1.Effects of mechanical factors on proliferation and differentiation of tissue-engineered stem cells

BACKGROUND:With the development of tissue engineering technology and the deep research of tendon regeneration, many problems caused by traditional tendon transplantation wil be solved. OBJECTIVE:To evaluate and prospect the effects of corresponding mechanical systems on seeding cel s based on the characters of tendon, stem cel s as wel as mechanical systems. METHODS:CBM, CNKI, CqVip and PubMed databases were retrieved for reviews and articles related to tissue-engineered tendon published from January in 2005 to December in 2015. The keywords were“tissue engineering, tendon, tendon stem cel , mechanical stimulation and seeding cel s”in Chinese and English, respectively. Final y a total of 63 articles were selected for overview. RESULTS AND CONCLUSION:As al kinds of stem cel s and the corresponding mechanical systems exhibit advantages and disadvantages when applied for tissue engineering, it is advisable to make appropriate choices according to the research needs. Tendon stem cel s show a broad application prospect in tendon regeneration, which wil grow into required tendon tissues through appropriate, accurate and gentle mechanical stimulations, thereby providing another alternative to improve the tendon healing.
2.The effects of antiphospholipid antibodies and EGb761 on the Schwann cells in vitro

Objective To detect the effect of the antiphospholipid antibodies on the Schwann cells in vitro and to find if the Extract of Ginkgo biloba 761(EGb761)has any protective effects.Methods The immunoglobulins(Ig)were extracted from the serum of the patient with autoimmune polyneuropathies and those with controls.Ig only or Ig plus calf serum(served as complement)were added into the culture solutions of the Schwann cells.The density of the cell in one high power field,the cell perimeter and area were detected and compared to those blank and those controls.Results The shape of the Schwann cells in both the Ig and Ig plus calf serum solution were greatly changed.The densities of the cells in them were both lower than the blank ones,with more severe in the cells with calf serum.There was no significant different between the solutions with or without EGb761.Conclusion Ig from the patients with autoimmune polyneuropathies could attack the Schwann cells in vitro,with the existence of complements,the destruction was even more severe.No protection of EGb761 could be found in these situations.
3.Study on expression and significance of inflammatory factors in dog atria with atrial fibrillation

Objective To investigate the expression and significance of interleukin-1β and C-reactive pro-tein(CRP)in atrial fibrillation(AF).Methods Sixteen mongrel dogs were randomly divided into two groups:pa-cing group(n=8)and control group(n=8).High frequency pacing was performed in the pacing group with perma-nent pacernaker but not in the control group.After pacing for 24 weeks,tissue samples were obtained from the left and right atria.The levels of IL-1βand CRP in atria were detected by immunohistochemistry.Results Compared to the control group,the expression level of IL-1β and CRP were significantly enhanced(P<0.05)in atrial tissues during AF.Conclusion IL-1β and CRP may participate in atrial fibrillation formation and play a role in generation and maintaining the artrial fibrillation.
4.Contrast-enhanced ultrasonography in diagnosing experimental abdominal blunt trauma

Objective To evaluate the diagnostic advantages of contrast-enhanced ultrasonography in experimental abdominal blunt trauma in comparison of routine ultrasonogrphy. Methods Liver, spleen and kidney blunt trauma models were established in 10 canines and 12 rabbits. Both routine ultrasonography and contrast-enhanced ultrasonography were performed to identify the possible injury. Accuracy of the two ultrasonic technologies was compared in diagnosing each kind of organ and various injury degrees. Results Fifty-six trauma models including 22 livers, 14 spleens and 20 kidneys were established. There was significant difference between two ultrasonic methods in diagnosing liver, spleen and kidney injury (P<0.01);and contrast-enhanced ultrasonogarphy had particular advantage in detecting minor injuries of Ⅰ-Ⅱ scale (P<0.01). Conclusion Contrast-enhanced ultrasonography greatly enhances diagnostic accuracy of blunt abdominal solid organ trauma, especially in minor injuries.
5.Value and safety of SonoVue in diagnosis of abdominal diseases

Objective To assess the value and safety of SonoVue in ultrasonic diagnosis of abdominal diseases. Methods A total of 204 patients (149 male, 55 female) with abdominal examination of contrast-enhanced ultrasonography (CEUS) with SonoVue from August 2007 to February 2009 were reviewed. Results Definitive diagnosis were obtained 215 times in 204 patients (233 times) with abdominal CEUS. The diagnostic accuracy of CEUS was 92.27%, significant higher than that of conventional ultrasonography (62.23%). Anaphylactic shock happened 1 time (0.43%) in 1 patient. Conclusion CEUS with SonoVue can provide a high diagnostic accuracy with relatively safety of the possibility of serious adverse event.
6.Classification of mandibular condyle fractures and selection of treatment

Objective To generalize a valuable classification of mandibular condyle fractures and explore proper measures for such kind of fractures so as to provide reference for clinical treatment. Methods The data of 304 cases with 358 mandibular condyle fractures were analyzed retrospectively. After one year of the initial treatment, X-ray inspection was employed to get the information including mouth opening, pattern of mouth opening, occlusion, facial symmetry and healing condition of mandibular condyles. Results According to classification of position,displacement and age (PDA), 180 mandibular condyle fractures belonged to type Ⅰ, 96 to type Ⅱ and 82 to type Ⅲ, which coincided with the different fractures treated surgically or nonsurgically. All surgical and nonsurgical treatments got satisfactory outcome. Conclusion The classification of PDA is effective in guiding the treatment choice for mandibular condyle fractures. Both surgical and nonsurgical treatment can obtain effective results for corresponding case.
7.Effects of recombinant human growth hormone on final adult height in children with Turner′s syndrome and analysis of the influencing factors

Objective To observe the final adult height (FAH) outcome and influencing factors in Turner′s Syndrome(TS) children treated with recombinant human growth hormone ( rhGH ).Methods Thirty TS children treated with rhGH were compared with 16 TS children without rhGH treatment and were followed up to achieve their FAH.Comparisons were made regarding predicted adult height (PAH),height standard deviation score for chronological age( HtSDScA ),height SDS for BA( HtSDSRA ),and growth velocity ( GV ) between rhGH treatment and without treatment groups and between the onset and by the end of rhGH treatment group.The factors determining FAH were also evaluated.Results FAH in rhGH treatment group was obviously improved as compared with untreatment group[ ( 149.5±6.3 vs 142.4±5.2) cm,P<0.01 ].FAH in treatment group was positively correlated with height standard deviation score for chronological age ( Ht0 SDSCA ),Hto SDS for BA ( Hto SDSBA ),height age ( HA0 ) at preliminary diagnosis,and correlated with duration of rhGH therapy,duration of estrogen-free rhGH therapy,and PAH0SDS at preliminary diagnosis.Stepwise regression analysis indicated that duration of estrogen-free rhGH therapy and PAH0 SDS were the variables with the greatest identified influence on FAH (F =11.56 and F =86.91,P< 0.01 ).FAH in the 45,XO group was significantly different from the mosaicism group (45,XO/46,XX ) [ ( 147.2 ± 6.3 vs 153.3±6.4) cm,P =0.038].Conclusion rhGH treatment is efficacious in improving FAH of TS children,but a variability in the magnitude of the response to rhGH is recognized.Duration of estrogen-free rhGH therapy and PAH0SDS are the variables with the greatest identified influence on FAH,and karyotype may be one of the influence factors.rhGH treatment should be initiated as early as possible and sufficient course of estrogen-free rhGH therapy is needed to yield a satisfactory FAH.
8.Efficacy of treatment with gonadotropin-releasing hormone analogue alone or combined with recombinant human growth hormone in pubertal short children

Objective To assess the efficacy of gonadotropin-releasing hormone analogue(GnRHa)with or without recombinant human growth hormone(rhGH)treatment in Chinese short pubertal children with non-growth hormone deficiency.Methods Of 42 short pubertal children(14 males,28 females)without growth hormone deftcieney,the average age was(11.6±0.8)year.30 children were treated with slow release GnRHa with initial dose (100μg·kg-1·d-1,28d)and maintenance dose(60-80μg·kg-1·d-1,28d)labd rgGH with initial dose(0.15IU·kg-1·d-1)and maintenance dose(0.10-0.15IU·kg-1·d-1)for at least 1year.16 of them were still ongoing till the end of the second year.12 children were treated with GnRHa alone by initial dose(100μg·kg-1·d-1,28d)and maintenance dose (60-80μg·kg-1·d-1,28d),and 7 of them remained on it for 2 years.Dynamic changes including annual growth velocity(GV),bone age(BA)/chronologic age(CA)ratio,Tanner stage,height SDS for CA (HtSDSCA),height SDS for BA(HtSDSBA),and predicted adult height (PAHSDS)were observed.Results By the end of the first year tretment with combination therapy,the following parameters:GV,HtSDSCA,HtSDSBA,and PAHSDS all increased significantly(all P<0.05).Treatment with GnRHa alone did not yield significant changes in GV,HtSDSCA,HtSDSBA,and PAHSDS(all P>0.05).Changes in GV,HtSDSBA,and PAHSDS between these two groups were statistically significant(all P<0.05).By the end of the second year treatment,in the combination group,GV slowed from 6.7 to 5.5 cm/year(P<0.05).HtSDSCA,HtSDSBA,PAHSDS increased(all P<0.05).In the group with GnRHa treatment alone,GV slowed from 4.0 to 3.6 cm/year(P>0.05).HtSDSCA,HtSDSBA,PAHSDS increased(all P>0.05).Changes in GV,HtSDSCA,HtSDSBA,and PAHSDS between these 2 groups were statistically significant respectively(all P<0.05).Conclusion This combined treatment regimen significantly impreved the growth by increasing growth rate and delaying bone matumtion in pubertal chidren without growth hormone deficiency.Further study is needed to verify beneficial effects on the final height gain.
9.Establishment of enzyme-linked immunosorbent assay one-step assay based on recombinant proteins derived from different genotypes of hepatitis E virus

Objective To establish an anti-hepatitis E virus (HEV) enzyme-linked immunosorbent assay (ELISA) one-step assay based on seven glutathione S-transferase (GST)-fusion recombinant proteins derived from different HEV genotypes and subtypes. Methods Concentration of the coating antigen was optimized by block titration. The cut-off values were determined for anti-HEV IgG and IgM, respectively. Assay sensitivity, specificity and reproducibility were investigated using samples with confirmed anti-HEV positive. Results An optimal concentration of mixture of recombinant proteins (Mix166) was 1.5 mg/L for antigen coating. Coefficient of variations (CV) of anti-HEV within-run and between-run were 8.67% and 10.85%, respectively. CV of anti-HEV IgM within-run and between-run were 4.56% and 5.99%, respectively. Positive rates of anti-HEV IgG and IgM were both 94% for 50 HEV-polymerase chain reaction (PCR) positive sera tested with the one step assay. Using one-step assay to detect 674 serum samples from healthy people, 52 samples were found anti-HEV IgG positive and 3 samples were anti-HEV lgM positive. A series of serum specimens collected at different time points until 76 weeks from a chimpanzee challenged with HEV Mexican strain were anti-HEV IgM positive during week 1--6 and anti-HEV IgG positive during week 2--76 determined by the one step ELISA. However, import ELISA kits were lack of both the IgM and lgG reactivity to all of the serial chimpanzee sera. Conclusions The sensitivity and specificity of anti-HEV ELISA one-step assay based on the Mix166 antigen are high and could be used for the diagnosis of HEV infection.
10.Effects of Dahuang Zhechong Pill on expressions of tissue inhibitor of metalloproteinase-1 and plasminogen activator inhibitor-1 in rats with glomerulosclerosis

OBJECTIVE: To explore the mechanisms of the beneficial effects of Dahuang Zhechong Pill (DHZCP), a Chinese patent herbal medicine, in treatment of chronic renal disease, and to investigate the effects of DHZCP on the expressions of renal tissue inhibitor of metalloproteinase-1 (TIMP-1) and plasminogen activator inhibitor-1 (PAI-1) mRNAs in rats with adriamycin-induced glomerulosclerosis. METHODS: Focal segmental glomerulosclerosis and diffuse mesangial proliferation were induced in rats by combined procedures, including unilateral nephrectomy, intravenous injection of adriamycin and giving high-fat foods. The rats were randomly divided into untreated group, benazepril-treated group and DHZCP-treated group. Another 6 rats were sham-operated as control group. After 12-week treatment, rats were sacrificed, reverse transcription polymerase chain reaction (RT-PCR) method and computerized image analytical technique were used to determine the expressions of TIMP-1 and PAI-1 mRNAs. RESULTS: Compared with the untreated group, the ratios of TIMP-1/beta-actin and PAI-1/beta-actin of the DHZCP-treated group were decreased, suggesting that DHZCP could down-regulate the expressions of TIMP-1 and PAI-1 mRNAs (P<0.05). Benazepril could significantly inhibit the expression of TIMP-1 mRNA compared with that of the untreated group (P<0.05) CONCLUSION: DHZCP can down-regulate the expressions of TIMP-1 and PAI-1 mRNAs in renal tissues of rats with focal segmental glomerulosclerosis and diffuse mesangial proliferation, which may be its action mechanism in treating chronic renal disease.
