1.Genealogical Investigation on Patients with Alopecia Areata

In the genealogical investigation into 1210 patients with alopecia areata(AA), we found 75 cases with a positive family history of AA, the familial incidence was 6.2%. The incidence of AA in the 75 families (1093 cases) was 14.91% (163 cases). In these families, there were 142 patients in first-grade relatives of the family members, 17 in second-grade, and 4 in third-grade. We observed 70 cases in 33 families within one generation, 87 cases in 40 families within two succesive generations, and 6 cases in 2 families within three successive generations, four pairs of monozygotic twins appeared in this investigation, but just one of each pair was affected. The relationship between pathogenesis of AA and genetic factor has been discussed.
2.Clinical observation on levofloxacin for the treatment of community acquired pneumonia in elderly patients

Objective To observe the efficacy and safety of levofloxacin for the theatment of com-munity acquired pneumonia(CAP) in the elderly patients. Methods Thirty-six elderly inpatients with CAP between May 2005 and May 2007 were treated with levofloxacin at a dosage intravenously of 500 mg once a day for 5 to 14 days treatment. Results Streptococcus pneumoniae of multiple drugs-resistant were found in sputum of 22 patients,pseudomonas aeruginosa in 8 patients and haemophilus influenzae in 6 patients de-pending on the results of the sputum culture. The total clinical efficacy rate of levofloxacin was 75.0% and bacteria elimination rate was 82.1%,and 16.7% patients showed related side effect of diarrhea,skin-rash and kidney injury. Conclusions Levofloxacin is an effective with little side effect in treatment of CAP in the elderly.
3.Doctor-patient relationship management information system:building and application

This paper introduced the construction and application of the doctor-patient relationship management information system at a tertiary general hospital in Zigong,and presented a preliminary evaluation of its application effect.On such basis,the authors proposed to enhance medical management capacity with informationization,and motivate medical workers with IT tools.At the national level,a standardized management for medical behaviors should be established,to encourage elevation of medical quality and safety,thus effectively improving the doctor-patient relationship.
4.Bronchoscopic high frequency electrocoagulation combined with balloon dilatation in treating tuberculosis inflammatory airway constriction

Objective To explore the efficacy and safety of the bronchoscopic high frequency electrocoagulation combined with balloon dilatation in treating tuberculosis inflammatory airway constriction. Methods According to the different methods of treatment, 55 patients with tuberculosis airway constriction were randomly divided into two groups, the balloon dilatation group (26 cases) and combination group (29 cases). The patients in balloon dilatation group underwent bronchoscopic balloon dilatation and the patients in combination group underwent bronchoscopic balloon dilatation combined with high frequency electrocoagulation. The patients of the two groups accepted endoscopic therapy once a week. Effective rate of recanalization for the narrow airway, frequency of effective treatment and the time of tuberculosis bacterium vanishing was recorded. Intraoperative and postoperative complications were also observed. Three months after the treatment, all patients accepted bronchoscopic to observe and assess the airway restenosis rate. Results After treatment, the effective rate in balloon dilatation group and combination group had no significant difference[69.2%(18/26) vs. 89.7% (26/29 )](P> 0.05 ),but frequency of effective treatment and time of tuberculosis bacterium vanishing had significant difference[(3.5 ±1.3) times vs. (1.5 ± 1.1) times, (23.3 ±3.6) d vs.(13.2 ±2.3) d](P<0.01). There was no significant difference on the intraoperative and postoperative complications between two groups (P>0.05). The airway restenosis rate was 33.3%(6/18) in balloon dilatation group and 7.7%(2/26) in combination group after treatment for 3 months (P <0.05). Conclusions Combination of bronchoscopic balloon dilatation and high frequency electrocoagulation is an efficacy and safety way for the tuberculosis inflammatory airway stenosis. It can reduce the frequency of interventional therapy, shorten the time of tuberculosis bacterium vanishing, and may also decrease the airway restenosis rate.
5.Factors related to heterotopic ossifications after treatment of bi-columnar acetabular fractures

Objective To discuss factors influencing the heterotopic ossifications (HO) after treatment of bi-column acetabular fractures. Methods One hundred and ninety-three cases of bi-column acetabular fractures were followed up, including 147 males and 46 females with an average age of 34. 4 years. Thirty-one cases were associated with craniocerebral trauma. The interval between injury and surgery was less than 1 week in 15 cases, 1 to 2 weeks in 121 cases, 2 to 3 weeks in 34 cases, and over 3 weeks in 23 cases. Factors that might have influenced HO, including surgical method, associated injury, and interval between injury and surgery, were analyzed retrospectively. Results All the cases were followed up for an average of 44. 2 (14 to 84) months. The mean operation time was 238 (150 to 330) minutes, and the average blood loss was 1453 (450 to 4400) mL. The incidence rate of HO was 39. 9% (77 in 193 cases), including 39 cases of degree one, 23 cases of degree two, and 15 cases of degree three. Of the 77 cases associated with HO, 14 had craniocerebral trauma and 63 did not (χ~2 = 0. 019, P = 0. 891) . HO was found in 2 cases that had been operated on in less than 1 week, 38 cases in 1 to 2 weeks, 21 cases in 2 to 3 weeks, and 16 cases in over 3 weeks. The Spearman correlation analysis between HO degree and clinical result showed no correlation ( R = 0. 041, P =0. 722). Only 15 cases (7. 8% ) were associated with HO after the debridement of the necrotic gluteus minimus. Conclusions HO after surgery for bi-column acetabular fractures may not be correlated with craniocerebral trauma, but highly correlated with the interval between injury and surgery. Debridement of necrotic gluteus minimus and other muscles can reduce the incidence of HO.

7.Effect of total flavonoids of ampelopsis grossedentata on blood lipid and hemorrheologyin in atherosclerosis rats

Objective To observe the effect of total flavonoids of ampelopsis grossedentata on blood lipid and hemorheology in atherosclerosis rats .Methods The atherosclerosis model in rats was made by freeing high grease food and intraperitoneal injection of vitamin D3 ,total flavonoids of ampelopsis grossedentata was gave by gavage ,and the level of serum lipid and blood rheology were tested 24 weeks later .Results Total flavonoids of ampelopsis grossedentata could obviously decreased the level of TG ,TC ,LDL , and increased HDL(P<0 .05) .It could significantly decrease whole blood viscosity ,plasma viscosity and hematocrit (P<0 .05) . Conclusion Total flavonoids of ampelopsis grossedentata can adjust fatty substance metabolism ,improve hemorheology of athero-sclerosis rats ,and has therapeutic effect for atherosclerosis .
8.Effects of Dihydromyricetin on Blood Lipid Metabolism and Antioxidation in Atherosclerosis Rats

Objective To observe the effect of dihydromyricetin of ampelopsis grossedentata on lipid metabolism and antioxidation in atherosclerosis (AS) rats. Methods Sixty Wistar rats were randomly divided into five groups (n = 12 each):normal control group, model control group, positive control group, high dose dihydromyricetin group (40 mg·kg-1 ) and low dose dihydromyricetin group (10 mg·kg-1 ). Except normal control group, rats in the other groups were injected with a single dose of vitamin D3(600 000 U·kg-1 ) and loaded with high fat diet to establish AS model. Simvastatin (5 mg·kg-1 ) was intragastrically administered to positive control group. High and low dose dihydromyricetin groups received intragastric administration of 40 and 10 mg·kg-1 dihydromyricetin, respectively. Equal volume of purified water was given to normal and model group. After 24 weeks of administration, serum levels of lipids, activities of superoxide-dismutase (SOD) and malonicdialdehyde (MAD) were determined. Results As compared with model control group, triglycerides (TG) of high and low dose dihydromyricetin groups was decreased [(191. 65±101. 10) vs. (111. 10±29. 29) and (120. 55±38. 12) mg·L-1 , respectively], total cholesterol (TC) was decreased [(151. 64±33. 62) vs. (148. 49±30. 14) and (118. 90±27. 38) mg·L-1 ], and high density lipoprotein cholesterol (HDL-C) was increased [(1. 29±0. 68) vs. (2. 10±0. 70) and (1. 62±0. 61) mmol·L-1 ], low density lipoprotein cholesterol (LDL-C) was decreased [(5. 01±1. 33) vs. (3. 97±0. 78) and (4. 28±0. 79) mmol·L-1 ], activity of SOD was increased [(141. 03±42. 52) vs. (187. 97±42. 08) and (150. 99±46. 17) U·mL-1 ], and MDA was decreased [(20. 51±3. 81) vs. (17. 64±1. 54) and (18. 52± 3. 42) nmol·mL-1 ], with significant differences (P<0. 05, P<0. 01). Conclusion Dihydromyricetin can reduce the level of serum lipid, improve antioxidation activity, and has therapeutic effect for atherosclerosis.
9.Effect of silencing gene DNA methyltransferase 3b expression by shRNA on proliferation of bladder cancer T24 cells

Objective To investigate the effect of recombinant plasmid pshRNA-DNMT3b on expression of DNMT3b mRNA and protein and on the proliferation of bladder cancer T24 cells,and research the function of DNMT3b in the process of bladder tumor formation.Methods There were three groups in this study,which are blank controller,HK and pshRNA-DNMT3b(24h,48h,72h),respectively.T24 cells were cultured routinely and transfected by the recombinant plasmids with lipfectamine 2000.The cells were detected by methods of RT-PCR,western blot and MTT.The varying level of DNMT3b mRNA and expression protein,and the conditions of cellular survival rate were observed.Results The recombinant plasmids were successfully transfected into T24 cell lines.The grey valHe of RT-PCR elctrophoretogram was analyzed by the software of Gel-pro analyzer,the rate of blank controller,HK and pshRNA-DNMT3b(24h,48h,72h),was (99.56±1.24)%,(99.12±1.35)%,(75.77±1.42)%,(44.69±1.05)%and(20.52±0.89)%,respectively.The analytical resuit of western blot image was(99.43±1.28)%,(98.90±1.31)%,(67.83±1.02)%,(43.43±1.05)%and(21.92±0.89)%.There was no statistically difference in survival between blank control and HK(P>0.05).The group of pshRNA-DNMT3b and other two groups had statistical difference only at the 72th hour and the cell inhibitory growth rate only increase 0.45%.Conclusions The recombinant ptasmid pshRNA-DNMT3b can inhibit the expression of mRNA and protein of DNMT3b effectively.However,it has slight function on inhibiting cell proliferation.
10.Preparation of curcumin prodrugs and their in vitro anti-tumor activities.

The curcumin prodrugs, which could be selectively activated in tumor cells, were prepared to establish a basis for the targeted chemotherapy for cancer. On the basis of the molecular structure of curcumin, the N-maleoyl-L-valine-curcumin (NVC), N-maleoyl- glycine-curcumin (NGC) were chemically synthesized and identified by IR and NMR spectroscopy. After treatment with these two prodrugs for 6 - 24 h, the rates of growth inhibition on human bladder cancer EJ cells and renal tubular epithelial (HKC) cells were detected by MTT colorimetry. Our results showed that after the treatment with 20 micromol/L - 40 micromol/L NVC and NGC for 6 - 24 h, the growth inhibitory effects on EJ cells were 6.71% - 65.13% (P < 0.05), 10.96% - 73.01% (P < 0.05), respectively, in both dose- and time-dependent manners. When compared with the curcumin of same concentrations, the growth inhibitory effects of these two prodrugs on HKC cells were significantly decreased (P < 0.01). It is concluded that activation of curcumin prodrugs via hydrolysis functions of cellular esterase could inhibit the growth activities of tumor cells, and reduce the side effects on normal diploid cells. This provided a novel strategy for further exploration of tumor-targeted chemotherapeutic drugs.
