osteoarthritis treatment with two therapeutic methods which are simple high cut of fibula and arthroscopic lavage debridement combined with fibular osteotomy.Methods Data of 45 consecutive patients with knee osteoarthritis were retrospectively observed and selected,and then they were divided into two groups according to the operation method.23 patients with simple high cut of fibula were selected as the control group,and 22 patients with arthroscopic lavage debridement combined with fibular osteotomy were selected as the observation group.Results The VAS and AKS scores of the control group at the time of 3m,6m,1 2m were (4.1 3 ±0.76)points,(1 07.04 ±21 .53)points;(2.70 ±0.64)points,(1 41 .1 7 ±1 2.57)points;(2.43 ±0.79)points,(1 48.26 ±5.81 )points;and the scores of the observation group were (3.45 ± 0.60)points,(1 26.64 ±1 4.24)points,(2.70 ±0.73)points,(1 46.45 ±7.26)points,(2.41 ±0.85)points and (1 48.26 ±5.81 )points.The differences between the two groups were statistically significant at the time of 3 months after surgery(t =3.32,-3.62,all P <0.05),the effect of observation group was better than the control group;but the other indices had no statistically significant differences (t =0.51 ,-1 .72,0.1 1 ,-0.20,all P >0.05 ). Conclusion The simple high cut of fibula with small trauma,less cost,less surgical difficulty,and long -term clini-cal curative effect is not less than the arthroscopic lavage and debridement combined with fibular osteotomy.There-fore,it should be carried out in the primary hospitals widely.
6.Two new sesquiterpenoids from basidiomycete Tyromyces chioneus.

Two new sesquiterpenoids, named as tyromols A and B (1 and 2), were isolated from cultures of basidiomycete Tyromyces chioneus, along with two previously reported 15-hydroxy-6 α, 12-epoxy-7β, 10αH, 11βH-spiroax-4-ene (3) and agripilol C (4). Compounds 1-4 were separated and purified by silica gel, RP-18, Sephadex LH-20 column chromatography. Their structures were elucidated on the basis of extensive spectroscopic analysis including IR, MS, 1D and 2D NMR experiments.
7.Effects of matrine on structure and function of K562 cells

Aim To investigate the effect of matrine on cytoskeletonof K562 cells. Methods Micropipette aspiration technique was adoptedto investigate the viscoelasticity of K562 cells, while the different ex-pression of cytoskeletal protein gene was analyzed by DNA microar-ray. Results In matrine-treated K562 cells, the viscoelastic propertiesKI, K2 and were decreased significantly from 726 ± 215 to 432 ±67,433 ±119 to 242±31, 72±38 to 50±15 respectively, and the geneexpression of prefoldin and ezrin was much stronger than that of controlcells. Conclusion The strueture and function can be changed in ma-trine-treated K562 cells.
8.Effects of fraction from Phyllanthus urinaria on thrombosis and coagulation system in animals

OBJECTIVE: To investigate the effects of PUW (a fraction containing 60% corilagin from a Chinese herbal plant Phyllanthus urinaria) on thrombosis and coagulation system. METHODS: Myers's method was used to evaluate the protection of intravenously administered PUW against the male mouse sudden death caused by injection of 75 mg/kg arachidonic acid in the tail vein; Charlton's method was modified to observe antithrombotic effect of PUW in rat electrically stimulated carotid artery thrombosis model; and the model of venous thrombosis was produced to investigate the antithrombotic effect of PUW. Rosette assay was used to observe the effect of PUW on platelet-neutrophil adhesion. The effects of PUW were monitored on euglobulin lysis time (ELT), prothrombin time (PT), kaolin partial thromboplastin time (KPTT), and bleeding time (BT) in rats, according to the methods of Kowalski, HUANG Zheng-Liang, and GU Yue-Fang, et al, respectively. RESULTS: The results showed that PUW administered intravenously significantly decreased the mouse mortality, prolonged the occlusion time of rat carotid arteries, and reduced the wet and dry thrombus weight of the inferior vena cava, respectively. PUW markedly inhibited the binding of activated platelets to neutrophils, obtaining 39.7 mg/L of the medium inhibitory concentration. Intravenously administered PUW significantly shortened ELT, prolonged KPTT while had no influence on PT; PUW increased BT in rat tail tips but the BT caused by PUW was much shorter than that by aspirin or urokinase. CONCLUSION: It is suggested that PUW has the potential of antithrombosis due to its inhibition of platelet-neutrophil adhesion. PUW shows the tendency to bleeding, however, it could not cause serious bleeding side effect as compared with aspirin or urokinase.
10. Clinical analysis of 125 cases with Mycoplasma pneumoniae pneumonia in children

Objective:
To investigate and analyze clinical symptoms, auxiliary examination results, therapeutic methods and the prognosis of the pneumonia caused by Mycoplasma pneumoniae in children.
Methods:
The clinical data including symptoms, auxiliary examination results, therapeutic methods of 125 children with Mycoplasma pneumoniae pneumonia in our hospital from September 2016 to February 2017 were retrospectively analyzed.
Results:
Among the 125 children with Mycoplasma pneumoniae pneumonia, the main symptoms were cough (100%) and fever (68%) and some cases accompanied by different extra-pulmonary complications. Varying degrees of changes were found from all the children’s abnormal chest X-ray films.
Conclusions
The symptoms of Mycoplasma pneumoniae pneumonia in children were serious, with slight clinical signs and varying degrees of change of the X-ray films. Early standardized application of macrolide antibiotics was beneficial for treatment.
