1.Growth hormone regulate the function of T lymphocytes

To study the effects of growth hormone on function of T lymphocytes. Methods: Tne expression plasmid PcDNA-GHcDNA was constructed,then the plasmids were transfected into Jurkat cells,on the other hand,Jurkat cells were treated with different doses ofrhGH, to investigate the effect of overexpression GH and rhGH on function of T lymphocytes. Results: Overexpression GH could increase the ex-pression of IL-2R(140%) and secretion of IL-2(160%) and IFN-γ( 198% ) in Jurkat cells after PHA activation,but these data were broughtdown by the treatment of GH antibody. rhGH could also enhancer the function of T lymphocytes after PHA activation. Conclusion: These resultssupport the idea that GH is an autocrine and paracrine factor, and can regulate the function of T lymphocytes.
2.Preliminary clinical significance of determination for serum insulin-like growth factor binding protein-3 protease activity

Objective To evaluate the clinical significance of determination for serum insulin like growth factor binding protein 3 (IGFBP 3) protease activity. Methods Serum IGFBP 3 protease activity in normal persons, parturient pregnant women, acromegalic patients and patients with growth hormone deficiency (GHD) was determined by Western blot technique. Results Serum IGFBP 3 protease activity in parturient pregnant women, acromegalic patients,normaladultsanduntreatedGHDpatientswere (75.2?2.2)%, (68.6?3.9)%, (22.7?3.1)% and (13.4?4.2)% respectively. Serum IGFBP 3 protease activity in GHD increased to (29.0?6.8)% after treatment with recombining human somatropin (rhGH) for six months. Conclusions Serum IGFBP 3 protease activity is modulated by GH and affects serum levels of members of GH IGF Ⅰ axis.

PRL receptors exist ubiquitously in all tissues and organs of immune system and PRL modulates local and systemic immune activities. PRL is involved in many physiological and pathological immune responses. The function of PRL in autoimmune diseases, organ transplantation and tumor occurrence has been explored in recent years. This article is to review the progress of prolactin in the immunology-related field.

7.Primary culture of human preadipocyte in a serum-free medium

Objective To establish a method of primary culture of human preadipocyte in a serum-free medium. Methods Collangenase digestion was used to dispart preadipocyte.The cells were identified by microscopy stained by oil red O and determination of the activity of Glycerol-3-phosphate dehydronase(G-3-PDH). Results The primary cultured human preadipocyte proliferated in the aserum-free medium successfully.In the differential serum-free medium the cells turned to be round on the 4 th day.The adipose drops began to cumulate in the cells,and to the most quantity until the 21 st day.Conclusion The human preadipocyte can be primarily cultured and induced to differentiate in serum-free medium,which is the base for researching the effects of hormones and factors to the proliferation and differentiation of preadipocyte.
8.Characterization of gene expression and adipokine secretion during differentiation of primary cultured human omental preadipocytes

Stromal preadipocytes from human omental adipose tissues were isolated with enzyme-digesting method,and cultured for inducing differentiation.Preadipocyte factor-1 was abundant during preadipoeyte stage,whereas lipoprotein lipase and PPARγ2 mRNA were rapidly up-regulated at early stage of differentiation and CCAAT/enhancer binding protein expression was increased rather late.In addition,resistin expression and secretion was primarily detectable in preadipocytes,leptin was gradually increased during adipogenesis,whereas adiponectin appeared specifically in mature adipocytes.Insulin-like growth factor-Ⅰ secretion was remarkably detected in both preadipocytes and mature adipocytes.
9.Cytokines regulate the promoter activities of human growth hormone gene in rat pituitary MtT/S cells

Objective:To study the effect of interleukin-11(IL-11),ciliary neurotropic factor (CNTF) and transforming growth factor-β (TGF-β) on the hGH gene promoter activity in rat pituitary MtT/S cells and the interaction with pituitary-specific transcription factor Pit-1.Methods:Stable transformed MtT/S cell line which contains hGH gene promoter -484-30 bp and luciferase reporter gene firstly established,then the concentration of GH in the medium and lysate of MtT/S cells and luciferase activities in MtT/S cells were measured after treatment these cells with the above cytokines,the effects of cytokines on secretion and synthesis of GH,and the promoter activity of the hGH gene were observed.Results:The results showed that IL-11(20 nmol/L),CNTF(10 nmol/L) and TGF-β(5 nmol/L) regulated secretion and synthesis of GH,and the luciferase expression in stable transformed MtT/S cells.IL-11 and CNTF had the stimulatory effect,whereas TGF-β had the inhibitory effect.Neither overexpression of Pit-1 nor inhibiting Pit-1 expression affected the regulatory role of these cytokines.Conclusion:IL-11,CNTF and TGF-β regulate the GH production in pituitary MtT/S cell line by regulating the hGH gene promoter activity.Pit-1 may not be involved in these actions.
10.Clinical value of LHRH exciting test in differential diagnosis of constitutional delayed puberty and male hypogonadotropic hypogonadism

Objective To analyse the clinical significance of LHRH exciting test in the differential diagnosis of constitutional delayed puberty (CDP) and hypogonadotropic hypogonadism (HH). Methods Eighty-one cases from 1982 to 1998 were investigated and followed up. They were all at genital stage Ⅰ. After injection of 100 ?g LHRH, the blood samples (3 ml) were taken at -15, 0, 15, 30, 45, 60, 90 and 120 min. The serum LH and FSH levels were determined by radioimmunoassay. Then they were followed up every 3-24 months. After they received LHRH exciting test, they were followed up until over 18 years old. According to their puberty development status, they were divided into 3 groups, normal group (n=34),CDP group (n=16) and HH group (n=31),andthemeanage,whenthey received LHRH exciting test, was (10.2?0.9, range 9-14) years, (16.0?1.0, range 14-18) years and (17.1?1.4, range 16-22) years respectively. Results There were no significant differences in serum LH baseline level and peak time in normal, CDP and HH groups, but the serum LH peak level, LH increment (peak LH level minus baseline LH level), LH increment ratio (peak level/baseline level of LH) and the area under LH curve (AUC LH ) of normal group were significantly higher than those of CDP group and HH group (all P
