1.Effect of dysadherin gene silencing mediated by RNA interference on metastasis and invasion of pancreatic cancer cells

Objective To investigate the effect of dysadherin gene silencing on metastasis and invasion in pancreatic cancer cell line PANC1,BxPC3 in vitro.Methods PANC1 and BxPC3 cells were divided into dysa group,negative siRNA control group(HK),liposomes control group(control),dysa group and HK group were tranfected by dysadherin siRNA and Negative siRNA,respectively.The expression of dysadherin mRNA and protein were detected by RT-PCR and immunohistochemical method.Transwell test was used to evaluate the invasion ability of pancreatic cancer cells.Results After transfected by dysadherin siRNA,the dysadherin mRNA levels in PANC1 and Bxpc3 cells were decreased by 95.4% and 52.1%.The expression of dysadherin protein was also down-regulated by 91.2% and 83.6%,respectively,when compared with HK groups (P<0.05 ).The numbers of invasive cells migrated in Transwell in PANC1 cells control group,HK group and dysa group were 163.2±15.5,154.4±17.3 and 53.6±7.9;the numbers of invasive cells in BxPC cells control group,HK group and dysa group were 30.7±3.2,27.5±2.8 and 4.7±2.4,respectively.The numbers in dysa group were significantly lower than those of HK group and control group (P<0.01 ).Conclusions Silencing the dysadherin gene of PANC1,BxPC3 by RNA interference could significantly inhibit the invasive and migratory ability of canceroas cells.
2.Effect of cold storage on nutrients and immune substances in human milk

Objective To explore the optimal storage standard of fresh human milk, and to observe the influence of different cold storage condition (time-temperature) on macronutrients (fat, protein, carbohydrates, TS and energy), immune sub-stances (sIgA, lactoferrin, IL-6, 8, 10 and TNF-α) and bacteria indicators of fresh human milk.Methods Fresh milk samples (n=30) were divided and stored at three temperature and nine time points, which are 4℃ (24 h, 48 h, 72 h), -18℃(72 h, 7 d, 14 d, 4 w, 8 w, 12 w), and -80℃ (12 w, 24 w). At each time point, the macronutrients , immune substance, and bacteria colony counts of each milk sample were measured and compared with fresh milk. Results Compared with fresh milk, all indicators with the exception of lactoferrin in stored human milk showed signiifcant difference (P<0.05). Under 4℃ refrigeration condition, fat, IL-6, and TNF-α decreased, bacteria colony counts and Gram-positive colony counts increased over 72 h storage (P<0.05). Under-18℃ freezing condition, fat, protein, TS, energy and IL-6 decreased from 72 h to 12 w storage (P<0.05); carbohydrates and sIgA also decreased from 4 w and 8 w storage, respectively (P<0.05). Under -80℃ freezing condition, fat, protein, TS, energy and IL-6 decreased over 24 W storage (P<0.05).Conclusions The macronutrients, immune substance, and bacteria indicators of human milk were affected obviously by cold storage. Refrigerated at 4℃ should not be longer than 72 h, -80℃ freezing condition should be chosen for more than two months storage.

4.Study on microscopic marginal extensions of malignant liver tumors based on tumor types and ultrasonic characteristics

Objective To study the relationships between microscopic marginal extensions and tumor types ,and ultrasonic characteristics of malignant liver tumors . Methods Two‐dimensional ultrasonography , contrast‐enhanced ultrasonography and ultrasound‐guided biopsy of hepatic tumors and surrounding hepatic tissues were performed in 78 patients with malignant liver tumor . Pathological microscopic extensions were observed after hematoxylin‐eosin staining of biopsy specimens . Results T he microscopic marginal extension rates of hepatocellular carcinoma ( HCC) ,intrahepatic cholangiocarcinoma ( ICC) and metastatic liver cancer ( M LC) were 62 .5% ( 30/48 ) ,87 .5% ( 7/8 ) ,and 91 .0% ( 20/22 ) ,respectively . For tumors with well‐defined and poorly‐defined two‐dimensional ultrasound boundary ,microscopic marginal extension rates were 50% ( 14/28) ,and 86% ( 43/50) . T umor type and two‐dimensional ultrasound boundary were independent predictors for microscopic extension rate ( P < 0 .05 ) . T he median microscopic extension distances of HCC ,ICC and M LC were 1 .0 ( 0 ,3 .0 ) mm ,4 .0 ( 2 .3 ,4 .0 ) mm ,and 2 .0 ( 1 .8 ,4 .0 ) mm ,respectively . T he distance of microscopic extension increased with tumor size . T umor type and tumor size were independent predictors for the distance of microscopic extension ( P <0 .05) . Conclusions ICC ,MLC ,and tumors with larger diameter or poorly‐defined two‐dimensional ultrasound boundary have a larger distance of microscopic extension .

The characteristic genetic abnormality of neuroendocrine neoplasms (NENs), a heterogeneous group of tumors found in various organs, remains to be identified. Here, based on the analysis of the splicing variants of an oncogene Focal Adhesion Kinase (FAK) in The Cancer Genome Atlas datasets that contain 9193 patients of 33 cancer subtypes, we found that Box 6/Box 7-containing FAK variants (FAK6/7) were observed in 7 (87.5%) of 8 pancreatic neuroendocrine carcinomas and 20 (11.76%) of 170 pancreatic ductal adenocarcinomas (PDACs). We tested FAK variants in 157 tumor samples collected from Chinese patients with pancreatic tumors, and found that FAK6/7 was positive in 34 (75.6%) of 45 pancreatic NENs, 19 (47.5%) of 40 pancreatic solid pseudopapillary neoplasms, and 2 (2.9%) of 69 PDACs. We further tested FAK splicing variants in breast neuroendocrine carcinoma (BrNECs), and found that FAK6/7 was positive in 14 (93.3%) of 15 BrNECs but 0 in 23 non-NEC breast cancers. We explored the underlying mechanisms and found that a splicing factor serine/arginine repetitive matrix protein 4 (SRRM4) was overexpressed in FAK6/7-positive pancreatic tumors and breast tumors, which promoted the formation of FAK6/7 in cells. These results suggested that FAK6/7 could be a biomarker of NENs and represent a potential therapeutic target for these orphan diseases.
