1.Tumor necrosis factor-? regulates the effect of glutamine on protein synthesis in rats' skeletal muscle

Objective:To find the regulations of glutamine(Gln) metabolism by tumor necrosis factor-?(TNF-?).Methods:Thirty male SD rats were assigned into three groups: A(total parenteral nutrition,TPN),B(TPN+Gln),C(TPN+ TNF-?+Gln).Besides all rats were supported by isonitrogenous and isocaloric TPN solutions for 3 days,rats in group B were supplied with Gln(in the form of alanylglutamine) for 3 days at the rate of 0.3 g/(kg?d),rats in group C were infused intravenously with TNF-? at the rate of 5 ?g/(kg?h) within the last 24 hours and also supplemented with Gln for three days at the same rate as in group B.At the last 0.5 hour,all animals were mainlined with \leucine at the dosage of 1.0 mmol/kg.TNF-? levels in plasma,concentrations of Gln in plasma and skeletal muscle were measured as the experiment ended after 72 hours,and the fractional synthesis rate(FSR) were also assessed.Results:The Gln concentrations in plasma and skeletal muscle increased markedly from group A,B to C.The levels of FSR decreased remarkably along with the sequence from group B,C to A.The statistical differences were significant(P
2.Mechanism of scarless healing of embryonic skin

OBJECTIVE: Post-traumatic healing style is an important link in the process of wound healing. Scarless healing of fetus skin after trauma is the ideal healing style that is hunted for by people. The results of studies on the mechanism of scarless healing were reviewed to explore the mechanism of scarless healing of fetus skin.DATA SOURCES: Computer was applied to retrieve Medline database on the articles relating to the mechanism of scarless healing of fetus skin from January 1970 to December 2003. Docuterms were "fetus, skin and scarless healing". Language in the articles was limited to English.STUDY SELECTION: The documents were examined at first to exclude the study on non-healing mechanism. Full texts were found in the surplus documents. The inclusive criterion was the foundational experiment on the mechanism of scarless healing. Clinical trial and phenomenal study on scarless healing were excluded.DATA EXTRACTION: Forty-eight articles on experimental study relating to embryonic scarless healing were collected in all. Sixteen experiments accorded with the inclusive criterion and 32 were excluded.DATA SYNTHESIS: Sixteen experiments discussed the relationship between the fetus and scarless healing on terms of growth environment, inflammation reaction to trauma and regulation of fibroblast in organism, extracellular matrix and nany cellular factors.CONCLUSION: Growth environment of fetus has not so much relationship with the mechanism of scarless healing after trauma. Low reaction of fetus to inflammation, different extracellular matrix, component of collagen fiber and cell factors may take part in scarless healing process. Regulation of gene expression may play an important role in the process. But the concrete mechanism of scarless healing is not clear yet and still needs more researches and elucidations.
3.Effects of gunshot wound on lipid peroxidation in plasma and pulmonary tissues and amount of superoxide anions released by PMNs in pigs

The amount of superoxide anions released by polymorphonuclear leukocytes (PMNs)and malonyldialdehyde (MDA) level in plasma,lung homoge-nate and bronchoalveolar lavage fluid (BALF) were determined in pigs after they were inflicted with gunshot wounds of steel-ball pellets.It was found that the amount of superoxide anions released spontaneously and after the stimulation of zymosa was increased and reached the peak in the 4th and 8th hour after injury respectively,and then returned to the preinjury level gradually.The changes of plasma MDA level was similar to those of superoxide anions released by PMNs.MDA level of lung homogenate and BALF was also significantly increased in the 24th hour postinjury.In addition,the relationship between PMNs function and lipid peroxidation in the plasma and the lungs was discussed.
4.The effects of naloxone and positive end expiratory pressure ventilation on respiratory function in dogs with experimental RDS

Respiratory distress syndrome was produced in dogs with the combination of oleic acid injection and gunshot wounding,and the therapeutic effects of naloxone(NAD and positive end expiratory pressure ventilation(PEEP)were assessed with the examinations of macropatholo-gy of the lungs,lung water ratio.respiratory rate(RR)and blood gas analysis.It was found that RR was rapidly and significantly increased tPaO2 decreased and Qs/Qt increased and there was severe pulmonary hemorrhage,edema and atelectasis in the controls after injury.NAL treatment could slightly alleviate the increase of RR and prevent the early of PaCO2,but it had no effects on the changes of PaO2,Qs/Qt and increase pulmonary hemorrhage,edema and atelectasis.PEEP could improve the decrease of PaO2 and the increase of Qs/Qt and prevent pulmonary atelectasis and alveolar edema from occurring,but it could not significantly stop the increase of lung water ratio.
5.Double-antigen sandwich ELISA for detection of total antibodies against hepatitis C virus

Objective To establish a double-antigen sandwich ELISA(S-ELISA) for detection of total antibodies against hepatitis C virus(HCV).Methods Recombinant HCV proteins fusion-expressed with His-tag and GST-tag were separately used as coating and HRP-labeling antigen of the S-ELISA.Serum samples were tested with both the S-ELISA and another commercial indirect ELISA(I-ELISA) kit(Beijing Wantai Biological Pharmacy Enterprise Co.Ltd.).HCV RNA in some of the samples were tested by RT-nested PCR.Results Among 1 968 tested samples,190(9.7%) were total anti-HCV-positive while 1 761(89.5%) were negative by both of the S-ELISA and I-ELISA,with a resultant concordance rate of 99.1% of the two ELISA assays.However,the results of 17(0.9%) were not consistent in the two assays,in which 14 were S-ELISA negative but I-ELISA positive and 3 were S-ELISA positive but I-ELISA negative.One of the 14 samples(0.7%) with S-ELISA negative was detected as HCV RNA-positive,while 2 of the 3(66.7%) samples with S-ELISA positive were detected as HCV RNA-positive.In addition,HCV RNA was detectable in 23 of 31(74.19%) samples which were total anti-HCV positive in both S-ELISA and I-ELISA.Taking the PCR data together into account,the sensitivity of the S-ELISA and I-ELISA were 99.48% and 98.96% and specificity were 99.94% and 99.27%,respectively.Conclusions The established S-ELISA in this study may provide a novel means for detection of HCV antibody with high sensitivity and specificity.
6.The changes and effect of the lungs and blood gas index after underwater blast injuries

Objective To observe effect of the pulmonary injury and the dynamic changes of the blood gas index on the mortality rate of the dogs suffered from underwater blast injury. Methods 38 adult dogs were brought out underwater blast injury by TNT. The dogs that died on the spot were anatomized immediately, while the survivors were anatomized after 6 hours; the blood samples were taken before the explosions, 30min and 6 hours after them to analyze the blood gas index by i-STAT. Results Most of the lung injuries were pulmonary hemorrhage and edema; the positive impulse of the blast wave and the severity of the lung injury were closely related to the mortality rate; the frequency of the respiratory and the blood oxygen and the carbon dioxide pressure can roughly exhibit how severe the lungs were injured. Conclusions The severe hypoxia induced by lung injury is the main cause of the early death; correlation between the severity of the injury and the positive impulse of blast wave is very obvious.

Objective To study the protective effects of low frequency electric stimulation at bilateral mastoid processes on the focal cerebral lesion induced by ischemia-reperfusion in rats. 10.ANALYSIS OF ROAD TRAFFIC ACCIDENTS IN CHONGQING IN RECENT TEN YEARS

To study the characteristics and preventive methods of road traffic accidents (RTA) in different regions of Chongqing, 2 police teams of traffic management in downtown, 2 teams at the arban suburb junction, 1 team in the county and 1 freeway team were selected randomly, and RTA occurring in the respective listrict, above middle RTA between 1988 to 1997 were investigated and analyzed. In a total of 13121 cases of RTA, there were 6201 RTA with casualties, 14 catastrophic RTA, and 977 serious RTA. In these RTA there were 1086 deaths, 3165 severely wounded and 7327 mildly wounded. Pedestrians and passenger weres constituted 44 0% and 42 5% of total casualties respectively. 57 9% of severely wounded and deaths occurred in pedestrians. As a whole,there was a tendency of rise in RTA every year, having a significant characteristic of time distribution. The majority RTA occurring in Chongqing was related with violation of driving regulations and recklessness in crossing roads. causes of RTA, time distribution and casualties were different in different regions. In order to decreese the rate of RTA,it is deemed essential to enhance propaganda to raise the awareness of traffic accidents and to reinforce traffic management appropriate for respective district of the city.
